Cisplatin-induced activation of TGF-βsignaling contributes to drug resistance

Growing evidence suggests an association between epithelial-mesenchymal transition(EMT),a hallmark of tumor malignancy,and chemoresistance to a number of anti-cancer ***,the mechanism of EMT induction in the process of acquiring anti-cancer drug resistance remains *** address this issue,we obtained a number of cisplatin-resistant clones from LoVo cells and found that almost all of them lost cell-cell *** these clones,the epithelial marker E-cadherin was downregulated,whereas the mesenchymal marker N-cadherin was ***,the expression of EMT-related transcription factors,including Slug,was *** the other hand,the upregulation of other mesenchymal marker Vimentin was weak,suggesting that the mesenchymal-like phenotypic changes occurred in these cisplatin-resistant *** mesenchymal-like features of cisplatin-resistant clones were partially reversed to parental epithelial-like features by treatment with transforming growth factor-β(TGF-β)receptor kinase inhibitors,indicating that TGF-βsignaling is involved in cisplatin-induced the mesenchymallike phenotypic ***,cisplatin was observed to enhance the secretion of TGF-βinto the culture media without influencing TGF-βgene *** results suggest that cisplatin may induce the mesenchymal-like phenotypic changes by enhancing TGF-βsecretion,ultimately resulting in drug resistance.

Two new spectroscopic methods to detect the optical activity of liquid-liquid interfaces have been developed. The first one is the centrifugal liquid membrane （CLM） method combined with a conventional circular dichroism （CD） spectropolarimetry and the second one is a more interfacial specific second harmonic generation CD （SHG-CD） spectrometry. In the CLM-CD method, a cylindrical glass cell containing small amounts of organic and aqueous phases was rotated at about 7000 r/min in a sample chamber of a CD spectropolarimeter to generate an interface with a high specific interfacial area between the two-phase liquid membranes. The CD spectra of the J-aggregate of protonated 5,10,15, 20-tetraphenylporphyrin formed at the toluene-sulfuric acid interface have been measured, As for the SHG-CD, a circularly polarized wavelength-variable fs-laser system was constructed to measure the interfacial SHG spectra of a flat liquid-liquid interface. The ion-associated aggregation of a water-soluble anionic porphyrin promoted with a cationic amphiphile at the heptane-water interface was observed by this technique and the observed SHG-CD spectra proved the generation of a characteristic oPtical activity accompanied by the formation of the interfacial aggregate of inherently achiral porphyrin molecules. These methods will pioneer a new field of interfacial chiral chemistry in the studies of solvent extraction mechanisms.

Luminescence study of europium(III) tris(β-diketonato)/phosphonate complexes in chloroform

The extraction of Eu（Ⅲ） with β-diketone, HA, and monodentate or bidentate Lewis bases, B, into chloroform and the luminescence properties of the extracted species were studied. Pivaloyltrifluoroacetone, Hpta, and 2-thenoyltfifluoroacetone, Htta, were used as the β-diketones. The Lewis bases, B, were tetraethyl methylene diphosphonate, POPO, which was bidentate, and diethyl benzylphosphonate, PhPO which was monodentate. Based on the extraction data, the stability constants, log β, of the first complexes between tfis（β-diketonato）Eu（Ⅲ） and the phosphonate, EuA3B, were determined to be 6.0 for the POPO complex and 3.40 for the PhPO complex. The Eu（Ⅲ） luminescence intensity in the EuA3POPO was larger than EuA3 where A was either pta or tta at similar concentrations of Eu（Ⅲ）, while that in Eu（pta）3PhPO was stronger than EuA3; however, in Eu（tta）3PhPO, it was weaker than Eu（tta）3. The POPO functions as a sensitizer, and the PhPO functions as a quencher for the tta chelate and as a sensitizer for the pta chelate. From the lifetime and quantum yield, φ, of the Eu（Ⅲ） luminescence in the complexes as well as the observation of the extractability of Eu（Ⅲ） with the Hpta and the phosphonates and of the luminescence spectra of the complexes, it was confirmed that the extraction of Eu（Ⅲ） was remarkably enhanced with a β-diketonate and a strong Lewis base, and also the ternary complex that was formed as the extracted species, showed luminescence enhancement. This phenomenon may be due the formation of a strong bond between the Eu（Ⅲ） and the strong Lewis base leading to more hydrophobicity in the extracted species and also to more effective energy transfer from the Lewis base to the Eu（Ⅲ）. It was not significant whether the donor atoms were N or O.

Wind Tunnel Study on Wind and Turbulence Intensity Profiles in Wind Turbine Wake

In recent years, there has been a rapid development of the wind farms in Japan. It becomes very important to investigate the wind turbine arrangement in wind farm, in order that the wake of one wind turbine does not to interfere with the flow in other wind turbines. In such a case, in order to achieve the highest possible efficiency from the wind, and to install as many as possible wind turbines within a limited area, it becomes a necessity to study the mutual interference of the wake developed by wind turbines. However, there is no report related to the effect of the turbulence intensity of the external flow on the wake behind a wind turbine generated in the wind tunnel. In this paper, the measurement results of the averaged wind profile and turbulence intensity profile in the wake in the wind tunnel are shown when the turbulence intensity of the external wind was changed. The wind tunnel experiment is performed with 500mm-diameter two-bladed horizontal axis wind turbine and the wind velocity in wake is measured by an I-type hot wire probe. As a result, it is clarified that high turbulence intensities enable to the entrainment of the main flow and the wake and to recover quickly the velocity in the wake.

Most adults with auditory processing disorder(APD) often have trouble at work. It is possible that these listening problems in adults with APD influence their mental health. Thus, we have to take the subject's personality and mental status into consideration when supporting subjects with APD. However, studies on APD in adults are comparatively rare. In this study, we aimed to evaluate the relationships between the results of auditory tests, APD symptoms, and psychological status in subjects with APD. This study included 22 adults who complained of listening problems in everyday life, and they underwent auditory processing tests and were questioned regarding their psychological *** the results of the auditory processing tests, all subjects showed low scores on at least one test; therefore, they were suspected of having APD. Especially, the scores were low on the auditory memory test. Furthermore, subjects with APD show relatively similar psychological characteristics to each other. However, there was no significant relationship between the results of the auditory processing tests and psychological *** have to take notice of a subject's psychological state when they perceive their listening difficulties as a large-scale problem and feel anxious as a result. For this purpose, we should take psychological characteristics into consideration at the time of the medical examination interview before audio-psychological testing.

Bacterial infection associated with the use of medical or dental devices is a serious *** devices made of ethylene vinyl acetate（EVA） are often used in the oral cavity,there are no established standards for their *** investigated bacterial survival on EVA sheets under various storage conditions to establish a standard for hygienic storage of such dental *** counts were evaluated,which showed a significant decrease after washing with sterilized water,mechanical brushing and rinsing,and using Mouthguard Cleaner as compared to untreated *** addition,no bacteria were detected on samples stored 2 days or longer in a ventilated environment,whereas they were detected for up to 14 days on samples without any cleaning stored in a closed *** counts for the untreated samples gradually declined,while surviving bacteria on samples treated with sterilized water and mechanical brushing showed a rapid ***,bacterial identification using polymerase chain reaction（PCR） revealed that Streptococcus oralis was dominantly detected on salivary samples after 14 days of storage among both two *** effective hygienic storage of dental devices made of EVA,washing with sterilized water is important to remove absorbed salivary compounds along with storage in a ventilated environment.

Bilateral Cochlear implants (CIs) improved speech intelligibility, speech perception in background noise, and sound localization in quiet and noisy situations. However, it is unclear whether these advantages essentially result in binaural integration of acoustic stimuli from each ear. In this study, we investigated the effectiveness of binaural integration by bilateral CIs placement using binaural hearing tests and subjective auditory perceptual assessment. A 61-year-old bilateral CIs subject underwent the following four tests:the Japanese Hearing in Noise Test (HINT-J), the dichotic listening test (DLT), the Rapidly Alternating Speech Perception (RASP) test, and subjective auditory perceptual assessment. The HINT-J score was significantly higher for bilateral CIs than for a unilateral CI. However, DLT and the RASP test revealed contradictory results. Subjective auditory perceptual assessment revealed active and bright impressions for bilateral hearing, which were also noisy and strong compared with those for unilateral hearing. The results of this study revealed that bilateral CIs improved speech perception in background noise and an improved auditory impression, although the bilateral integration abilities were not improved. This was probably because the patient was required to combine information from the two ears into a single perception in DLT and the RASP test. More longitudinal data should be collected and analyzed in future studies to evaluate the long-term effects of bilateral CIs.
