Identification and Characterization of hmr19 Gene Encoding a Multidrug Resistance Efflux Protein from Streptomyces hygroscopicus *** Strain 10-2.

The hmr19 gene was cloned from Streptomyces hygroscopicus subsp. yingchengensis strain 10- 2., a bacterium strain producing agricultural antibiotics. Sequence similarity comparison indicates that hmr19 gene may encode a predicted protein with 14 putative transmembrane α-helical spanners, belonging to the drug:H+ antiporter-2.family of the major facilitator superfamily. The expression of hmr19 in the mycelium of strain 10-2. was detected by Western blotting analysis. Gene replacement technology was employed to construct an hmr19 disruption mutant. The growth inhibition test against different antibiotics indicated that the mutant strain was 5–2. fold more susceptible to tetracycline, vancomycin and mitomycin C than the parental wild type strain. The mutant took up tetracycline much faster and accumulated more antibiotics than the wild type strain 10-2.. While with the addition of an energy uncoupler, carbonyl cyanide m- chlorophenylhydrazone, the characteristics of the accumulation of [3H]tetracycline in these two strains were almost the same. It was thus concluded that hmr19 encoded a multidrug resistance efflux protein.

Sorption of a triazol derivative by soils: importance of surface acidity

The sorption of a triazol derivative, 1-(4-chlorophenyl)- 4,4-dimethyl-2.(1H-1,2.4-triazol-1-yl)penten-3-ol with a common name of S3307D, on fifteen soils and three H_2._2.treated soils was investigated. The sorption isotherm for each untreated and treated soil was non-linear, and was best fitted to Freundlich sorption equation. Soils containing high amount of clay content or organic matter or both sorbed much higher amounts of the chemical than soils that had low contents of these soil constituents. H_2._2.treated soils showed considerable sorptive affinity for S3307D. It was concluded that both organic matter and mineral fraction in natural soils contributed to the sorption of the basic compound. Sorption by the H_2._2.treated soils increased as suspension pH decreased, but all suspension pHs exceeded the pKa of the compound by more than two units. This implies that organic base protonation can occur on surfaces of soil components, and surface acidity (exchangeable acidity ) is important in sorption process of the organic base rather than suspension pH.

A Resistivity Gradient Piezoelectric FGM Actuator

A resistivity gradient actuator based on lead zirconate titanate ceramics was successfully developed and the bending deflections up to 140 μm were obtained. The actuator material was a matrix of PZT ceramic into which smooth gradient of piezoelectric activity was introduced. The application of an electric field then causes the actuator to bend due to differential strains induced by the piezoelectric effect. The resistivity gradient of the actuator was achieved by doping PZT with suitable donor and acceptor dopants. PZT powder was modified and synthesized by using two stage powder fabrication method. The actuator was fabricated by uniaxial pressing followedby isostatic pressing with two layers of different resistivities.

Self-assembly of peptides into fibrils and other morphologies has attracted much attention inmany fields,especially in nanofabrication,pathology and *** this paper,self-assembly of GAV-9peptide in organic solvents,ethanol and acetone,was investigated using atomic force microscopy(AFM)and attenuated total reflectance-Fourier transform infrared spectroscopy(ATR-FTIR).The results indicatedthat GAV-9 self-assembled into various nanostructures in both solvents after deposited and evaporated *** with β-sheet conformation were observed in both solvents when the peptide concentrationwas higher than 2.0 μ***,ordered fibrils with β-sheet conformation were formed in ethanol,butnot in acetone,with a peptide concentration ranging from 7 μM to 2. μ*** attribute the formation ofvarious nanostructures to the different physicochemical properties of the polar organic solvents on the self-assembly of GAV-9 peptide.

The low-abundantly expressed genes composed the majorities of the mRNAs expressed in the central nervous system (CNS), and were thought to be important for the normal brain functions. Through differential screening a low-abundance cDNA sublibrary with mRNA from neuropathic pain of chronic constriction injury (CCI) model, we have identified a novel rat gene, rat spinal-cord expression protein 4 gene (RSEP4). The total length of RSEP4 cDNA is 2.06 bp, with a 501 nucleotide open reading frame (ORF) that encodes a 167 amino acid polypeptide. Northern blot revealed that RSEP4 was expressed specifically in the CNS. In situ hybridization showed that the mRNA of RSEP4 was strongly expressed in the CA1, CA2. CA3 and DG regions of hippocampus, the Purkinje cells of cerebellum, and the small sensory neurons of dorsal horn and large motor neurons of ventral horn of spinal cord. Over-expression of RSEP4-EGFP fusion protein in the human embryonic kidney 2.3T cells showed that RSEP4 protein was mainly localized in the cell cytoplasm. These results suggest that RSEP4 may play some roles in the CNS.

Physical Simulation of Nonmetallic Particle Movement in Al Melt under Electromagnetic Field

Physical simulation is used to study the movement of nonmetallic particles in Al melt in electro- magnetic field. It is found that the terminal velocity of particles in different Reynolds number range has different functions. By confirming drag force coefficient of nonmetallic particles with Reynolds number in the range of 0.2.10 and 10-2. respectively, two functions of terminal ve- locity for spherical nonmetallic particles have been got accordingly, which provide a theoretical basis for separating nonmetallic inclusions from Al melt in electromagnetic field.

OPTIMAL CONTROL APPLIED IN AUTOMATIC CLUTCH ENGAGEMENTS OF VEHICLES

Start-up working condition is the key to the research of optimal engagementof automatic clutch for AMT. In order to guarantee an ideal dynamic performance of the clutchengagement, an optimal controller is designed by considering throttle angle, engine speed, gearratio, vehicle acceleration and road condition. The minimum value principle is also introduced toachieve an optimal dynamic performance of the nonlinear system compromised in friction plate wearand vehicle drive quality. The optimal trajectory of the clutch engagement can be described in theform of explicit and analytical expressions and characterized by the deterministic and accuratecontrol strategy in stead of indeterministic and soft control techniques which need thousands ofexperiments. For validation of the controller, test work is carried out for the automated clutchengagements in a commercial car with an traditional mechanical transmission, a hydraulic actuator, agroup of sensors and a portable computer system. It is shown through experiments that dynamicbehaviors of the clutch engagement operated by the optimal control are more effective and efficientthan those by fuzzy control.

A three-dimensional representation of vasculature can be extremely important in image-guided neurosurgery, pre-surgical planning. In this paper, a spatial continuity incorporated multi-attribute fuzzy clustering algorithm (MAFCM S) is proposed to segment entire blood vessels from TOF MRA images. This clustering method takes both the intensity information and the geometrical information into account, while most of the cur- rent clustering methods only deal with the former. In this method, a new dissimilarity measure, which integrates the intensity and the geometry shape dissimilarity, is introduced. Because of the presence of the geometrical information, the new measure is able to differentiate the pixels with similar intensity values within different geometrical shape structures. Experimental results show that the new algorithm can get better segmentation.

MOLECULAR DYNAMICS SIMULATION OF INTERFACIAL DEFECTS WITH MODIFIED POTENTIAL BASED ON THE FIRST-PRINCIPLE

The interfacial molecular structure of materials with different lattice constants is simulated by using the method of molecular dynamics （MD）. Potential of modified analytical embedded atom method （MAEAM） is used for nominterfacial atoms. To simulate the state of interracial atoms, further modifications are applied to the MAEAM potential based on the first- principle simulation results. It is concluded that a small change of potential may greatly influence the interfacial molecular structure. Void and diffusions can be observed in the simulation results. It is also found that the pre-existed dot defects before bonding can decrease the number of interfacial defects greatly, thereby, can increase the strength of the interface.
