Oligodendrocytes(Ols),glial cells that provide myelin sheaths to axons in the central nervous system(CnS),have a dual role.First,this myelin fatty cover around the axons protects the nerves and is crucial for the saltatory propagation of action potentials.Second and not less important,Ols provide trophic support to neurons and axons.Thus,Ol metabolism is important for Ol well-being and the proper accomplishment of their functions,and Ol metabolism energetics directly affect the energy metabolism of neighboring neurons.

Although many causes of Alzheimer’s disease(Ad)may exist,both the original amyloid cascade and tau hypotheses posit that abnormal misfolding and accumulation of amyloid-β(Aβ)and tau protein is the central event causing the pathology.However,that conclusion could be only partly true,and there is conflicting evidence about the role of both proteins in Ad,being able to precede and influence one another.Some researchers argue that these proteins are mere executors rather than primary causes of pathology.Therefore,there have been continuing refinements of both hypotheses,with alternative explanations proposed.Aβand tau proteins may be independently involved in specific neurotoxic pathways;yet there may be other crucial processes going on in early Ad.Moreover,accumulating evidence suggests that Aβand tau act synergistically,rather than additively in disease onset(Jeremic et al.,2021,2023a).

Unmet needs exist in metabolic dysfunction-associated steatotic liver disease(MASld)risk stratification.Our ability to identify patients with MASld with advanced fibrosis and at higher risk for adverse outcomes is still limited.Incorporating novel biomarkers could represent a meaningful improvement to current risk predictors.With this aim,omics technologies have revolutionized the process of MASld biomarker discovery over the past decades.While the research in this field is thriving,much of the publication has been haphazard,often using single-omics data and specimen sets of convenience,with many identified candidate biomarkers but lacking clinical validation and utility.If we incorporate these biomarkers to direct patients’management,it should be considered that the roadmap for translating a newly discovered omics-based signature to an actual,analytically valid test useful in MASld clinical practice is rigorous and,therefore,not easily accomplished.This article presents an overview of this area’s current state,the conceivable opportunities and challenges of omics-based laboratory diagnostics,and a roadmap for improving MASld biomarker research.

This work reports the growth and characterization of p-AlInn layers doped with Mg by plasma-assisted molecular beam epitaxy(PAMBE).AlInn was grown with an Al molar fraction of 0.80 by metal-modulated epitaxy(MME)with a thickness of 180 nm on Si(111)substrates using Aln as buffer layers.low substrate temperatures were used to enhance the incorporation of indium atoms into the alloy without clustering,as confirmed by X-ray diffraction(XRd).Cathodoluminescence measurements revealed ultraviolet(UV)range emissions.Meanwhile,Hall effect measurements indicated a maximum hole mobility of 146 cm^(2)/(V∙s),corresponding to a free hole concentration of 1.23×10^(19)cm^(−3).The samples were analyzed by X-ray photoelectron spectroscopy(XPS)estimating the alloy composition and extracting the Fermi level by valence band analysis.Mg-doped AlInn layers were studied for use as the electron-blocking layer(EBl)in lEd structures.We varied the Al composition in the EBl from 0.84 to 0.96 molar fraction to assess its theoretical effects on electroluminescence,carrier concentration,and electric field,using SIlVACO Atlas.The results from this study highlight the importance and capability of producing high-quality Mg-doped p-AlInn layers through PAMBE.Our simulations suggest that an Al content of 0.86 is optimal for achieving desired outcomes in electroluminescence,carrier concentration,and electric field.

Mitochondrial dysfunction is a key driver of cardiovascular disease(CVd)in metabolic syndrome and diabetes.This dysfunction promotes the production of reactive oxygen species(ROS),which cause oxidative stress and inflammation.Angiotensin II,the main mediator of the renin-angiotensin-aldosterone system,also contributes to CVd by promoting ROS production.Reduced activity of sirtuins(SIRTs),a family of proteins that regulate cellular metabolism,also worsens oxidative stress.Reduction of energy production by mitochondria is a common feature of all metabolic disorders.High SIRT levels and 5’adenosine monophosphate-activated protein kinase signaling stimulate hypoxia-inducible factor 1 beta,which promotes ketosis.Ketosis,in turn,increases autophagy and mitophagy,processes that clear cells of debris and protect against damage.Sodiumglucose cotransporter-2 inhibitors(SGlT2i),a class of drugs used to treat type 2 diabetes,have a beneficial effect on these mechanisms.Randomized clinical trials have shown that SGlT2i improves cardiac function and reduces the rate of cardiovascular and renal events.SGlT2i also increase mitochondrial efficiency,reduce oxidative stress and inflammation,and strengthen tissues.These findings suggest that SGlT2i hold great potential for the treatment of CVd.Furthermore,they are proposed as anti-aging drugs;however,rigorous research is needed to validate these preliminary findings.

Protein aggregation has been linked with many neurodegenerative diseases,such as Alzheimer’s disease(Ad)or Parkinson’s disease.Ad belongs to a group of heterogeneous and incurable neurodegenerative disorders collectively known as tauopathies.They comprise frontotemporal dementia,Pick’s disease,or corticobasal degeneration,among others.The symptomatology varies with the specific tau protein variant involved and the affected brain region or cell type.However,they share a common neuropathological hallmark-the formation of proteinaceous deposits named neurofibrillary tangles.neurofibrillary tangles,primarily composed of aggregated tau(Zhang et al.,2022),disrupt normal neuronal functions,leading to cell death and cognitive decline.

Memory deficit,which is often associated with aging and many psychiatric,neurological,and neurodegenerative diseases,has been a challenging issue for treatment.Up till now,all potential drug candidates have failed to produce satisfa ctory effects.Therefore,in the search for a solution,we found that a treatment with the gene corresponding to the RGS14414protein in visual area V2,a brain area connected with brain circuits of the ventral stream and the medial temporal lobe,which is crucial for object recognition memory(ORM),can induce enhancement of ORM.In this study,we demonstrated that the same treatment with RGS14414in visual area V2,which is relatively unaffected in neurodegenerative diseases such as Alzheimer s disease,produced longlasting enhancement of ORM in young animals and prevent ORM deficits in rodent models of aging and Alzheimer’s disease.Furthermore,we found that the prevention of memory deficits was mediated through the upregulation of neuronal arbo rization and spine density,as well as an increase in brain-derived neurotrophic factor(BdnF).A knockdown of BdnF gene in RGS14414-treated aging rats and Alzheimer s disease model mice caused complete loss in the upregulation of neuronal structural plasticity and in the prevention of ORM deficits.These findings suggest that BdnF-mediated neuronal structural plasticity in area V2 is crucial in the prevention of memory deficits in RGS14414-treated rodent models of aging and Alzheimer’s disease.Therefore,our findings of RGS14414gene-mediated activation of neuronal circuits in visual area V2 have therapeutic relevance in the treatment of memory deficits.

Antarctic Peninsula is experiencing one of the largest global warming events worldwide.Shallow water bodies generated by the melting of snow in summer are numerous,and they might act as sentinels of climate change due to their rapid response and ability to integrate catchment information.Shifts in climate can influence the structure of microbial communities which dominate these freshwaters ecosystems.Here,we characterize three ponds at Cierva Point(Antarctic Peninsula)by examining their physico-chemical and morphological characteristics and we explored how different factors modify the structure of the microbial community.We studied the abundance and biomass of heterotrophic bacteria,picocyanobacteria and picoeukaryote algae during January and February of two consecutive summers(2017 and 2018).We found that ponds had different limnological characteristics,due to their location,geomorphological features and presence of the surrounding flora and fauna.Physico-chemical parameters as well as microbial community differed between ponds,months and years.In 2017,most ponds were oligo to mesotrophic states.The larger accumulated rainfall(as a result of environmental changes on the Antarctic Peninsula)during 2018,particularly in February,causes nutrient runoff into water bodies.This affects those ponds with the highest seabird circulation,such as gentoo penguin,increasing eutrophication.As a result,picoplanktonic abundances were higher,and the community structure shifts to a largely heterotrophic bacteria dominated one.These results suggest that these communities could act as sentinels to environmental changes,anticipating a future with mostly hypertrophic ponds.
