Wavelength-dependent three-dimensional single-molecule superlocalization imaging for yoctomole detection of thyroid-stimulating hormone on a quantum dot nanobiosensor

A wavelength-dependent three-dimensional(3D)superlocalization imaging method on gold nanoislands(GNIs)chip was developed as a supersensitive single-molecule thyroid-stimulating hormone(TSH)*** and fluorescent signals from gold nanoislands on the substrate and quantum dots(QDs)nanoprobes were simultaneously isolated and acquired within an evanescent field layer generated by total internal reflection(TIR)of incident light using a dual-view *** 3D TIR fluorescence images of TSH-bound QDs on the GNIs were obtained using z-axis optical sectioning at 10nm intervals before/after immunoreaction to identify the optimal conditions for *** localized centroid position of QD nanoprobes and GNI were distinguished at a subdiffraction limit resolution using 3D Gaussian fitting to the point spread *** QD TSH nanobiosensor using wavelength-dependent 3D TIR fluorescence-based single-molecule localization microscopy(3D TIRF-SLM)imaging technique showed an excellent detection limit of 90 yoctomoles(~54 molecules)and a wide linear dynamic range of 1.14 zmol/L-100 pmol/L for *** detection sensitivity was about 4.4×10^(9)times higher than conventional enzyme-linked immunosorbent assay and could successfully quantify TSH in human *** wavelength-dependent 3D TIRF-SLM technique may emerge as a reliable platform for ultrahigh-sensitive nanobiosensors at the single-molecule level and early diagnosis with quantification of disease-related ultra-tracebiomolecules.

Base pair distance analysis in single DNA molecule by direct stochastic optical reconstruction microscopy

Precise fluorescence imaging of single l-DNA molecules for base pair distance analysis requires a superresolution technique, as these distances are on the order of diffraction limit. Individual l-DNA molecules intercalated with the fluorescent dye YOYO-1 were investigated at subdiffraction spatial resolution by direct stochastic optical reconstruction microscopy（d STORM）. Various dye-to-DNA base pair ratios were imaged by photoswitching YOYO-1 between the fluorescent state and the dark state using two laser sources. The acquired images were reconstructed into a super-resolution image by applying Gaussian fitting to the centroid of the point spread function. By measuring the distances between localized fluorophores, the base pair distances in single DNA molecules for dye-to-DNA base pair ratios of 1：50,1：100, and 1：500 were calculated to be 17.1 0.8 nm, 34.3 2.2 nm, and 170.3 8.1 nm[17_TD$IF], respectively,which were in agreement with theoretical values. These results demonstrate that intercalating dye in a single DNA molecule can be photoswitched without the use of an activator fluorophore, and that super-localization precision at a spatial resolution of 17 nm was experimentally achieved.

Super-resolution morphological dissemination of intercalating dye in single DNA molecules via binding activated localization microscopy

Fluorescent dye （YOYO-I） intercalated with single DNA molecules were investigated via bindingactivated localization microscopy （BALM） at sub-diffraction limit resolutions. Various dye-to-DNA base pair （bp） ratios were imaged using the blinking property of YOYO-1 dye under optimum BALM switching buffer conditions. Individual DNA molecules exhibited regular/irregular intercalating phenomena with respect to dye-to-DNA bp ratio. The acquired images were reconstructed into super-resolution images by applying a Gaussian fit to the centroid of the point spread function. The YOYO-1 intercalated with λ-DNA possessed a non-homogeneous region due to the different binding modes of YOYO-1 with λ-DNA. Each binding mode was imaged at the sub-diffraction limit super-resolution. The distance between homogenously localized intercalating dyes within the DNA molecules was measured to be 34nm （n= 10; dye：DNAbp= 1：100） without photocleavage in 50mmol/L β-mercaptoethylamine buffer. The results were similar to those of the theoretical values without photocleavage in the base pairs of single DNA molecules below the diffraction limit. The results paved the way for an in-depth microscopic analysis of molecular variation with single λ-DNA molecules. With this method, it should be possible to analyze the exact base pair breakdown during various stages of cell apoptosis.

Material properties and machining performance of hybrid Ti_2AlN bulk material for micro electrical discharge machining

Mn+1AXn(MAX) phases are a family of nanolaminated compounds that possess unique combination of typical ceramic properties and typical metallic *** a member of MAX phase,Ti2 AlN bulk materials are attractive for some high-temperature *** synthesis,characteristics and machining performance of hybrid Ti2 AlN bulk materials were focused on in this *** bulk samples mainly consisting of Ti2 AlN MAX phase with density close to theoretic one were synthesized by a spark plasma sintering *** electron microscopy results indicate homogenous distribution of Ti2 AlN grains in the ***-hardness values are almost constant under different loads (6-6.5 GPa).A machining test was carried out to compare the effect of material properties on micro-electrical discharge machining (micro-EDM) performance for Ti2 AlN bulk samples and Ti6242 *** machining performance of the Ti2 AlN sample is better than that of the Ti6242 *** inherent mechanism was discussed by considering their electrical and thermal conductivity.

Due to highly underdetermined nature of Single Image Super-Resolution(SISR)problem,deep learning neural networks are required to be more deeper to solve the problem *** of deep neural networks successful in the Super-Resolution(SR)problem is ResNet which can render the capability of deeper networks with the help of skip ***,zero padding(ZP)scheme in the network restricts benefits of skip connections in SRResNet and its performance as the ratio of the number of pure input data to that of zero padded data *** this *** consider the ResNet with Partial Convolution based Padding(PCP)instead of ZP to solve SR *** training of deep neural networks using patch images is advantageous in many aspects such as the number of training image data and network complexities,patch image based SR performance is compared with single full image based *** experimental results show that patch based SRResNet SR results are better than single full image based ones and the performance of deep SRResNet with PCP is better than the one with ZP.

Brittle and ductile characteristics of intermetallic compounds in magnesium alloys: A large-scale screening guided by machine learning

In the present work,we have employed machine learning(ML)techniques to evaluate ductile-brittle(DB)behaviors in intermetallic compounds(IMCs)which can form magnesium(Mg)*** procedure was mainly conducted by a proxy-based method,where the ratio of shear(G)/bulk(B)moduli was used as a proxy to identify whether the compound is ductile or *** from compounds information(composition and crystal structure)and their moduli,as found in open databases(AFLOW),ML-based models were built,and those models were used to predict the moduli in other compounds,and accordingly,to foresee the ductile-brittle behaviors of these new *** results reached in the present work showed that the built models can effectively catch the elastic moduli of new *** was confirmed through moduli calculations done by density functional theory(DFT)on some compounds,where the DFT calculations were consistent with the ML prediction.A further confirmation on the reliability of the built ML models was considered through relating between the DB behavior in MgBe_(13) and MgPd_(2),as evaluated by the ML-predicted moduli,and the nature of chemical bonding in these two compounds,which in turn,was investigated by the charge density distribution(CDD)and electron localization function(ELF)obtained by DFT *** ML-evaluated DB behaviors of the two compounds was also consistent with the DFT calculations of CDD and *** findings and confirmations gave legitimacy to the built model to be employed in further prediction ***,as examples,the DB characteristics were investigated in IMCs that might from in three Mg alloy series,involving AZ,ZX and WE.

Mo and Ta addition in NbTiZr medium entropy alloy to overcome tensile yield strength-ductility trade-off

In this study,single-phase NbTiZr and NbTiZr(MoTa)_(0.1) medium-entropy alloys(MEAs)were investigated for their use in biomedical *** alloys were prepared by arc melting,and were then cold-rolled,annealed,and characterized in terms of phase analysis,mechanical properties,fractography,and wear *** alloys showed a single body-centered cubic phase with superior mechanical,and tribological properties compared to commercially available biomedical *** and Ta-containing MEAs showed higher tensile yield strength(1060±18 MPa)and higher tensile ductility(~20%),thus overcoming the strength-ductility trade-off with no signs of transformation-induced plasticity,twinning,or *** generalized stacking fault energy(GSFE)calculations on the{112}slip system by the first-principles calculations based on density functional theory showed that the addition of less than0.2 molar fraction of Mo and Ta lowers the GSFE *** behavior posits the increase in ductility of the alloy by facilitating slips although strength is also increased by solid solution *** wear resistance of both alloys against hardened steel surfaces was superior to that of commercial biomedical ***,we concluded that NbTiZr(MoTa)_(0.1)MEA with good tensile ductility is a potential candidate for biomedical implants.

Fossil-Winged Fruits of Fraxinus(Oleaceae) and Liriodendron(Magnoliaceae) from the Duho Formation,Pohang Basin,Korea

A total of 16 specimens of fossil-winged fruits were found from the Middle Miocene marine deposits, Duho Formation, Pohang Basin, Korea. They were identified into two structurally different groups： 15 specimens into a winged fruit of Fraxinus, and one specimen of Liriodendron. The most samaras （13 specimens） were identified as Fraxinus oishii, which is characterized by narrowly ovate or ovate-elliptic shapes that are 2.7-3.6 cm in length and 0.7-1 cm in width （i/w ratio=3.4-4）. The apexes of the Fraxinus oishii samara are round or slightly emarginated, and a seed of the samara is always located at the base, of which the general shape is narrow rhombic-ellipsoidal. The seed is 1.2-2 cm long and 0.5-0.7 cm wide. Two specimens are different from the samara of Fraxinus oishii. They have a 6.6 length/width ratio （3.3 cm long and 0.5 cm wide）, and thus, are temporarily classified into the Fraxinus sp. One specimen was recognized as a winged seed of Liriodendron meisenense. The wing is broadly lanceolate to elliptic in shape, has a smooth, acute apex, and is approximately 3 cm long and 0.7 cm wide. Samaras of Fraxinus oishii and Liriodendron meisenense were early reported from the Middle Miocene deposits from North Korea, but these specimens are the first discovery in South Korea. Further study of the Duho Formation may connect flora relationships between North and South Korea.
