Milling degree affects the fermentation properties of rice:perspectives from the composition of nutrients and gut microbiota via in vitro fermentation

Fermentation substrates of rice with different milling degrees(MDs) were prepared and fermented with human feces to compare their fermentation properties and effects on gut microbiota.MD 0s,MD 5s and MD 60s represented brown rice,moderately-milled rice and white rice,respectively.After in vitro fermentation,the MD 5s group showed higher starch utilization,compared with the MD 0s and 60s groups evaluated by Fourier transform infrared spectrometer,and confocal laser scanning microscope.Effects of fermentation substrates of rice with different MDs on gut microbiota were evaluated by 16S rDNA sequencing.All the sample groups reduced the pH and produced short-chain fatty acids(SCFAs) and branched-chain fatty acids.The MD 5s group exhibited higher α-diversity than the MD 0s and 60s groups.Abundances of Phascolarctobacterium,Blautia and norank_f_Ruminococcaceae were higher in the MD 0s and 5s groups,compared with the MD 60s group.These bacteria were also positively correlated with the SCFAs production via Spearman correlation analysis.In vitro culture assay revealed that fermentation substrates of MD 0s and 5s promoted the growth of two probiotics(Akkermansia muciniphila and Bifidobacterium adolescentis).Our results showed that moderate milling might be an appropriate way to produce rice products with richer nutrients and better fermentation properties.

BACKGROUND liver fibrosis is a compensatory response during the tissue repair process in chronic liver injury,and finally leads to liver cirrhosis or even hepatocellular carcinoma.The pathogenesis of hepatic fibrosis is associated with the progressive accumulation of activated hepatic stellate cells(HSCs),which can transdiffer-entiate into myofibroblasts to produce an excess of the extracellular matrix(ECM).Myofibroblasts are the main source of the excessive ECM responsible for hepatic fibrosis.Therefore,activated hepatic stellate cells(aHSCs),the principal ECM producing cells in the injured liver,are a promising therapeutic target for the treatment of hepatic fibrosis.AIM To explore the effect of taurine on aHSC proliferation and the mechanisms involved.METHODS Human HSCs(LX-2)were randomly divided into five groups:Normal control group,platelet-derived growth factor-BB(PDGF-BB)(20 ng/mL)treated group,mmol/L,respectively)with PDGF-BB(20 ng/mL)treated group.Cell Counting Kit-8 method was performed to evaluate the effect of taurine on the viability of aHSCs.Enzyme-linked immunosorbent assay was used to estimate the effect of taurine on the levels of reactive oxygen species(ROS),malondialdehyde,glutathione,and iron concen-tration.Transmission electron microscopy was applied to observe the effect of taurine on the autophagosomes and ferroptosis features in aHSCs.Quantitative real-time polymerase chain reaction and Western blot analysis were performed to detect the effect of taurine on the expression ofα-SMA,Collagen I,Fibronectin 1,LC3B,ATG5,Beclin 1,PTGS2,SLC7A11,and p62.RESULTS Taurine promoted the death of aHSCs and reduced the deposition of the ECM.Treatment with taurine could alleviate autophagy in HSCs to inhibit their activation,by decreasing autophagosome formation,downregulating LC3B and Beclin 1 protein expression,and upregulating p62 protein expression.Meanwhile,treatment with taurine triggered ferroptosis and ferritinophagy to eliminate aHSCs characterized by iron overload,lipid ROS accumu-lation,glutathi

Oat avenanthramide B alleviates high-fat diet-induced obesity via regulating fatty acid metabolism involved in gut bacteria and fungi remodeling

Oat avenanthramides (AVNs) have been found to exhibit novel lipid-lowering effects. However， the mechanism remains unclear. In this study， the effect of avenanthramide B (AVN B)， as one of the major AVNs， on high-fat diet (HFD)-induced mice was investigated. Results showed that AVN B significantly inhibited weight gain and improved hepatic and serum lipid biochemical indices. Hepatic RNA-Sequencing analysis suggested that AVN B significantly modulates fatty acid (FA) metabolism. Hepatic real-time qualitative polymerase chain reaction (RT-qPCR) and Western blot results indicated that AVN B could alleviate FA synthesis by activating the AMPK- SREBP1c-FAS， and increase FA oxidation by activating the AMPK/CPT1A and PPARα. Additionally， AVN B had a regulating effect on ileum lipid metabolism by inhibiting intestinal cell differentiation and down-regulating the expression levels of FA absorption-related protein and gene. Moreover， AVN B promoted the growth of beneficial bacteria and fungi such as Coriobacteriaceae＿UCG-002， Parvibacter， Enterococcus， and Aspergillus， while decreasing the abundance of Roseburia， unclassified＿f＿Lachnospiraceae Cladosporium， Eurotium， unclassified＿f＿Aspergillaceae and unclassified＿f＿Ceratocystidaceae. All these results provided new points of the lipid-lowing mechanism of AVNs and oats via the gut-liver axis.

Current state and influencing factors of social alienation in patients with stress urinary incontinence

Objective To understand the current level of social alienation among patients with stress urinary incontinence(Sul)andanalyse the influencing factors to provide a foundation for better clinical support for patients'reintegration into society.Methods We conductedacomprehensive surveyand analysis involving 230 patients with SUl using a general information questionnaire,a general sense of alienation scaleandasocial impactscale.Results The general sense of alienation scale score of patientswith SUlwas25.43±13.994,whilethe social impact score was 39.25±12.167.Multiple linear regression analysis revealed that marital status,primary caregiver,presence of concurrent illnesses and severity of urinary incontinence were the key influencing factors contributing to SUl-related social alienation(plienation.Tailored interventions are recommended,especiallyfor individuals who are divorced,rely on friends or relatives as primary caregivers,have concurrentillnesses or experience severe Sul,to enhance their social integration.

Geometric phase-encoded stimuli-responsive cholesteric liquid crystals for visualizing real-time remote monitoring:humidity sensing as a proof of concept

liquid crystals are a vital component of modern photonics,and recent studies have demonstrated the exceptional sensing properties of stimuli-responsive cholesteric liquid crystals.However,existing cholesteric liquid crystal-based sensors often rely on the naked eye perceptibility of structural color or the measurement of wavelength changes by spectrometric tools,which limits their practical applications.Therefore,developing a platform that produces recognizable sensing signals is critical.In this study,we present a visual sensing platform based on geometric phase encoding of stimuli-responsive cholesteric liquid crystal polymers that generates real-time visual patterns,rather than frequency changes.To demonstrate this platform’s effectiveness,we used a humidity-responsive cholesteric liquid crystal polymer film encoded with a q-plate pattern,which revealed that humidity causes a shape change in the vortex beam reflected from the encoded cholesteric liquid crystal polymers.Moreover,we developed a prototype platform towards remote humidity monitoring benefiting from the high directionality and long-range transmission properties of laser beams carrying orbital angular momentum.Our approach provides a novel sensing platform for cholesteric liquid crystals-based sensors that offers promising practical applications.The ability to generate recognizable sensing signals through visual patterns offers a new level of practicality in the sensing field with stimuli-responsive cholesteric liquid crystals.This platform might have significant implications for a broad readership and will be of interest to researchers working in the field of photonics and sensing technology.

Pre-mating nitenpyram exposure in male mice leads to depression-like behavior in offspring by affecting tryptophan metabolism in gut microbiota

Several studies have confirmed that the health status of the paternal affects the health of the offspring,however,it remains unknown whether paternal exposure to pesticides affect the offspring health.Here,we used untargeted metabolomics and 16S rRNA sequencing technology,combined with tail suspension test and RT-qPCR to explore the effects of paternal exposure to nitenpyram on the neurotoxicity of offspring.Our results found that the paternal exposure to nitenpyram led to the offspring’s depressive-like behaviors,accompanied by the reduction of tryptophan content and the disorder of microbial abundance in the gut of the offspring.Further,we determined the expression of tryptophan metabolism-related genes tryptophanase(tnaA)and tryptophan hydroxylase 1(TpH1)in gut bacteria and colonic tissues.We found that tryptophan is metabolized to indoles rather than being absorbed into colonocytes,which coursed the reduce of tryptophan availability after nitenpyram exposure.In conclusion,our study deepens our understanding of the intergenerational toxic effects of pesticides.

Large animal models for Huntington's disease research

Huntington'sdisease(HD)isahereditary neurodegenerative disorder for which there is currently no effectivetreatmentavailable.Consequently,the development of appropriate disease models is critical to thoroughly investigate disease progression.The genetic basis of HD involves the abnormal expansion of CAG repeats in the huntingtin(HTT)gene,leading to the expansion of a polyglutamine repeat in the HTT protein.Mutant HTT carrying the expanded polyglutamine repeat undergoes misfolding and forms aggregates in the brain,which precipitate selective neuronal loss in specific brain regions.Animal models play an important role in elucidating the pathogenesis of neurodegenerative disorders such as HD and in identifying potential therapeutic targets.Due to the marked species differences between rodents and larger animals,substantial efforts have been directed toward establishing large animal models for HD research.These models are pivotal for advancing the discovery of novel therapeutic targets,enhancing effective drug delivery methods,and improving treatment outcomes.We have explored the advantages of utilizing large animal models,particularly pigs,in previous reviews.Since then,however,significant progress has been made in developing more sophisticated animal models that faithfully replicate the typical pathology of HD.In the current review,we provide a comprehensive overview of large animal models of HD,incorporating recent findings regarding the establishment of HD knock-in(KI)pigs and their genetic therapy.We also explore the utilization of large animal models in HD research,with a focus on sheep,non-human primates(NHPs),and pigs.Our objective is to provide valuable insights into the application of these large animal models for the investigation and treatment of neurodegenerative disorders.

Cuproptosis mediates copper-induced testicularspermatogenic cell death

Cuproptosis,a novel mechanism of programmed cell death,has not been fully explored in the context of spermatogenic cells.Thisstudy investigated the potential involvement of cuproptosis in spermatogenic cell death using a mouse model of copper overload.Sixty male Institute of Cancer Research(ICR)mice were randomly divided into four groups that received daily oral gavage withsodium chloride(control)or copper sulfate(CuSO4)at 50 mg kg−1,100 mg kg−1,or 200 mg kg−1,for 42 consecutive days.Micesubjected to copper overload exhibited a disruption in copper homeostasis.Additionally,significant upregulated expression of keycuproptosis factors was accompanied by a significant rise in the rates of testicular tissue cell apoptosis.Immunohistochemicalanalysis revealed the presence of ferredoxin 1(Fdx1)in Sertoli cells,Leydig cells,and spermatogenic cells at various stages oftesticular development,and the Fdx1-positive staining area was significantly increased in copper-overloaded mice.Mitochondrialdysfunction and decreased adenosine triphosphate levels were also observed,further implicating mitochondrial damage undercuproptosis.Further analyses revealed pathological lesions and blood−testis barrier destruction in the testicular tissue,accompaniedby decreased sperm concentration and motility,in copper-overloaded mice.In summary,our results indicate that copper-overloadedmice exhibit copper homeostasis disorder in the testicular tissue and that cuproptosis participates in spermatogenic cell death.These findings provide novel insights into the pathogenic mechanisms underlying spermatogenic cell death and provide initialexperimental evidence for the occurrence of cuproptosis in the testis.

Uniqueness Problem for the Backward Differential Equation of a Continuous-State Branching Process

The distributional properties of a multi-dimensional continuous-state branching process are determined by its cumulant semigroup, which is defined by the backward differential equation.We provide a proof of the assertion of Rhyzhov and Skorokhod(Theory Probab. Appl., 1970) on the uniqueness of the solutions to the equation, which is based on a characterization of the process as the pathwise unique solution to a system of stochastic equations.

Open surgical approach for two coincidental splenic artery aneurysms:a case report

Several factors can contribute to the formation of aneurysms,including hemodynamic changes,polyarteritis nodosa,bacterial endocarditis,vasculitis,fibromuscular dysplasia,vascular malformation,and cystic medial necrosis.^([1,2]) Surgery is recommended for splenic artery aneurysms (SAAs) greater than 25 mm in diameter,and several surgical approaches are used,including open surgery,laparoscopic surgery,and percutaneous embolization.Laparoscopic surgery might be associated with an increased risk of pancreatic leakage compared to the open surgery approach.
