Rbfox1 controls alternative splicing of focal adhesion genes in cardiac muscle cells

Alternative splicing is one of the major cellular processes that determine the tissue-specific expression of protein ***,it remains challenging to identify physiologically relevant and tissue-selective proteins that are generated by alternative ***,we investigated the target spectrum of the splicing factor Rbfox1 in the cardiac muscle context in more *** using a combination of in silico target prediction and in-cell validation,we identified several focal adhesion proteins as alternative splicing targets of *** focused on the alternative splicing patterns of vinculin(metavinculin isoform)and paxillin(extended paxillin isoform)and identified both as potential Rbfox1 *** analyses suggested that both isoforms are promoted by Rbfox1 due to binding in the *** adhesions play an important role in the cardiac muscle context,since they mainly influence cell shape,cytoskeletal organization,and cell–matrix *** data confirmed that depletion of Rbfox1 changed cardiomyoblast morphology,cytoskeletal organization,and multinuclearity after differentiation,which might be due to changes in alternative splicing of focal adhesion ***,our results indicate that Rbfox1 promotes alternative splicing of focal adhesion genes in cardiac muscle cells,which might contribute to heart disease progression,where downregulation of Rbfox1 is frequently observed.

Direct Pointwise Comparison of FE Predictions to StereoDIC Measurements:Developments and Validation Using Double Edge-Notched Tensile Specimen

To compare finite element analysis(FEA)predictions and stereovision digital image correlation(StereoDIC)strain measurements at the same spatial positions throughout a region of interest,a field comparison procedure is *** procedure includes(a)conversion of the finite element data into a triangular mesh,(b)selection of a common coordinate system,(c)determination of the rigid body transformation to place both measurements and FEA data in the same system and(d)interpolation of the FEA nodal information to the same spatial locations as the StereoDIC measurements using barycentric *** an aluminum Al-6061 double edge notched tensile specimen,FEA results are obtained using both the von Mises isotropic yield criterion and Hill’s quadratic anisotropic yield criterion,with the unknown Hill model parameters determined using full-field specimen strain measurements for the nominally plane stress *** Hill’s quadratic anisotropic yield criterion,the point-by-point comparison of experimentally based full-field strains and stresses to finite element predictions are shown to be in excellent agreement,confirming the effectiveness of the field comparison process.

Objective of this work was to develop a novel method for characterizing real 3D shapes of particles smaller than 20μm by X-ray microtomography(X-RMT).Multidimensional separation of heterogenous solids through agglomeration in suspension will improve recycling processes as the particle shape and the agglomerate size are used for shape-selective *** the present paper we discuss the fundamentals of X-ray tomography and the experimental setup for selective spherical agglomeration in suspension.A specific preparation method of the particulate sample for X-RMT followed by 3D image processing,are essential for the shape analysis expressed as *** also discuss the limitation of this method due to the so-called Partial Volume Effect and particle clusters in the order of magnitude of X-RMT *** proof of concept,we used a mixture of graphite platelets and spheronized graphite particles for a shape selective-agglomeration in *** remaining fines were analyzed and showed more platelets than in the *** indicates that spheronized particles are preferably bound in the *** findings show that,based on the discussed sample preparation and a 3D image analysis in connection with X-RMT,particle shapes of micronized particles can be discriminated.

Launching jets from accretion belts

We propose that sub-Keplerian accretion belts around stars might launch jets. The sub-Keplerian inflow does not form a rotationally supported accretion disk, but it rather reaches the accreting object from a wide solid angle. The basic ingredients of the flow are a turbulent region where the accretion belt interacts with the accreting object via a shear layer, and two avoidance regions on the poles where the accretion rate is very low. A dynamo that is developed in the shear layer amplifies magnetic fields to high values. It is likely that the amplified magnetic fields form polar outflows from the avoidance regions. Our speculative belt-launched jets model has implications on a rich variety of astrophysical objects, from the removal of common envelopes to the explosion of core collapse supernovae by jittering jets.

Binary interactions with high accretion rates onto main sequence stars

Energetic outflows from main sequence stars accreting mass at very high rates might account for the powering of some eruptive objects, such as merging main sequence stars, major eruptions of luminous blue variables, e.g., the Great Eruption of Eta Carinae, and other intermediate luminosity optical transients（ILOTs; red novae; red transients）. These powerful outflows could potentially also supply the extra energy required in the common envelope process and in the grazing envelope evolution of binary systems. We propose that a massive outflow/jets mediated by magnetic fields might remove energy and angular momentum from the accretion disk to allow such high accretion rate flows. By examining the possible activity of the magnetic fields of accretion disks, we conclude that indeed main sequence stars might accrete mass at very high rates, up to≈10^-2M⊙yr^-1for solar type stars, and up toion≈1 M⊙yr^-1for very massive stars. We speculate that magnetic fields amplified in such extreme condits might lead to the formation of massive bipolar outflows that can remove most of the disk＇s energy and angular momentum. It is this energy and angular momentum removal that allows the very high mass accretion rate onto main sequence stars.

Priority setting in health care: Attitudes of physicians and patients

Background: The opinion of physicians clearly counts in prioritizing health care, but there is little information on the rationales underlying treatment decisions and whether these rationales are accepted by patients. Objective: To compare physicians and patients regarding their understanding and use of therapeutic benefit and treatment costs as criteria for prioritizing health care. Methods: Seven physicians and twelve patients were purposefully selected to yield a heterogeneous sample. Participants were interviewed face-to-face, following a semi-structured topic guide comprising three scenarios that focused on interventions with low or unproven therapeutic benefit and high costs, respectively. For data analysis we used qualitative content analysis. Results: We found that patients and physicians differed in their understanding of therapeutic benefit, their expectations of what medicine can do and their use of costs as criteria for prioritizing health care. Physicians were less likely to assess a certain intervention as effec tive, and they less often accepted upper funding limits in health care. Unlike the physicians, patients raised non-medical aspects in decision making such as the patient’s consent and social inequalities. Conclusions: The revealed differences point toward the necessity to strengthen the doctor-patient communication, to improve information for patients about the possibilities and limits of health care and to gain a deeper understanding of their attitudes, wishes and concerns to reach an agreement by physicians and patients on the treatment to be implemented.

A process similar to delta-sigma modulation can be used to shape the noise caused by element mismatch in a multi-element DAC. This paper presents a design of the "vector quantizer," a key component in the element selection logic which accomplishes this noise-shaping.A 4-order△Σmodulator is implemented on a Xilinx FPGA along with a 2-order element selection logic which makes use of the vector *** FPGA is used to drive an 8- element switched-capacitor(SC) DAC and a 30 dB reduction in the harmonic distortion is observed when mismatch-shaping is employed.

AN INTRODUCTION TO THE NASA EAST PACIFIC ORIGINS AND CHARACTERISTICS OF HURRICANES(EPOCH) FIELD CAMPAIGN

Over the past fi ve years, tropical activity in the East Pacifi c has increased, while declining in the Atlantic Basin. In addition, during El Ni?o years, warmer than average sea surface temperatures further increase the likelihood of tropical cyclone formation in the East Pacifi c. Hurricane fi eld campaigns used the Ku-/Ka-band HighAltitude Wind and Rain Airborne Profi ler(HIWRAP) radar on the Global Hawk(GH) unmanned aircraft, in GRIP(Genesis and Rapid Intensifi cation Processes 2010), HS3(Hurricane and Severe Storm Sentinel 2012-14), and the NOAA Sensing Hazards with Operational Unmanned Technology(SHOUT 2015-16) fi eld campaigns. Although originally designed for the GH, the X-band high-altitude RADar(EXRAD) has yet to be integrated and fl own on an unmanned aerial vehicle. EXRAD will provide data with less attenuation of signal over deep convection as well as better estimates of three-dimensional winds with its nadir-pointing beam. As part of the NASA Hand On Project Experience(HOPE) Training Opportunity, our team proposed to fl y the AV-6 GH aircraft with the EXRAD radar, the High Altitude MMIC Sounding Radiometer(HAMSR), and NOAA Advanced Vertical Atmospheric Profi ling System(AVAPS) dropsondes to investigate genesis and/or rapid intensifi cation(RI) of an East Pacifi c hurricane by measuring both the environment and interior structures. Information on planned activities primarily focused on the EXRAD high-altitude radar integration for the July-August 2017 science fl ight will be presented.
