Thermogelling hydrogels,such as poly(N-isopropylacrylamide)[P(NiPAAm)],provide tunable constructs leveraged in many regenerative biomaterial ***,our lab developed the crosslinker poly(glycolic acid)-poly(ethylene glycol)-poly(glycolic acid)-di(but-2-yne-1,4-dithiol),which crosslinks P(NiPAAm-co-glycidyl methacrylate)via thiol-epoxy reaction and can be functionalized with azide-terminated peptides via alkyne-azide click *** study’s aim was to evaluate the impact of peptides on the physicochemical properties of the *** physicochemical properties of the hydrogels including the lower critical solution temperature,crosslinking times,swelling,degradation,peptide release and cytocompatibility were *** gels bearing peptides increased equilibrium swelling indicating hydrophilicity of the hydrogel *** sol fractions were found for all groups,indicating that inclusion of peptides does not impact ***,the inclusion of a matrix metalloproteinase-sensitive peptide allowed elucidation of whether release of peptides from the network was driven by hydrolysis or enzymatic *** hydrophilicity of the network determined by the swelling behavior was demonstrated to be the most important factor in dictating hydrogel behavior over *** study demonstrates the importance of characterizing the impact of additives on the physicochemical properties of *** characteristics are key in determining design considerations for future in vitro and in vivo studies for tissue regeneration.

Multiple z-Score Based Method for Noninvasive Prenatal Test Using Cell-Free DNA in Maternal Plasma

Objective: To improve the detecting accuracy of chromosomal aneuploidy of fetus by non-invasive prenatal testing (NIPT) using next generation sequencing data of pregnant women’s cell-free DNA. Methods: We proposed the multi-Z method which uses 21 z-scores for each autosomal chromosome to detect aneuploidy of the chromosome, while the conventional NIPT method uses only one z-score. To do this, mapped read numbers of a certain chromosome were normalized by those of the other 21 chromosomes. Average and standard deviation (SD), which are used for calculating z-score of each sample, were obtained with normalized values between all autosomal chromosomes of control samples. In this way, multiple z-scores can be calculated for 21 autosomal chromosomes except oneself. Results: Multi-Z method showed 100% sensitivity and specificity for 187 samples sequenced to 3 M reads while the conventional NIPT method showed 95.1% specificity. Similarly, for 216 samples sequenced to 1 M reads, Multi-Z method showed 100% sensitivity and 95.6% specificity and the conventional NIPT method showed a result of 75.1% specificity. Conclusion: Multi-Z method showed higher accuracy and robust results than the conventional method even at low coverage reads.

Outcome of transarterial chemoembolization-based multi-modal treatment in patients with unresectable hepatocellular carcinoma

AIM:To investigate the efficacy and safety of transarterial chemoembolization(TACE)-based multimodal treatment in patients with large hepatocellular carcinoma(HCC).METHODS:A total of 146 consecutive patients were included in the analysis,and their medical records and radiological data were reviewed ***:In total,119 patients received TACE-based multi-modal treatments,and the remaining 27 received conservative *** survival(P<0.001)and objective tumor response(P=0.003)were significantly better in the treatment group than in the conservative *** subgroup analysis,survival benefits were observed not only in the multi-modal treatment group compared with the TACE-only group(P=0.002)but also in the surgical treatment group compared with the loco-regional treatment-only group(P<0.001).Multivariate analysis identified tumor stage(P<0.001)and tumor type(P=0.009)as two independent pre-treatment factors for *** adjusting for significant pre-treatment prognostic factors,objective response(P<0.001),surgical treatment(P=0.009),and multi-modal treatment(P=0.002)were identified as independent post-treatment prognostic ***:TACE-based multi-modal treatments were safe and more beneficial than conservative *** surgery after successful downstaging resultedin long-term survival in patients with large,unresectable HCC.

地震引起的水坝损坏将对社会造成巨大损失。因此,应该进行大坝及相关附属设施的抗震性能评估和维护,以保护人民的生命和财产。在韩国,随着2017年修订的抗震设计准则的发布,水坝的抗震设计准则也在修订中,以反映修订的抗震设计准则的变更。根据韩国水资源公社对水坝及其附属设施进行抗震性能评估的经验和结果,本文详细讨论了2011年发布的现行水坝的抗震设计准则存在的问题及后续的问题。此外,本文还介绍了韩国水资源公社经营的水坝及其附属设施的抗震性能评估现状。

Risk factors for hepatocellular carcinoma in patients with drug-resistant chronic hepatitis B

AIM:To investigate the risk factors and characteristics of hepatocellular carcinoma(HCC) in the patients with drug-resistant chronic hepatitis B(CHB).METHODS:A total of 432 patients with drug-resistant CHB were analyzed retrospectively from January 2004to December 2012. The patients were divided into two groups:the HCC group(n = 57) and the non-HCC group(n = 375). Two groups compared using logistic regression for various patients and viral characteristics in order to identify associated risk factors for ***,patient and tumor characteristics of HCC patients with na ve CHB(N group,n = 117) were compared to the HCC group(R group,n = 57) to identify any difference in HCC characteristics between ***:A significant difference was found for age,platelet count,alpha-fetoprotein(AFP),positivity of HBeAg,seroconversion rate of HBeAg,virologic response,the Child-Pugh score,presence of rtM204I,and the duration of antiviral treatment in non-HCC and HCC group. Cirrhosis,age(> 50 years),HBeAg(+),virologic non-responder status,and rtM204I mutants were independent risk factors for the development of HCC. The R group had lower serum C-reactive protein(CRP) and AFP levels,earlier stage tumors,and a shorter mean tumor surveillance period than the N group. However,the total follow-up duration was not significantly different between the two ***:13.2% of patients with drug-resistant CHB developed HCC. Age,cirrhosis,YIDD status,HBeAg status,and virologic response are associated with risk of HCC. Patients with drug-resistant CHB and these clinical factors may benefit from closer HCC surveillance.

Dofs(DNA-binding with one zinc finger proteins)是植物特有的转录因子家族。Dof蛋白参与许多生物过程,如植物激素的产生,种子发育和环境适应。Dof蛋白已经在多种植物中被鉴定,但在辣椒中尚未开展类似的研究。我们在辣椒中鉴定出了33个可能的Dof基因(CaDofs)。为了了解CaDof基因的特征,我们分析了其系统进化关系、蛋白质基序和进化历史。我们将含有25个基序且分布在8条染色体上的33个CaDof基因分成4个族。我们发现CaDof基因的扩张可追溯到最近的复制事件。茄科种系形成之前,片段复制在CaDof家族基因中占主导地位。通过RNA-seq分析获得了CaDof基因表达谱,发现在对包括两种TMV菌株,辣椒斑驳病毒和辣椒疫霉菌的生物胁迫响应过程中,Ca Dof家族基因表现出了时间和病原特异性变化,这表明CaDof基因的功能具有多样性。这些结果不仅为研究Dof基因在生物胁迫中的响应提供有价值的信息,同时也有助于更好地理解Dof基因在辣椒和其他物种中的多重功能。

Clinical significance and risk factors of postembolization fever in patients with hepatocellular carcinoma

AIM:To investigate tumor response and survival in patients with postembolization fever(PEF) and to determine the risk factors for ***:Four hundred forty-three hepatocellular carcinoma(HCC) patients who underwent the first session of transcatheter arterial chemoembolization(TACE) between January 2005 and December 2009 were analyzed *** was defined as a body temperature greater than 38.0 ℃ that developed within 3 d of TACE without evidence of *** tumor progression-free interval was defined as the interval from the first TACE to the second TACE based on mRECIST *** staging was based on the American Joint Committee on Cancer tumor,node,metastases(TNM) classification of malignant *** patients were admitted before their 1 st TACE treatment,and blood samples were obtained from all patients before and after *** variables and host-related variables were compared between two groups:patients with PEF vs patients without ***,variables related to 20-mo mortality and tumor progression-free survival were ***:The study population comprised 370(85.4%) men and 73(14.6%) women with a mean age of 62.29 ± 10.35 years.A total of 1836 TACE sessions were conducted in 443 patients,and each patient received between 1 and 27(mean:4.14 ± 3.57) TACE *** mean follow-up duration was 22.23 ± 19.6 mo(range:0-81 mo).PEF developed in 117 patients(26.41%) at the time of the first TACE *** was not associated with 20-mo survival(P = 0.524) or computed tomography(CT) response(P = 0.413) in a univariate analysis.A univariate analysis further indicated that diffuse-type HCC(P = 0.021),large tumor size(≥ 5 cm)(P = 0.046),lipiodol dose(≥ 7 mL,P = 0.001),poor blood glucose control(P = 0.034),alanine aminotransferase(ALT) value after TACE(P = 0.004) and C-reactive protein(CRP) value after TACE(P = 0.036) served as possible risk factors correlated with *** ALT value after TACE(P = 0.02

Biopathologic features and clinical significance of micrometatasis in the lymph node of early gastric cancer

AIM: To evaluate the biopathologic features and clinical significance of nodal micrometastasis(MI) in early gastric cancer(EGC).METHODS: Among 1022 EGC patients who underwent gastrectomy with lymphadenectomy of D1 + β or more from March 2001 to December 2005 at the Korean National Cancer Center, available nodal metastasis was found in 90 p T1N1 patients. Nodal metastasis was confirmed by immunohistochemistry(IHC) with cytokeratin and patients were classified into MI and macrometastasis(MA) groups based on the main tumor burden according to the 6th International Union Against Cancer/American Joint Committee on Cancer staging system; the main tumor burden with a diameter of greater than 0.2 mm but no greater than 2 mm as MI, and greater than 2 mm as MA of the representative metastatic node. Proliferative and apoptotic activities of the primary tumor and the nodal metastasis were measured by IHC with Ki-67 and terminal deoxynucleotidyl transferase d UTP nick end labeling, respectively. Biopathologic and clinical features of the patients were analyzed and compared between MI and MA groups. Patients with recurrence were compared with those without recurrence to identify risk factors for ***: Thirty-seven patients showed MI and the other 53 patients revealed MA in the lymph node; the incidence of patients with MI and MA was 41.1% and 58.9%. The main tumor burden was 0.9 and 4.6 mm in the representative metastatic node, respectively. Japanese N2 stations were more frequently involved in MA group(20.9%) than in MI group(10.3%) butthe difference was not statistically different(P = 0.338). Proliferative and apoptotic activities of MI were decreased than those of MA(26.7% vs 40.5%, P = 0.004 and 1.0% vs 3.0%, P < 0.001, respectively). However, nodal MI in the current study showed a relatively high proliferative activity and an equivalent apoptotic activity compared to other cancers in the previously published studies.
