Rationale:Comorbidity of depression and anxiety is one of the most common mental disorders in *** experimental procedures,however,have focused on identifying mixed depression-and anxiety-like behaviors in animal ***:To develop a novel testing paradigm,the so-called Liang’s contextual-stress Box(briefly,Liang’s Box),for evaluating depression-and/or anxiety-like behaviors in ***:Liang’s Box consisted of a central area(CA)with three peripheral arms(PA).mice were placed individually in the end of a peripheral arm facing to the central area and were allowed to travel around Liang’s *** a test period,behavioral parameters were recorded and collected by a video-tracking system named SmART 3.0(Panlab,Spain),including①Latency to CA,②Time spent in CA(CA-time),③Distance traveled in CA(CA-distance),④Distance traveled in PA(PA-distance),⑤Transition number between CA and PA,⑥Global activity,⑦CA-time/cm2,⑧PA-time/cm2,⑨CAdistance/cm2,and⑩PA-distance/*** order to assess the reliability and validity of Liang’s Box and get baseline information,we undertook a series of experiment,including verifying consistency and stability in the various strains of mice(Km,ICR,and C57 BL/6 J mice),imposing high illumination,restraint and tail suspension stress,pre-treating with an anxiolytic(diazepam,buspirone)or antidepressant(desipramine,mianserin).Results:①There were not remarkable differences in all the parameters among Km,ICR,and C57 BL/6 J mice;②High illumination,restraint and tail suspension significantly increased the latency to CA and decreased the CA-time,CA-distance and transition ***,these three types of stresses evoked different effect on global *** illumination increased activity,but restraint stress had no significant *** contrast with high illumination,tail suspension stress markedly reduced activity in mice;③The anxiolytics diazepam(0.25 mg·kg-1 and 0.5 mg·kg-1,i.p.)and buspirone(0.5 mg·kg-1 and 1.0 mg·kg-1,i.p.)

At the China Spallation Neutron Source(CSNS), we have developed a custom gas-filling station, a glassblowing workshop, and a spin-exchange optical pumping(SEOP) system for producing high-quality ^(3)He-based neutron spin filter(NSF) cells. The gas-filling station is capable of routinely filling ^(3)He cells made from GE180 glass of various dimensions, to be used as neutron polarizers and analyzers on beamlines at the CSNS. Performance tests on cells fabricated at our gas-filling station are conducted via neutron transmission and nuclear-magneticresonance measurements, revealing nominal filling pressures, and a saturated ~3He polarization in the region of 80%, with a lifetime of approximately 240 hours. These results demonstrate our ability to produce competitive NSF cells to meet the ever-increasing research needs of the polarized neutron research community.

Based on the optimal estimation method, a satellite XCO2 retrieval algorithm was constructed by combining LBLRTm with VLIDORT. One-year GOSAT/TANSO observations over four TCCON stations were tested by our algorithm, and retrieval results were compared with GOSAT L2 B products and ground-based FTS measurements. meanwhile, the influence of CO2 line mixing effect on retrieval was estimated, and the research showed that neglecting CO2 line mixing effect could result in approximately 0.25% XCO2 underestimation. The accuracy of XCO2 retrievals was similar to GOSAT L2 B products at cloud-free footprints with aerosol optical depth less than 0.3, and 1% accuracy of XCO2 retrievals can be reached based on the validation result with TCCON measurements.

mounting evidence indicates that the orbital period of the solar system’s movement through the milky Way has had a controlling effect on processes of the Earth’s system throughout the *** decipher the response of a given terrestrial basin’s rhythms to galactic dynamics,for the first time,we report long-term cycles recorded in the mesozoic Ordos Basin(Central China).The astronomical time scale(ATS)was established for each successive sedimentary sequence,and the duration of unconformity episodes was estimated based on the chronological *** this timescale,time-series analysis of the deposition rate is carried out through the mesozoic Ordos Basin,which reveals compelling periodicities of 93,33,9,3-5,and *** radial solar system motions around the galactic centre and plate tectonic cycles act cooperatively to impact the magmatic tempo of the Qinling orogenic belt and the 93-myr depositional cycle of the mesozoic Ordos *** mesozoic 33-myr cycle is a sedimentary response to the half-period of the solar system’s vertical oscillation about the galactic plane.A rational explanation is that galactic oscillation affects mantle convection,which is responsible for periodic asthenospheric upwelling and ultimately controls the vertical crust oscillation of the Ordos *** 9-myr and the higher-frequency 3-5and 2.4-myr depositional cycles can be sedimentary responses to the Earth’s orbital eccentricity,which affected the temporal variation in depositional environments in the Ordos *** from the galactic-geologic correlations,long-term cycles recorded within a sedimentary basin should also consider the tectonically driven mechanisms at these *** framework provides a new perspective for revealing the astronomical origin of Earth’s rhythms.

Dolomite genesis is a century-old mystery in *** reveal the mechanism of dolomite genesis,two core problems need to be *** first is the origin and migration mechanism of mg^(2+)-rich fluids during the dolomitization *** second is the kinetic barrier caused by mg^(2+)hydration during dolomite precipitation at low *** address these problems,our study,based on detailed petrological,sedimentological,geochemical(major and trace elements),and isotopic(C-O-mg)analysis,clarified the source and migration of mg^(2+)-rich fluids and the kinetic barrier mechanism of lowtemperature dolomite precipitation in the Upper Sinian Qigebulake Formation and the Lower Cambrian Xiaoerbulake Formation in the Tarim ***,we found that the mg^(2+)-rich fluids required for the dolomitization of dolomite in the Xiaoerbulake Formation were primarily derived from the Early Cambrian marine *** the interface of the sedimentary cycle,δ26mg values fluctuated considerably,indicating that the sequence interface was the starting point and channel for the migration of dolomitized *** level variation plays a major role in controlling the dolomitization process of the Xiaoerbulake ***,the Qigebulake Formation contains low-temperature dolomite with mg^(2+)-rich fluids supplied by seawater,microorganisms,and sedimentary organic *** analysis shows that the dolomite of the Qigebulake Formation was formed by microbial induction by anaerobic methane ***,the properties and sources of dolomitization fluids and the formation process of dolomite were the reasons for the difference in the mg isotope composition of dolomite during the Sinian-Cambrian *** study reveals the genetic mechanism of the Sinian-Cambrian dolomite in the Tarim Basin and establishes a new method to explain the genesis of microbial dolomite by C-O-mg isotopes,providing a reference for the reconstruction of the formation and evolutio

近年来,动物实验证实,糖摄取过多可造成组织损伤,这种组织损伤是由相关的氧化和羰基应激的细胞毒性和炎症引起.我们也已证实如果肝细胞暴露于由枯否细胞或浸润淋巴细胞所产生的无毒剂量的过氧化氢(H202),果糖对干细胞的毒性将会增大100倍.为明确果糖毒性所涉及的机制,本研究首先采用2,4-二硝基苯肼法测定以果糖及其代谢产物蛋白质羰基化作用,其次以用不同浓度的果糖代谢产物(甘油醛)诱导肝细胞毒性,探讨其细胞毒性机制.结果发现:果糖本身并不导致牛血清白蛋白(BSA)的酰基化,但在Fenton体系(FeⅡ/H202或CuⅠ/H202)作用下可酰基化蛋白,FeⅡ/H202的作用比CuⅠ/H202强.果糖代谢产物(甘油醛或乙醇醛)在Fenton反应体系下对蛋白质的羰基化作用显着增加.乙二醛捕获剂氨基胍可以完全阻止果糖、甘油醛和乙醇醛蛋白质羰基化.羟基自由基清除剂可以抑制FeⅡ催化的Fenton反应导致的蛋白质羰基化,但羟基自由基清除剂没有抑制CuⅡ/H202所导致的蛋白质羰基化.在肝细胞炎症模型中,果糖本身的毒性为1.5m,而在无毒剂量的H202存在时,其肝细胞毒性为10 mm,毒性在FeⅢ:HQ(8-羟基喹啉)的存在下进一步增加.甘油醛在H202存在时肝细胞毒性增加,在FeⅢ:HQ/H202存在时毒性进一步增加.乙醇醛的肝细胞毒性在CuⅡ:HQ/H202的诱导下也增加,但羟自由基清除剂只抑制FeⅢ:HQ/H202诱导的细胞毒性而不抑制CuⅡ:HG/H202诱导的细胞毒性.氨基胍可以抑制FeⅢ:HQ/H202和CuⅡ:HQ/H202诱导的细胞毒性及蛋白质羰基化.结论:果糖或果糖代谢物在Fenton体系反应下肝细胞毒性增加可能是NASH的第二次打击,肝细胞所受到的氧化应激环境可以继续增加肝细胞毒性和诱发NASH.

AIm: To identify and characterize the function of non-mu-scle myosin Ⅱ (Nmm Ⅱ) isoforms in primary rat hepatic stellate cells (HSCs).mETHODS: Primary HSCs were isolated from male Spra-gue-Dawley rats by pronase/collagenase digestion. Total RNA and protein were harvested from quiescent and culture-activated HSCs. Nmm Ⅱ isoform (Ⅱ-A, Ⅱ-B and Ⅱ-C) gene and protein expression were measured by RealTime polymerase chain reaction and Western blot analyses respectively. Nmm Ⅱ protein localization was visualized in vitro using immunocytochemical analysis. For in vivo assessment, liver tissue was harvested from bile duct-ligated (BDL) rats and Nmm Ⅱisoform expression determined by immunohistochemistry. Using a selective myosin Ⅱ inhibitor and siRNA-mediated knockdown of each isoform, Nmm Ⅱ functionality inprimary rat HSCs was determined by contraction and migration ***: Nmm Ⅱ-A and Ⅱ-B mRNA expression was increased in culture-activated HSCs (Day 14) with sig-niflicant increases seen in all pairwise comparisons (Ⅱ-A: 12.67 ± 0.99 (quiescent) vs 17.36 ± 0.78 (Day 14), P milar expression patterns (Ⅱ-A: 1.87 ± 2.50 (quiescent) vs 58.64 ± 8.76 (Day 14), P mm Ⅱ-C mRNA and protein expression between quiescent and activated HSCs. In culture-activated HSCs, Nmm Ⅱ-A and Ⅱ-B merged with F-actin at the cellular periphery and throughout cytoplasm respectively. In vitro stud-ies showed increased expression of Nmm Ⅱ-B in HSCs activated by BDL compared to sham-operated animals. There were no apparent increases of Nmm Ⅱ-A and Ⅱ-C protein expression in HSCs during hepatic BDL injury.
