towards a standard diet-induced and biopsy-confirmed mouse model of non-alcoholic steatohepatitis: Impact of dietary fat source

BACKGROUND the trans-fat containing AMLN(amylin liver non-alcoholic steatohepatitis,NASH)diet has been extensively validated in C57BL/6J mice with or without the Lep^ob/Lep^ob(ob/ob)mutation in the leptin gene for reliably inducing metabolic and liver histopathological changes recapitulating hallmarks of NASH.Due to a recent ban on trans-fats as food additive,there is a marked need for developing a new diet capable of promoting a compatible level of disease in ob/ob and C57BL/6J mice.AIM to develop a biopsy-confirmed mouse model of NASH based on an obesogenic diet with trans-fat substituted by saturated fat.MEtHODS Male ob/ob mice were fed AMLN diet or a modified AMLN diet with trans-fat(Primex shortening)substituted by equivalent amounts of palm oil[Gubra amylin NASH,(GAN)diet]for 8,12 and 16 wk.C57BL/6J mice were fed the same diets for 28 wk.AMLN and GAN diets had similar caloric content(40%fat kcal),fructose(22%)and cholesterol(2%)level.RESULtS the GAN diet was more obesogenic compared to the AMLN diet and impaired glucose tolerance.Biopsy-confirmed steatosis,lobular inflammation,hepatocyte ballooning,fibrotic liver lesions and hepatic transcriptome changes were similar in ob/ob mice fed the GAN or AMLN diet.C57BL/6J mice developed a mild to moderate fibrotic NASH phenotype when fed the same diets.CONCLUSION Substitution of Primex with palm oil promotes a similar phenotype of biopsyconfirmed NASH in ob/ob and C57BL/6J mice,making GAN diet-induced obese mouse models suitable for characterizing novel NASH treatments.

Understanding the solid target dynamics resulting from the interaction with an ultrashort laser pulse is a challenging fundamental multi-physics problem involving atomic and solid-state physics,plasma physics,and laser physics.Knowledge of the initial interplay of the underlying processes is essential to many applications ranging from lowpower laser regimes like laser-induced ablation to high-power laser regimes like laser-driven ion acceleration.Accessing the properties of the so-called pre-plasma formed as the laser pulse’s rising edge ionizes the target is complicated from the theoretical and experimental point of view,and many aspects of this laser-induced transition from solid to overdense plasma over picosecond timescales are still open questions.On the one hand,laser-driven ion acceleration requires precise control of the pre-plasma because the efficiency of the acceleration process crucially depends on the target properties at the arrival of the relativistic intensity peak of the pulse.On the other hand,efficient laser ablation requires,for example,preventing the so-called“plasma shielding”.By capturing the dynamics of the initial stage of the interaction,we report on a detailed visualization of the pre-plasma formation and evolution.Nanometer-thin diamond-like carbon foils are shown to transition from solid to plasma during the laser rising edge with intensitiest near-infrared probe transmission measurements evidence sub-picosecond dynamics of an expanding plasma with densities above 10^(23)cm^(−3)(about 100 times the critical plasma density).the complementarity of a solid-state interaction model and kinetic plasma description provides deep insight into the interplay of initial ionization,collisions,and expansion.

Gold complexes have emerged as promising anticancer metallodrugs due to their efficient thioredoxin reductase(trxR)inhibition,which disturbs the redox balance of cancer cells.However,in this model,the role of the ligand(s)coordinated to gold is often overlooked.In this work,we present a series of tetrapyridyl Au(Ⅲ)complexes that exhibit thiol-induced release of a Au(Ⅰ)ion and a tetrapyridyl ligand.the formation of a free Au(Ⅰ)center is responsible for the expected trxR inhibition.Additionally,the released ligand,which was visible in cells due to its intense blue fluorescence,showed excellent binding properties to the hERG potassium channel.Moreover,these ligands ended up in the lysosomes,resulting in significant lysosome damage.Altogether,the Au(Ⅲ)complexes presented in this work showed broad-spectrum anticancer properties,both in hypoxic 2D monolayers and 3D tumor spheroids.We suggest that the interaction of the released Au(Ⅰ)center and the tetrapyridyl ligand with two different protein targets may combine into prodrugs that overcome hypoxia-induced drug deactivation.

Charge carrier mobilities of organic semiconductors:ab initio simulations with mode-specific treatment of molecular vibrations

the modeling of charge transport in organic semiconductors usually relies on the treatment of molecular vibrations by assuming a certain limiting case for all vibration modes,such as the dynamic limit in polaron theory or the quasi-static limit in transient localization theory.these opposite limits are each suitable for only a subset of modes.Here,we present a model that combines these different approaches.It is based on a separation of the vibrational spectrum and a quantum-mechanical treatment in which the slow modes generate a disorder landscape,while the fast modes generate polaron band narrowing.We apply the combined method to 20 organic crystals,including prototypical acenes,thiophenes,benzothiophenes,and their derivatives.their mobilities span several orders of magnitude and we find a close agreement to the experimental mobilities.Further analysis reveals clear correlations to simple mobility predictors and a combination of them can be used to identify high-mobility materials.

A recent research article published in Science by Ritter et al.reported that endosomal sorting complexes required for transport(ESCRt)proteins mediate repair of lesions in the cell membrane caused by the pore-forming toxin perforin at sites of cytotoxic t cell engagement.1 Subsequent entry of granzymes and initiation of apoptosis of cancer cells is thus limited,leading to reduced sensitivity against t cell‑mediated cytotoxicity.1 Along these lines,this could constitute a new critical resistance mechanism for t cellbased treatment approaches.

Long-term ozone(O_(3))exposure may lead to non-communicable diseases and increase mortality risk.However,cohort-based studies are relatively rare,and inconsistent exposure metrics impair the credibility of epidemiological evidence synthetization.to provide more accurate meta-estimations,this study updates existing systematic reviews by including recent studies and summarizing the quantitative associations between O_(3) exposure and cause-specific mortality risks,based on unified exposure metrics.Cross-metric conversion factors were estimated linearly by decadal observations during 1990-2019.

Comparison of EUV Photomask Metrology Between CD-AFM and tEM

Accurate metrology of extreme ultraviolet (EUV) photomask is a crucial task. In this paper, two different methods for reference EUV photomask metrology are compared. One is the critical dimension atomic force microscopy (CD-AFM). In the measurements, the contribution of its AFM tip geometry is usually the dominant error source, as measured AFM images are the dilated results of measured structures by the AFM tip geometry. to solve this problem, a bottom-up approach has been applied in calibrating the (effective) AFM tip geometry where the result is traceably calibrated to the lattice constant of silicon crystals. the other is transmission electron microscopy (tEM). For achieving measurement traceability, structure features are measured in pairs in tEM images;thus the distance between the structure pair calibrated by a metrological AFM in prior can be applied to determine the magnification of the tEM image. In this study, selected photomask structures are calibrated by the CD-AFM, and then sample prepared and measured by high-resolution tEM nearly at the same location. the results are then compared. Of six feature groups compared, the results agree well within the measurement uncertainty, indicating excellent performance of the developed methodology. this research supports the development of a photomask standard, which is applied as a “reference ruler” with improved low measurement uncertainty in photomask fabs.

Influenza A virus and Staphylococcus aureus are common causative agents of pneumonia.Co-infections with these two pathogens frequently occur and are characterized,among others,by higher morbidity and mortality due to hyper-inflammation of the lungs.Here,we aimed to profile systemic and local cytokine composition at early acute stages of pneumonia in amurinemodel.Allmice recovered from single influenza A virus and/or staphylococcal infections.In contrast,co-infections led to a severe clinical outcome.While distinct cytokine patterns were detected in lungs of single-pathogen-infected animals,co-infections combined both virus-and bacteria-driven responses.However,analyses of infected human primarymonocytic cells as well as bronchial epithelial cells did not reflectmurine profiles.Based on infectious dose,mainly bacteria-driven responses were noted.the impact of single cells to cytokine composition of the lungs and translation of murine studies to humans remains uncertain and warrants further studies.

Adhesion and cellsurface properties of wild species of spore formers against enteric pathogens

Objective:to investigate the adhesion potential and cell surface properties against enteric pathogens Salmonella typhi.Salmonella para typhi A and Vibrio cholera.Methods:Adhesion potentials of spore and vegetative phase were studied separately for the isolates.Hydrophobic nature was measured on the basis of affinity towards the xylene.Autoaggregalion and coaggregation were studied on the basis of clumping of cells.In vitro adhesion studies were done on mucous which were prepared from infant child faeces.Biofilm production of superior adhesive isolate was confirmed by SEM analysis.Results:Spore and vegetative phases of isolates possessed a different rate of adhesion potentials on intestinal mucous,which indicated that cell surface properties were involved in adhesion process.Spores showed a higher hydrophobicity than their vegetative cells which remained less or non hydrophobic.Vegetative phases showed capabilities for autoaggregation and coaggregation.Spores were found to be more adhesive on intestinal mucous than vegetative phase.Among enteric pathogens Vibrio cholera registered higher adhesion potentials with supporting cell surface properties.Among the five sporeforming isolates,isolate BM-3 possess superior adhesion than enteric pathogens and also exhibited biofilm formation which enhances colonization potential.Conclusions:Spore and vegetative cell phases shows differences in adhesion potentials.Cell surface properties and adhesion studies reveals that isolate BM-3 can be selected as superior isolate which is capable for biofilm production.In short,isolate BM-3 possesses an enhanced adhesion potential than enteric pathogens towards intestinal mucous which is a desirable probiotic character.
