Participants in mixed-species bird flocks(MSFs)have been shown to associate with species that are similar in body size,diet,and evolutionary history,suggesting that facilitation structures these assemblages.In addition,several studies have suggested that species in MsFs resemble each other in their plumage,but this question has not been systematically investigated for any MsF system.During the nonbreeding season of 2020 and 2021,we sampled 585 MSFs on 14 transects in 2 habitats of tongbiguang Nature Reserve in western Yunnan Province,China.We performed social network analysis and the Multiple Regression Quadratic Assignment Procedure to evaluate the effect of 4 species traits(body size,overall plumage color,distinctive plumage patterns,and diet)and evolutionary history on species association strength at the whole-MSF and within-MSF levels.All 41 significant relationships showed that species with stronger associations were more similar in their various traits.Body size had the strongest effect on association strength,followed by phylogeny,plumage patterns,and plumage color;diet had the weakest effect.Our results are consistent with the hypotheses that the benefits of associating with phenotypically similar species outweigh the potential costs of interspecific competition,and that trait matching can occur in plumage characteristics,albeit more weakly than in other traits.Several explanations exist as to why similarities in plumage may occur in MSFs,including that they could reduce predators'ability to target phenotypically"odd"individuals.Whether trait matching in plumage occurs through assortative processes in ecological time or is influenced by co-evolution requires furtherstudy.

Due to a lack of resources, rural communities often face challenges when planning catastrophic events. this project involved applying systems thinking and model-based systems engineering to develop a proof-of-concept, multi-method computer simulation and then determining whether the simulation could be used to assess the efficacy of disaster planning approaches on health outcomes in rural communities, as a function of primary healthcare. the project focus was a rural or non-urban healthcare system experiencing a natural hazard. Both system dynamics and discrete event models were incorporated to represent subsystem operations, crucial disaster responses, as well as three key response systems: public health, emergency management, and healthcare. the subsystem models included several components: policies/procedures, communications,resources, exercises/drills/training, healthcare space and staff, and the flow of affected people into and through the system.the combined simulation can serve as a first step to a more comprehensive approach to helping rural communities achieve more efficient and effective healthcare planning for disaster responses.

Introduction:Biological soil crusts(BSCs)can dominate surface cover in dry lands worldwide,playing an integral role in arid land biogeochemistry,particularly in N fertilization through fixation and cycling.Nitrification is a characteristic and universal N transformation in BSCs that becomes important for the export of N beyond the microscopic bounds of the crust itself.the contribution of ammonia-oxidizing bacteria(AOB)in BSCs has been shown,but the role and extent of the recently discovered ammonia-oxidizing archaea(AOA)have not.Methods:We sampled various types of crusts in four desert regions across the western United States and characterized the composition and size of ammonia-oxidizing communities using clone libraries and quantitative PCR targeting the amoA gene,which codes for the ammonia monooxygenase enzyme,universally present in ammonia-oxidizing microbes.Results:All archaeal amoA sequences retrieved from BSCs belonged to the thaumarchaeota(Nitrososphaera associated Group I.1b).Sequences from the Sonoran Desert,Colorado Plateau,and Great Basin were indistinguishable from each other but distinct from those of the Chihuahuan Desert.Based on amoA gene abundances,archaeal and bacterial ammonia oxidizers were ubiquitous in our survey,but the ratios of archaeal to bacterial ammonia oxidizers shifted from bacterially dominated in northern,cooler deserts to archaeally dominated in southern,warmer deserts.Conclusions:Archaea are shown to be potentially important biogeochemical agents of biological soil crust N cycling.Conditions associated with different types of BSCs and biogeographical factors reveal a niche differentiation between AOA and AOB,possibly driven by temperature.

the cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies.Over the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats.Yet,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging.In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database Fungaltraits operating at genus and species hypothesis levels.Combining the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,respectively.this resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental stud-ies.In order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ItS sequences.On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.

β-cells are a type of endocrine cell found in pancreatic islets that synthesize,store and release insulin.In type 1 diabetes(t1D),t-cells of the immune system selectively destroy the insulin-producingβ-cells.Destruction of these cells leads to a lifelong dependence on exogenous insulin administration for survival.Consequently,there is an urgent need to identify novel therapies that stimulateβ-cell growth and induceβ-cell function.We and others have shown that pancreatic ductal progenitor cells are a promising source for regeneratingβ-cells for t1D owing to their inherent differentiation capacity.Default transcriptional suppression is refractory to exocrine reaction and tightly controls the regenerative potential by the EZH2 methyltransferase.In the present study,we show that transient stimulation of exocrine cells,derived from juvenile and adult t1D donors to the FDAapproved EZH2 inhibitors GSK126 and tazemetostat(taz)influence a phenotypic shift towards aβ-like cell identity.the transition from repressed to permissive chromatin states are dependent on bivalent H3K27me3 and H3K4me3 chromatin modification.targeting EZH2 is fundamental toβ-cell regenerative potential.Reprogrammed pancreatic ductal cells exhibit insulin production and secretion in response to a physiological glucose challenge ex vivo.these pre-clinical studies underscore the potential of small molecule inhibitors as novel modulators of ductal progenitor differentiation and a promising new approach for the restoration ofβ-like cell function.

this review highlights some established and some more contemporary mechanisms responsible for heart failure(HF)-induced skeletal muscle wasting and weakness.We first describe the effects of HF on the relationship between protein synthesis and degradation rates,which determine muscle mass,the involvement of the satellite cells for continual muscle regeneration,and changes in myofiber calcium homeostasis linked to contractile dysfunction.We then highlight key mechanistic effects of both aerobic and resistance exercise training on skeletal muscle in HF and outline its application as a beneficial treatment.Overall,HF causes multiple impairments related to autophagy,anabolic-catabolic signaling,satellite cell proliferation,and calcium homeostasis,which together promote fiber atrophy,contractile dysfunction,and impaired regeneration.Although both wasting and weakness are partly rescued by aerobic and resistance exercise training in HF,the effects of satellite cell dynamics remain poorly explored.

Knee biomechanics of patients with total knee replacement during downhill walking on different slopes

Purpose:the purpose of this study was to compare knee biomechanics of the replaced limb to the non-replaced limb of total knee replacement(tKR)patients and healthy controls during walking on level ground and on decline surfaces of 5°,10°,and 15°.Methods:twenty-five tKR patients and 10 healthy controls performed 5 walking trials on different decline slopes on a force platform and an instrumented ramp system.two analyses of variance,2×2(limb×group)and 2×4(limb×decline slope),were used to examine selected biomechanics variables.Results:the replaced limb of tKR patients had lower peak loading-response and push-off knee extension moment than the non-replaced and the matched limb of healthy controls.No differences were found in loading-response and push-off knee internal abduction moments among replaced,non-replaced,and matched limb of healthy controls.the knee flexion range of motion,peak loading-response vertical ground reaction force,and peak knee extension moment increased across all slope comparisons between 0°and 15°in both the replaced and non-replaced limb of tKR patients.Conclusion:Downhill walking may not be appropriate to include in early stage rehabilitation exercise protocols for tKR patients.

Metabolic flexibility has emerged as a critical determinant of CD8+t-cell antitumor activity,yet the mechanisms driving the metabolic flexibility of t cells have not been determined.In this study,we investigated the influence of the nuclear cap-binding complex(CBC)adaptor protein ARS2 on mature t cells.In doing so,we discovered a novel signaling axis that endows activated CD8+t cells with flexibility of glucose catabolism.ARS2 upregulation driven by CD28 signaling reinforced splicing factor recruitment to pre-mRNAs and affected approximately one-third of t-cell activation-induced alternative splicing events.Among these effects,the CD28-ARS2 axis suppressed the expression of the M1 isoform of pyruvate kinase in favor of PKM2,a key determinant of CD8+t-cell glucose utilization,interferon gamma production,and antitumor effector function.Importantly,PKM alternative splicing occurred independently of CD28-driven PI3K pathway activation,revealing a novel means by which costimulation reprograms glucose metabolism in CD8+t cells.
