Alterations of Gefitinib Pharmacokinetics by Co-administration of Herbal Medications in Rats

Objective： To evaluate the potential pharmacokinetic interactions of the anticancer agent gefitinib （Iressae） and the oriental medications Guipi Decoction （归脾汤, GPD, Guibi-tang in Korean） and Bawu Decoction （八物汤, BWD, Palmul-tang in Korean）. Methods： Methylceliulose （MC, control）, GPD （1,200 mg/kg）, or BWD （6,000 mg/kg） was orally administered to rats either as a single dose or multiple doses prior to gefitinib administration. To examine the effects of a single dose of the herbal medicines, gefitinib （10 mg/kg） was orally administered after 5 min or 1 h of MC or the herbal medicine pretreatments. To examine the effects of the multiple doses of the herbal medicines, gefltinib （10 mg/kg） was orally administered following 7 consecutive days of the administration of MC or each herbal medicine. The plasma concentrations of gefitinib were determined with liquid chromatography-tandem mass spectrometry assay. The plasma concentration-time profiles of gefitinib were analyzed with a noncompartmental analysis. Results： Gefitinib was rapidly absorbed and showed a mono- exponential decline with an elimination half-life of 3.7-4.1 h. The pharmacokinetics of gefitinib was not affected by GPD pretreatment. However, a significantly lower maximum plasma concentration （Crux, P〈0.05） and area under the curve （P〈0.05）, and a delayed time to reach Cmax （Tmax, P〈0.01） were observed in both single- and multiple- dose BWD-pretreated rats compared with the control rats. Conclusions： BWD and not GPD might delay and interfere with gefitinib absorption. Further evaluations of the clinical significance of these findings are needed.

Genetic variation of the PSCA gene(rs2294008) is not associated with the risk of prostate cancer

Prostate stem cell antigen（PSCA） is a cell-membrane glycoprotein consisting of 123 amino acids and highly expressed in the prostate,but there have been few reports on the relationship between rs2294008 of PSCA and prostate cancer in the literature.Therefore,we evaluated the association between rs2294008 and the risk of prostate cancer.A total of 240 prostate cancer patients and 306 controls（patients with benign prostatic hyperplasia） were enrolled.Genotype analysis of rs2294008 of PSCA was performed using PCR.Logistic regression analysis was performed according to the genotype of PSCA rs2294008.We found that CT and TT genotypes were associated with an insignificant risk of prostate cancer compared with the CC genotype（P= 0.627 and 0.397,respectively）.In addition,there was no significant difference in rs2294008 according to clinicopathological parameters,such as age,Gleason score,prostate-specific antigen（PSA）,stage,and metastasis in prostate cancer（P 〉 0.05 for each）.Age,Gleason score,PSA,pathologic stage,and metastasis did not modify the association between PSCA and the risk of prostate cancer（each P 〉 0.05 for each）.Taken together,the genetic polymorphism of PSCA rs2294008 was not associated with the risk of prostate cancer.Our results suggest that rs2294008 may not play a role in prostate carcinogenesis.

Gene editing particle system as a therapeutic approach for drug-resistant colorectal cancer

The epidermal growth factor receptor(EGFR)pathway plays an important role in the progression of colorectal cancer(CRC).Anti-EGFR drugs based on antibodies have been widely used for treating CRC through inducing the cell death pathway.However,the majority of CRC patients will inevitably develop drug-resistance during anti-EGFR drug treatment,which is mainly caused by a point mutation in the KRAS oncogene.We developed a nanoliposomal(NL)particle containing the Cas9 protein and a single-guide RNA(sgRNA)complex(Cas9-RNP),for genomic editing of the KRAS mutation.The NL particle is composed of bio-compatible lipid compounds that can effectively encapsulate Cas9-RNP.By modifying the NL particle to include the appropriate antibody,it can specifically recognize EGFR expressing CRC and effectively deliver the gene-editing complexes.The conditions of NL treatment were optimized using a KRAS mutated CRC in vivo mouse model.Mice with KRAS-mutated CRC showed drug resistance against cetuximab,a therapeutic antibody drug.After treating the mice with the KRAS gene-editing NL particles,the implanted tumors showed a dramatic decrease in size.Our results demonstrated that this genomic editing complex has great potential as a therapeutic carrier system for the treatment of drug-resistant cancer caused by a point mutation.

Pediatric schwannoma of the tongue:A case report and review of literature

BACKGROUND Neurogenic tumors account for about ten percent of all tumors of childhood,and benign tumor originating from Schwann cells is rare in peripheral nerves.Schwannoma of the tongue is quite rare in children.CASE SUMMARY We present the case of an 8-year-old male with schwannoma in the anterolateral tongue.The mass was slow-growing for one year with no pain and discomfort.He underwent transoral mass excision under general anesthesia.Gross examination revealed a smooth surfaced,17 mm×14 mm×7 mm sized,encapsulated nodule with a clear resection margin.Schwannoma of the tongue was confirmed by the pathological exam.He reported no motor or sensory change,such as dysgeusia or paresthesia,or phonation difficulty during postoperative 12 mo follow-up.CONCLUSION Schwannoma of the tongue is a rare benign neoplasm in childhood.If a painless firm mass is encountered in the tongue of a child,solid tumors like schwannoma should be considered in the differential diagnosis.

Porous 2D materials with high conductivity and large surface area have been proposed for potential electromagnetic interference(EMI)shielding materials in future mobility and wearable applications to prevent signal noise,transmission inaccuracy,system malfunction,and health hazards.Here,we report on the synthesis of lightweight and flexible flash-induced porous graphene(FPG)with excellent EMI shielding performance.The broad spectrum of pulsed flashlight induces photo-chemical and photo-thermal reactions in polyimide films,forming 5×10 cm^(2)-size porous graphene with a hollow pillar structure in a few milliseconds.The resulting material demonstrated low density(0.0354 g cm^(−3))and outstanding absolute EMI shielding effectiveness of 1.12×10^(5) dB cm^(2) g^(−1).The FPG was characterized via thorough material analyses,and its mechanical durability and flexibility were confirmed by a bending cycle test.Finally,the FPG was utilized in drone and wearable applications,showing effective EMI shielding performance for internal/external EMI in a drone radar system and reducing the specific absorption rate in the human body.

Background:Aducanumab(Adu),which is a human IgG1 monoclonal antibody that targets oligomer and fibril forms of beta-amyloid,has been reported to reduce amyloid pathology and improve impaired cognition after administration of a high dose(10 mg/kg)of the drug in Alzheimer’s disease(AD)clinical trials.The purpose of this study was to investigate the effects of a lower dose of Adu(3 mg/kg)with enhanced delivery via focused ultrasound(FUS)in an AD mouse model.Methods:The FUS with microbubbles opened the blood-brain barrier(BBB)of the hippocampus for the delivery of Adu.The combined therapy of FUS and Adu was performed three times in total and each treatment was performed biweekly.Y-maze test,Brdu labeling,and immunohistochemical experimental methods were employed in this study.In addition,RNA sequencing and ingenuity pathway analysis were employed to investigate gene expression profiles in the hippocampi of experimental animals.Results:The FUS-mediated BBB opening markedly increased the delivery of Adu into the brain by approximately 8.1 times in the brains.The combined treatment induced significantly less cognitive decline and decreased the level of amyloid plaques in the hippocampi of the 5×FAD mice compared with Adu or FUS alone.Combined treatment with FUS and Adu activated phagocytic microglia and increased the number of astrocytes associated with amyloid plaques in the hippocampi of 5×FAD mice.Furthermore,RNA sequencing identified that 4 enriched canonical pathways including phagosome formation,neuroinflammation signaling,CREB signaling and reelin signaling were altered in the hippocami of 5×FAD mice receiving the combined treatment.Conclusion:In conclusion,the enhanced delivery of a low dose of Adu(3 mg/kg)via FUS decreases amyloid deposits and attenuates cognitive function deficits.FUS-mediated BBB opening increases adult hippocampal neurogenesis as well as drug delivery.We present an AD treatment strategy through the synergistic effect of the combined therapy of FUS and Adu.

Hybrid functional IrO_2-TiO_2 thin film resistor prepared by atomic layer deposition for thermal inkjet printheads

IrO2-TiO2 thin films were prepared by atomic layer deposition using Ir(EtCp)(COD) and titanium isopropoxide (TTIP). The resistivity of IrO2-TiO2 thin films can be easily controlled from 1 500 to 356.7 μΩ·cm by the IrO2 intermixing ratio from 0.55 to 0.78 in the IrO2-TiO2 thin films. The low temperature coefficient of resistance(TCR) values can be obtained by adopting IrO2-TiO2 composite thin films. Moreover, the change in the resistivity of IrO2-TiO2 thin films was below 10% even after O2 annealing process at 600 °C. The step stress test results show that IrO2-TiO2 films have better characteristics than conventional TaN0.8 heater resistor. Therefore, IrO2-TiO2 composite thin films can be used as a heater resistor material in thermal inkjet printhead.

Risks of non-conservative design according to ASME B31.1 for high-temperature piping subjected to long-term operation in the creep range

This study investigates the risks of non-conservative piping design according to ASME B31.1 for hightemperature piping subjected to long-term operation at high temperature in a creep regime based on a sensitivity analysis of the hold time. Design evaluations of hightemperature piping were conducted over a range of hold times in the creep regime according to ASME B31.1,which implicitly considers the creep effects, and the French high-temperature design code of the RCC-MRx, which explicitly considers the creep effects. Conservatisms were quantified among the codes in terms of the hold times. In the case of B31.1, the design evaluation results do not change depending on the hold time at high temperature,whereas in the case of RCC-MRx, they do. It was shown that the design limits of RCC-MRx were exceeded when the hold time exceeded certain values, whereas those of B31.1 were satisfied regardless of the hold times. Thus, the design evaluations according to B31.1 did not consistently yield conservative results and might lead to non-conservative results in the case of long-term operations in the creep range.

Comparative study for predictability of type 1 gastric variceal rebleeding after endoscopic variceal ligation:High-frequency intraluminal ultrasound study

BACKGROUND The efficacy of endoscopic ultrasonography for the follow-up of gastric varices treated with endoscopic variceal ligation(EVL)has not been established.AIM To evaluate the diagnostic correlation of esophagogastroduodenoscopy(EGD)and high-frequency intraluminal ultrasound(HFIUS)for type 1 gastric varices(GOV1)after EVL and to identify the predictability for rebleeding of EGD and HFIUS.METHODS In liver cirrhosis patients with GOV1,we performed endoscopic follow-up using EGD and HFIUS synchronously after EVL for hemorrhage from GOV1.Endoscopic grading and red color signs were analyzed using EGD,and the largest variceal cross-sectional areas were measured using HFIUS.In addition,1-year follow-up was performed.Variceal rebleeding was defined as the presence of hematemesis,hematochezia,or melena without other evidence of bleeding on endoscopic follow-up.RESULTS In 26 patients with GOV1,variceal cross-sectional areas on HFIUS of GOV1 was poorly correlated with EGD grading of GOV1(r=0.36).In 17 patients who completed the 1-year follow-up,variceal cross-sectional areas on HFIUS was a good predictor of subsequent rebleeding,whereas EGD grading was not a predictor of subsequent rebleeding.CONCLUSION HFIUS measurement is more predictive of GOV1 rebleeding than EGD grading,so HFIUS measurement may be necessary for endoscopic follow-up after EVL in patients with GOV1.
