AIM： To investigate c-met expression during early pancreatic carcinogenesis. METHODS： We used 46 bulk tissues and 36 microdissected samples, including normal pancreas, chronic pancreatitis, and pancreatic cancer, for quantitative realtime reverse transcription-polymerase chain reaction. RESULTS： In bulk tissue analyses, pancreatic cancer tissues expressed significantly higher levels of c-met than did chronic pancreatitis and normal pancreas tissues. c-met levels did not differ between chronic pancreatitis and normal pancreas tissues. In microdissection-based analyses, c-met was expressed at higher levels in microclissected pancreatic cancer cells and pancreatitisaffected epithelial cells than in normal ductal epithelial cells （both, P 〈 0.01）. Interestingly, pancreatitis-affected epithelial cells expressed levels of c-met similar to those of pancreatic cancer cells. CONCLUSION： Overexpression of c-met occurs during the early stage of pancreatic carcinogenesis, and a single alteration of c-met expression is not sufficient for progression of chronic pancreatitis-affected epithelial cells to pancreatic cancer cells.

Retinal ischemia causes several vision-threatening diseases, including diabetic retinopathy, retinal artery occlusion, and retinal vein occlusion. Intracellular adenosine triphosphate（ATP） depletion and subsequent induced endoplasmic reticulum（ER） stress are proposed to be the underlying mechanisms of ischemic retinal cell death. Recently, we found that a naphthalene derivative can inhibit ATPase activity of valosin-containing protein, universally expressed within various types of cells, including retinal neural cells, with strong cytoprotective activity. Based on the chemical structure, we developed novel valosin-containing protein modulators, Kyoto University Substances（KUSs）, that not only inhibit intracellular ATP depletion, but also ameliorate ER stress. Suppressing ER stress by KUSs is associated with neural cell survival in animal models of several neurodegenerative diseases, such as glaucoma and retinal degeneration. Given that a major pathology of ischemic retinal diseases, other than intracellular ATP depletion, is ER stress-induced cell death, KUSs may provide a novel strategy for cell protection in ischemic conditions. Hence, we investigated the efficacy of KUS121 in a rat model of retinal ischemic injury. Intravitreal injections of KUS121, which is clinically preferable route of drug administration in retinal diseases, significantly suppressed inner retinal thinning and retinal cell death, and maintained visual functions. Valosin-containing protein modulation by KUS is a promising novel therapeutic strategy for ischemic retinal diseases.

Biogas purification via water scrubbing produces effluent containing dissolved CH4, H2S,and CO2, which should be removed to reduce greenhouse gas emissions and increase its potential for water regeneration. In this study, a reactor built with air supplies at the top and bottom was utilized for the treatment of biogas purification effluent through biological oxidation and physical stripping processes. Up to 98% of CH4 was removed through biological treatment at a hydraulic retention time of 2 hr and an upper airflow rate of 2.02 L/day. Additionally, a minimum CH4 concentration of 0.04% with no trace of H2S gas was detected in the off gas. Meanwhile, a white precipitate was captured on the carrier showing the formation of sulfur. According to the developed mathematical model, an upper airflow rate of greater than 2.02 L/day showed a small deterioration in CH4 removal performance after reaching the maximum value, whereas a 50 L/day bottom airflow rate was required to strip the CO2 efficiently and raise the effluent p H from 5.64 to 7.3. Microbiological analysis confirmed the presence of type 1 methanotroph communities dominated by Methylobacter and Methylocaldum. However, bacterial communities promoting sulfide oxidation were dominated by Hyphomicrobium.

A monitoring system based on a CaF2 flow-cell detector was previously developed for measuring the tritium concentration in water. The flow-cell detector was fabricated using a granular CaF2(Eu) solid scintillator. In the present study, the effect of lead shielding on the measurement sensitivity was investigated. First, measurements were carried out both on tritium water and tritium-free water without the use of shielding. Counts were accumulated for either 600 or 10 000 s and the detection limits without shielding were calculated. Second, the effect of shielding was evaluated using background radiation. Lead blocks with a thickness of 5 cm were used as shielding materials. Full shielding around the entire measurement system reduced the background count by 53.8%. Approximating this to a background reduction of 2 times, the detection limit with full shielding was then calculated. It was concluded that 5-cm-thick lead shielding reduced the detection limit for tritium by at least 35%. © 2014, Science Press. All rights reserved.
