LHP1 Interacts with ATRX through Plant-Specific Domains at Specific Loci Targeted by PRC2

Heterochromatin Protein 1 （HP1） is a major regulator of chromatin structure and function. In animals, the network of proteins interacting with HP1 is mainly associated with constitutive heterochromatin marked by H3K9me3. HP1 physically interacts with the putative ortholog of the SNF2 chromatin remodeler ATRX, which controls deposition of histone variant H3.3 in mammals. In this study, we show that the Arabidopsis thaliana ortholog of ATRX participates in H3.3 deposition and possesses specific conserved domains in plants. We found that plant Like HP1 （LHP1） protein interacts with ATRX through domains that evolved specifically in land plant ancestors. Loss of ATRX function in Arabidopsis affects the expression of a limited subset of genes controlled by PRC2 （POLYCOMB REPRESSIVE COMPLEX 2）, including the flowering time regulator FLC. The function of ATRX in regulation of flowering time requires novel LHPl-interacting domain and ATPase activity of the ATRX SNF2 helicase domain. Taken together, these results suggest that distinct evolutionary pathways led to the interaction between ATRX and HP1 in mammals and its counterpart LHP1 in plants, resulting in distinct modes of transcriptional regulation.

AIM: To study the significance of cap-fitted colonoscopy in improving cecal intubation time and polyp detection rate. METHODS: This study was a prospective randomized controlled trial conducted from March 2008 to February 2009 in a tertiary referral hospital at Sydney. The primary end point was cecal intubation time and the secondary endpoint was polyp detection rate. Consecutive cases of total colonoscopy over a 1-year period were recruited. Randomization into either standard colonoscopy (SC) or cap-assisted colonoscopy (CAC) was performed after consent was obtained. For cases randomized to CAC, one of the three sizes of cap was used: D-201-15004 (with a diameter of 15.3 mm), D-201-14304 (14.6 mm) and D-201-12704 (13.0 mm). All of these caps were produced by Olympus Medical Systems, japan. Independent predictors for faster cecal time and better polyp detection rate were also determined from this study. RESULTS: There were 200 cases in each group. There was no signif icant difference in terms of demographic characteristics between the two groups. CAC, when compared to the SC group, had no signif icant difference in terms of cecal intubation rate (96.0% vs 97.0%, P = 0.40) and time (9.94 ± 7.05 min vs 10.34 ± 6.82 min, P = 0.21), or polyp detection rate (32.8% vs 31.3%, P = 0.75). On the subgroup analysis, there was no significant difference in terms of cecal intubation time by trainees (88.1% vs 84.8%, P = 0.40), ileal intubation rate (82.5% vs 79.0%, P = 0.38) or total colonoscopy time (23.24 ± 13.95 min vs 22.56 ± 9.94 min, P = 0.88). On multivariate analysis, the independent determinants of faster cecal time were consultant-performed procedures (P < 0.001), male patients (P < 0.001), non-usage of hyoscine (P < 0.001) and better bowel preparation (P = 0.01). The determinants of better polyp detection rate were older age (P < 0.001), no history of previous abdominal surgery (P = 0.04), patients not having esophagogastroduodenoscopy in the same setting (P = 0.003),

Alzheimer’s disease (AD) is a complex neurodegenerative disease, pathologically characterized by the accumulation of β-amyloid peptide (Aβ) and intraneuronal neurofibrillary tangles. Production of the Aβpeptide by the regulated intra-membrane proteolysis of the β-amyloid precursor protein (APP) is a central event in *** decline observed in AD models is dependent on Aβgeneration. One of the downstream effects mediated by soluble Aβoligomersincludes the hyperactivation of calcineurin (CaN), a Ca2+and calmodulin (CaM)-dependent, serinethreonine protein phosphatase. CaN is one of the major phosphatase associated with cognitive and neurodegenerative effects of Aβ. A specific isoform of CaN, CaNAβ, is one of the most up-regulated mRNAs in brains of early stage AD patients. While Aβ’s ability to activate CaN has been well studied, not much is known about how increased levels of CaN in AD brain can contribute towards Aβgeneration. In this study we specifically investigate if increased levels of CaNAβ regulate APP metabolism. Our results demonstrate that increased CaNAβ expressionleads to increased APP levels and increased proteolytic products of APP, including Aβ. The data suggest that upregulation of CaN levels could contribute to neurodegeneration observed in AD by increasing levels ofAβ, potentially forming a pathogenic feed forward loop.

Livedo Reticularis in a Patient with EBV Mononucleosis

A 23-year-old woman with serologically confirmed EBV mononucleosis presented with fever, exudative pharyngitis, periorbital and lid edema, cervical lymphadenopathy, and splenomegaly. Two weeks after the onset of symptoms she developed a generalized symmetrical reticular eruption of unbroken hexagons characteristic of livedo reticularis that resolved over several weeks as her condition improved. To our knowledge, this is the first reported case of livedo reticularis to occur in a patient with EBV mononucleosis.

Earth Observation(EO)has been recognised as a key data source for supporting the United Nations Sustainable Development Goals(SDGs).Advances in data availability and analytical capabilities have provided a wide range of users access to global coverage analysis-ready data(ARD).However,ARD does not provide the information required by national agencies tasked with coordinating the implementation of ***,standardised,scalable mapping of land cover and its change over time and space facilitates informed deci-sion making,providing cohesive methods for target setting and reporting of *** aim of this study was to implement a global framework for classifying land *** Food and Agriculture Organisation’s Land Cover Classification System(FAO LCCS)provides a global land cover taxonomy suitable to comprehensively support SDG target setting and *** present a fully implemented FAO LCCS optimised for EO data;Living Earth,an open-source software package that can be readily applied using existing national EO infrastructure and satellite *** resolve several semantic challenges of LCCS for consistent EO implementation,including modifications to environmental descriptors,inter-dependency within the mod-ular-hierarchical framework,and increased flexibility associated with limited data *** ensure easy adoption of Living Earth for SDG reporting,we identified key environmental descriptors to provide resource allocation recommendations for generating routinely retrieved input *** Earth provides an optimal platform for global adoption of EO4SDGs ensuring a transparent methodology that allows monitoring to be standardised for all countries.

The Future of Artificial Intelligence for Alzheimer’s Disease Diagnostics

Alzheimer’s disease (AD) is a leading cause of death, yet there is no disease-modifying drug therapy currently available. It is critical to establish a diagnosis of AD before clinical system onset so that drug therapies can start earlier. Unfortunately, this is not the current standard practice. Artificial intelligence (AI) holds tremendous promise for identifying AD related structural changes in brain scan images. This paper discusses the recent applications and potential future directions for AI in AD diagnostics. Annual brain scanning and computer vision-assisted early diagnosis is encouraged, so that disease-modifying drug therapy could begin earlier in the progressive pathology.

Digital ischemic necrosis caused by pegylated interferon in a patient with hepatitis C

Pegylated interferon plus ribavirin remains the firstline treatment for patients with hepatitis C virus(HCV) . Interferonαhas the most extensive clinical application and is used for the treatment of chronic hepatitis B virus and hepatitis D virus as well as acute and chronic HCV *** attachment of polyethylene glycol to interferon increases its half-life by reducing the rate of absorption after injection,reducing renal and cellular clearance and also decreasing *** this case report,we have described a patient with chronic hepatitis C who developed ischemic necrosis of her fingertips after completing her third course of pegylated interferon and *** patient underwent a very extensive workup in order to determine the underlying cause of her digital ischemia which was finally determined to be secondary to the use of pegylated interferon.
