Invasive hybridization and introgression pose a serious threat to the persistence of many native species. Understand- ing the effects of hybridization on native populations （e.g., fitness consequences） requires numerous species-diagnostic loci dis- tributed genome-wide. Here we used RAD sequencing to discover thousands of single-nucleotide polymorphisms （SNps） that are diagnostic between rainbow trout （RBT, Oncorhynchus mykiss）, the world＇s most widely introduced fish, and native westslope cutthroat trout （WCT, （9. clarkii lewisi） in the northern Rocky Mountains, USA. We advanced previous work that identified 4,914 species-diagnostic loci by using longer sequence reads （100 bp vs. 60 bp） and a larger set of individuals （n = 84）. We sequenced RAD libraries for individuals from diverse sampling sources, including native populations of WCT and hatchery broodstocks of WCT and RBT. We also took advantage of a newly released reference genome assembly for RBT to align our RAD loci. In total, we discovered 16,788 putatively diagnostic SNps, 10,267 of which we mapped to anchored chromosome locations on the RBT genome. A small portion of previously discovered putative diagnostic loci （325 of 4,914） were no longer diagnostic （i.e., fixed between species） based on our wider survey of non-hybridized RBT and WCT individuals. Our study suggests that RAD loci mapped to a draft genome assembly could provide the marker density required to identify genes and chromosomal regions in- fluencing selection in admixed populations of conservation concern and evolutionary interest [Current Zoology 61 （1）： 146-154, 2015].

经一次冷冻解冻处理的囊胚、桑椹胚和8-细胞胚胎的形态,正常率及存活率较高,分别为85.3％、96.2％和85.2％及89.9％、96.2％和96.3％,此三组间无显著差异,4-细胞和1-细胞期胚胎则较低,正常率和存活率分别为69.2％和51.5％及84.6％和60.8％。重复冷冻解冻胚胎的正常率和存活率分别为75.8％和90.9％。一次冷冻和重复冷冻胚胎(囊胚和桑椹胚)移植至受体后的附植率分别为63.6％和34.8％。本研究结果证明:(1)较晚龄的胚胎(8-细胞、桑椹胚和囊胚)较适宜于作超低温冷冻保存,(2)冷冻保存胚胎解冻后未使用者可作重复冷冻保存,(3)重复冷冻解冻胚胎移植至受体后仍可继续发育。

Leptospira binds to host extracellular matrix(ECM)through surface exposed outer membrane proteins called adhesin in order to initiate *** various adhesins present on the surface of the spirochete, Leptospira-immunoglobulin like proteins(Lig proteins)and LipL32 are most abundant,widely distributed among pathogenic serovars and well *** fragments of Lig proteins(Ligcon4,Ligcon4-7.5,LigBcen2) and C-terminus fragment of LipL32 all of that bind to host ECM were fused,expressed and purified in soluble form as *** week hamsters were immunized subcutaneously with various fusion proteins emulsified in EMULSIGEN-D adjuvant and subsequently boosted at 3 *** protective efficacy of these novel fusion proteins was evaluated against subsequent challenge with highly virulent *** serovar pomona(MLD50-100).Our results indicate that fusion protein based vaccine induced significant protection against acute infection with respect to pBS-adjuvant vaccinated controls as revealed by enhanced survival and reduced pulmonary ***,the protection mediated by these novel proteins was higher than that of conserved region of Lig protein(Ligcon,established protective antigen)and correlated to the level of ***32 failed to impart significant protection,however fusing its immunogenic C-terminus domain to Lig fragments slightly delayed the morbidity of the infected *** results demonstrate that this novel strategy could be promising in developing effective subunit vaccine to combat this zoonotic infection.

The phosphatidylinositol 3 kinase （pI3K）/AKT pathway is genetically targeted in more pathway components and in more tumor types than any other growth factor signaling pathway, and thus is frequently activated as a cancer driver. More importantly, the pI3K/AKT pathway is composed of multiple bifurcating and converging kinase cascades, providing many potential targets for cancer therapy. Renal cell carcinoma （RCC） is a high-risk and high-mortality cancer that is notoriously resistant to traditional chemotherapies or radiotherapies. The pI3K/AKT pathway is modestly mutated but highly activated in RCC, representing a promising drug target. Indeed, pI3K pathway inhibitors of the rapalog family are approved for use in RCC. Recent large-scale integrated analyses of a large number of patients have provided a molecular basis for RCC, reiterating the critical role of the pI3K/AKT pathway in this cancer. In this review, we summarize the genetic alterations of the pI3K/AKT pathway in RCC as indicated in the latest large-scale genome sequencing data, as well as treatments for RCC that target the aberrant activated pI3K/AKT pathway.

Therapeutic horseback riding(THR)as an animal-assisted intervention is one of the innovative approaches emer-ging in the treatment for children with autism spectrum disorder(ASD).The current study was designed to inves-tigate the effects of a 12-week,twice a week THR program on motor skills in sixty-eight children with ASD aged 5–10 years *** participants selected met the DSM-V criteria for ASD,and a total ofﬁfty-three participants completed the study.A randomized controlled trial design was utilized for the *** was collected via a pre-THR test,interim-THR test,and post-THR test to investigate the possible changes in motor skills throughout the 12-week THR *** showed that the THR program signiﬁcantly improved overall motor skills across time points(pp and two-hand catch(as compared to the control group,pprogram may be an effective option for improving motor skills in children with ASD and further investigation with a longer period of intervention is warranted.

polyetheretherketone(pEEK)is an alternative to metallic implants and a material of choice in many applications,including orthopedic,spinal,trauma,and *** titanium(Ti)and Ti-alloys are widely used in many intraosseous implants due to its biocompatibility and ability to osseointegrate,negatives include stiffness which contributes to shear stress,radio-opacity,and *** surgeons prefer to use pEEK due to its biocompatibility,similar elasticity to bone,and radiolucency,however,due to its inert properties,it fails to fully integrate with *** Neutral Atom Beam(ANAB)technology has been successfully employed to demonstrate enhanced bioactivity of pEEK both in vitro and in *** this study,we further characterize surfaces of pEEK modified by ANAB as well as elucidate attachment and genetic effects of dental pulp stem cells(DpSC)exposed to these *** modification resulted in decreased contact angle at 72.9±4.5°as compared to 92.4±8.5°for control(ppSC attachment and proliferation with considerable genetic differences showing earlier progression towards osteogenic *** surface modification is achieved without adding a coating or changing the chemical composition of the pEEK *** together,we show that ANAB processing of pEEK surface enhances the bioactivity of implantable medical devices without an additive or a coating.

本研究目的在于探索牛卵泡卵母细胞的耐冻性和解冻后体外成熟的可能性。利用从屠宰母牛卵巢采集的1407个卵母细胞在四种冷冻液中进行超低温冷冻保存。解冻后,卵母细胞的形态正常率和存活率分别为88.1％和86.5％。四种冷冻液中卵母细胞的存活率无显著差异(p>0.05)。然而,慢解冻的卵母细胞存活率显著高于快解冻者(p<0.01)。经冷冻保存和解冻后尚存活的牛卵泡卵母细胞作体外培养时,有一定比例(15.1％)出现第一极体达到成熟阶段。研究结果证明,牛卵巢卵母细胞可作超低温冷冻长期保存,解冻后可以体外培养成熟。

AIM: To investigate the effects of intraocular lens(IOL) implantation on visual field(VF) in patients with glaucoma and comorbid cataracts(G&C) with different disease ***: Totally 56 eyes of 50 patients with primary G&C were included. All patients were divided into three groups based on the severity of the VF defect: the mild, moderate, and severe stage. phacoemulsification was performed for cataract removal combined with IOL implantation. Visual acuity(VA) and VF tests were performed for all enrolled patients, up to 3 mo after surgery. Changes in VF threshold and global VF index in various groups were also recorded before and after surgery. The mean light sensitivity(MS) values and the changes following surgery(DMS) were compared between the three groups. Advanced Glaucoma Intervention Study(AGIS) scoring was analyzed on all VF results for analysis of changes in VF before and after ***: Following surgery, the MS values of the three groups of G&C increased significantly, while the AGIS scores decreased statistically in all groups. The DMS values for the three zones in moderate and severe stage but not mild stage were statistically different between zones. The DMS value was significantly higher in zone I than those in zone II and III(zone I>zone II>zone III;ppatients(zone I>zone II>zone III;ppatients was significantly higher in zone I than those in zone II and III(zone I>zone II>zone III;ppatients increased significantly after cataract removal and IOL implantation. Variations in the severity and distribution of characteristics of VF defects result in differences in postoperative VF improvements after cataract surgery. The magnitude of increase in VF sensitivity is associated with VF defect characteristic in glaucoma.

Whittingham等(1971,1972)报道小鼠胚胎超低温(-196℃和-269℃)冷冻获得成功,得到科学界的广泛重视,发展甚速,接着相继报道了牛、羊、兔等的胚胎超低温冷冻。 Tsunoda等(1976)报导,取仓鼠未受精卵置于塑料细管中冷冻,并于液氮中保存30分钟5525小时,解冻后仓鼠卵的形态正常率为87—94％。1981年,Tsunoda等采用不同的防冻剂(二甲亚砜、乙二醇和甘油)冷冻8-细胞期仓鼠胚胎,于0℃添加和稀释乙二醇防冻剂者形态正常率最高(89％),于37℃添加防冻剂者均较低(14—45％)。
