Failure of P-selectin blockade alone to protect the liver from ischemia-reperfusion injury in the isolated blood-perfused rat liver

AIM： To determine if blockade of P-selectin in the isolated blood-perfused cold ex vivo rat liver model protects the liver from ischemia-reperfusion injury. METHODS： The effect of P-selectin blockade was assessed by employing an isolated blood-perfused cold ex vivo rat liver with or without P-selectin antibody treatment before and after 6 h of cold storage in University of Wisconsin solution. RESULTS： In our isolated blood-perfused rat liver model, pre-treatment with P-selectin antibody failed to protect the liver from ischemia-reperfusion injury, as judged by the elevated aspartate aminotransferase activity. In addition, P-selectin antibody treatment did not significantly reduced hepatic polymorphonuclear leukocyte accumulation after 120 min of perfusion. Histological evaluation of liver sections obtained at 120 min of perfusion showed significant oncotic necrosis in liver sections of both ischemic control and P-selectin antibody-treated groups. However, total bile production after 120 rain of perfusion was significantly greater in P-selectin antibody-treated livers, compared to control livers. No significant difference in P-selectin and ICAM-1 mRNAs and proteins, GSH, GSSG, and nuclear NF-kB was found between control and P-selectin antibody-treated livers. CONCLUSION： In conclusion, we have shown that blockade of P-selectin alone failed to reduced polymorphonuclear leukocyte accumulation in the liver and protect hepatocytes from ischemia-reperfusion injury in the isolated blood-perfused cold-ex vivo rat liver model.

Human embryonic stem cells(hESCs)undergo self-renewal while maintaining ***,the molecular mechanism that demonstrates how these cells maintain their undifferentiated state and how they self-renew is poorly ***,we characterized an aneuploidy H1 hESC subline(named H1T)using karyotyping and comparative genomic hybridization(CGH)*** the H1T hESC line displays a self-renewal advantage while maintaining an undiffer-entiated state,we speculated that the expression patterns of speciﬁc genes which are related to pluripotency or differentiation were altered;therefore,we attempted to screen for molecules that are propitious for maintenance of stemness by performing a combination of mRNA and CGH microarray analysis which compared the aneuploidy H1T hESC subline versus the euploid H1 hESC *** is discovered that some genes are up-regulated in H1T hESC subline such as TBX2 and Wnt3,while some are downregulated,for example,Fbxo7 and ***ﬁndings should fascilitate the study of the complex signaling network which maintains hESC pluripotency and function.

Understanding how plants respond to nitrogen in their environment is crucial for determining how they use it and how the nitrogen use affects other processes related to plant growth and development. Under nitrogen limitation the activity and affinity of uptake systems is increased in roots, and lateral root formation is regulated in order to adapt to low nitrogen levels and scavenge from the soil. Plants in the legume family can form associations with rhizobial nitrogen-fixing bacteria, and this association is tightly regulated by nitrogen levels. The effect of nitrogen on nodulation has been extensively investigated, but the effects of nodulation on plant nitrogen responses remain largely unclear. In this study, we integrated molecular and phenotypic data in the legume Medicago truncatula and determined that genes controlling nitrogen influx are differently expressed depending on whether plants are mock or rhizobia inoculated. We found that a functional autoregulation of nodulation pathway is required for roots to perceive, take up, and mobilize nitrogen as well as for normal root development. Our results together revealed that autoregulation of nodulation, root development, and the location of nitrogen are processes balanced by the whole plant system as part of a resource-partitioning mechanism.

We aimed to characterize, and analyze the presence of factors such as polypharmacy and personal medical history that predispose to Adverse Drug Reactions （ADRs） and potential preventability of these. The Civil Hospital of Culiacan （HCC）, in Sinaloa, where the study was conducted has 80 beds in total, in which the Department of Internal Medicine with 24 beds： 12 lbr men and 12 for women, we found the part of the share to contribute to the 200,000 cases of ADRs that according with the WHO each of the countries members report ever）, year to the Uppsala Monitoring Centre. We have not only but also ordered frequency of drugs, pathologies, and analytical tests of the hospitalized patients.
