Apoptosis and hypertrophy of cardiomyocytes are the primary causes of heart failure （HF）, a global leading cause of death, and are regulated through the complicated intracellular signaling network, limiting the development of effective treatments due to its complexity. To identify effective therapeutic strategies for HF at a system level, we develop a large-scale comprehensive mathematical model of the cardiac signaling network by integrating all available experimental evidence. Attractor landscape analysis of the network model identifies distinct sets of control nodes that effectively suppress apoptosis and hypertrophy of cardiomyocytes under ischemic or pressure overload-induced HF, the two major types of HF. Intriguingly, our system-level analysis suggests that intervention of these control nodes may increase the efficacy of clinical drugs for HF and, of most importance, different combinations of control nodes are suggested as potentially effective candidate drug targets depending on the types of HF. Our study provides a systematic way of developing mechanism-based therapeutic strategies for HF.

Predictors of poor outcomes in patients with wild mushroom-induced acute liver injury

To identify early predictive markers of poor outcomes in patients with acute liver injury from wild mushroom *** observational, retrospective record review involved adults aged ≥ 18 years admitted to emergency department with mushroom intoxication from January 2005 to December 2015. The diagnosis of mushroom intoxication was based on the following: (1) a positive history of recent wild mushroom intake (either raw or cooked); (2) the onset of gastrointestinal symptoms, such as watery diarrhea, vomiting, and/or abdominal pain, after ingestion; and (3) the exclusion of other possible causes of acute liver injury. Acute liver injury was defined by a > 5-fold elevation of liver enzymes or moderate coagulopathy [international normalized ratio (INR) > 2.0]. Clinical and laboratory findings were compared in survivors and *** 93 patients with mushroom intoxication, 23, 11 men (47.8%) and 12 women (52.2%), of median age 61 years, developed acute liver injury. The overall in-hospital mortality rate was 43.5% (10/23). Among the laboratory variables, mean serum alkaline phosphatase (73.38 ± 10.89 mg/dL vs 180.40 ± 65.39 mg/dL, P howed that total bilirubin concentration (OR = 3.58, 95%CI: 1.25-10.22), indirect/direct bilirubin ratio (OR = 0.14, 95%CI: 0.02-0.94) and aPTT (OR = 1.30, 95%CI: 1.04-1.63) were significantly associated with mortality. Background:Cholangiocarcinoma was considered as a dismal disease with very poor prognosis until *** is increasingly found due to increased life *** surgical and medical management were advanced recently,data on the prognosis,especially extrahepatic cholangio-carcinoma(ECC),were *** study aimed to identify clinicopathologic features and prognosis of patients with ***:Patients followed up and diagnosed with ECC between January 2014 and December 2016 at a tertiary hospital were included,whereas those with intrahepatic cholangiocarcinoma,gallbladder cancer,and ampullary cancer were ***:A total of 83 patients were followed after the treatment(49 men and 34 women;median age 73.3 years).Cancer location was classified as distal common bile duct(25 patients),proximal com-mon bile duct(24 patients),common hepatic duct(20 patients),and hilar(14 patients).About 14.5%of patients had history of another malignant neoplasm,and 24.1%patients had chronic ***-cal resection was performed in 54 patients(65%)and dysplasia was combined in 63%(34/54).Adjuvant chemotherapy was performed in 54%(29/54),but only 7 underwent palliative chemotherapy in 29 non-surgical *** median overall survival in all patients was 30.9 *** analyzing the treatment modality,median survival of adjuvant chemotherapy,surgery only,palliative chemotherapy,and support-ive care groups were 42.9,30.9,12.0,and 8.9 months,respectively(P<0.05).In the Cox regression analy-sis of survival,age,surgical resection,chemotherapy,and comorbidity were significant prognostic factors,and the comorbidity was the only significant prognostic factor in the multivariable analysis(hazard ratio[HR]=2.80;95%CI:1.32–5.95;P=0.007).In a subgroup analysis of surgical patients,the presence of dysplasia was a favorable prognostic factor in the multivariable analysis(HR=0.29;95%CI:0.09–0.91;P=0.033).Conclusions:The overall survival of patients with ECC wa

Seafood,especially sea cucumbers and oysters are an expensive delicacy in several Asian *** sun-drying of these products takes 3 to 4 days and fetches a lower market *** industrial drying machines used for seafood drying are unable to dry sea cucumbers and oysters without texture and color degradation as they are delicate,temperature-sensitive and have longer drying *** economical drying system that does not cause texture or color degradation is the heat pump drying system and is commonly applied for agriculture and fabric *** this study,the heat pump technology is applied to develop two hybrid dryer models(bottom discharge and front discharge)to dry shellfish and sea cucumbers in large scale for storage and *** model is tested in a laboratory-scale with wet sponges as input material for its dehumidification capacity and the power consumed to attain the target *** front discharge model is found to have rapid drying capability and *** fluid dynamic tools are used to study the hot air flow behavior and flow uniformity inside the drying *** front discharge model showed smooth uniform flow over the trays with fewer losses in the flow *** front discharge model with higher drying rate,lower power consumption and uniform airflow distribution over the trays is chosen for the industrial-scale design.

AIM To measure exogenous corticotropin-releasing factor(CRF)-induced motility of the isolated rat colon and to demonstrate the effect of pharmacologic inhibition on CRF-induced motility. METhoDS The isolated vascularly-perfused rat colon was used. Luminal pressure was monitored via microtip catheter pressure transducers in the proximal and distal colon. At first, exogenous CRF was administered in a stepwise manner and the concentration of CRF yielding maximal colonic motility was selected. After recording basal colonic motility, hexamethonium, phentolamine, propranolol, atropine and tetrodotoxin were infused into the isolated colon. Initially, only the test drug was infused; then, CRF was added. The motility index was expressed as percentage change over basal level. RESULTS Administration of 1.4, 14.4, 144 and 288 pmol/L CRF progressively increased colonic motility in the proximal and distal colon. Infusion of atropine or tetrodotoxin reduced CRF-induced motility of both the proximal and distal colon, whereas hexamethonium, phentolamine and propranolol had no *** CRF-induced colonic motility appears to be mediated by local cholinergic signaling via muscarinic receptors. Muscarinic receptors are potential targets for counteracting CRF-induced colonic hypermotility.

Metaverse is one of the main technologies in the daily lives of several people,such as education,tour systems,and mobile application ***,the number of users of mobile metaverse applications is increasing owing to the merit of accessibility *** provide an improved service,it is important to analyze online reviews that contain user *** previous studies have utilized traditional methods,such as the structural equation model(SEM)and technology acceptance method(TAM)for exploring user satisfaction,using limited survey *** methods may not be appropriate for analyzing the users of mobile *** overcome this limitation,several researchers perform user experience analysis through online reviews and star ***,some online reviews occasionally have inconsistencies between the star rating and the sentiment of the *** variation disturbs the performance of machine *** alleviate the inconsistencies,Valence Aware Dictionary and sEntiment Reasoner(VADER),which is a sentiment classifier based on lexicon,is *** current study aims to build a more accurate sentiment classifier based on machine learning with *** this study,five sentiment classifiers are used,such as Naïve Bayes,K-Nearest Neighbors(KNN),Logistic Regression,Light Gradient Boosting Machine(LightGBM),and Categorical boosting algorithm(Catboost)with three embedding methods(Bag-of-Words(BoW),Term Frequency-Inverse Document Frequency(TF-IDF),Word2Vec).The results show that classifiers that apply VADER outperform those that do not apply VADER,excluding one classifier(Logistic Regression with Word2Vec).Moreover,LightGBM with TF-IDF has the highest accuracy 88.68%among other models.
