Comparison of RFFIT Tests with Different standard sera and Testing Procedures

The World Health Organization (WHO) standard assay for determining antibody level is the rapid fluorescent focus inhibition test (RFFIT) and is used to determine the degree of immunity after vaccination against rabies. To compare the difference in RFFIT results between the laboratories of The National Institute of Infectious Disease in Japan (NIID) and the Chinese Centre for Disease Control (CCDC) as well the influence of the choice of standard serum (sTD) for the detection, the two laboratories detection methods were simultaneously manipulated by RFFIT. The reference serums used in NIID and the WHO standard serum used in CCDC were compared in the same RFFIT detection to determine the titer of four sera samples C1, s1, s2 and s4 in parallel, and the titers of the detected sera samples were calculated using the standard formula for neutralizing antibody titer. No significant difference was found in RFFIT methods from the two laboratories and the RFFIT testing procedures of the two laboratories have good consistency. However, different titers were obtained with the tentative internal standard serum (TI-sTD) produced by adjusting to 2.0 IU of WHO standard serum in NIID and the WHO sTD. The titer determined with the TI-sTD was higher than that determined with WHO sTD, This difference appears to be significant and requires further investigation.

Energy Assessment of Building Integrated Photovoltaics and Fuel Cell systems: Design study for Building(s) of Mie, Japan

A building integrated energy system (photovoltaic (PV) and fuel cell (FC)) is proposed for assessment of the energy self-sufficiency rate in five cities of Mie prefecture in Japan. In this work, it is considered that the electricity requirement of the building is provided by the building integrated photovoltaic (BIPV) system and the gap between the energy demand and BIPV supply is fulfilled by the FC. The FC is powered by the electrolytic H2 produced from the surplus power of PV. A design study of using the proposed system in five cities in Mie prefecture, which are in center part of Japan, has been performed. It has been observed that the monthly power production from BIPV is higher in spring and summer, while it is lower in autumn and winter at all considered locations. The self-sufficiency rate of the FC system is higher with decreasing households’ number and it has been observed that the 12 households are more suitable for full cover of the electricity demand by the combined system of PV and FC. The relationship between the households’ number and self-sufficiency rate of the FC system per solar PV installation area can be expressed by exponential curve. The coefficient of the exponential curve can predict the suitable city for the BIPV system with FC system utilizing electrolytic H2 generated by using excess energy from the PV system.

We describe a multi-lingual speech-to-speech translation （s2sT） system, which is mainly focused on translation between English and Asian languages （Japanese and Chinese）. There are three main modules of our s2sT system: large vocabulary continuous speech recognition, machine text-to-text （T2T） translation, and text-to-speech synthesis. All of them are multilingual and are designed using state-of-the-art technologies. A corpus-based statistical machine learning framework forms the basis of our system. We use a parallel multilingual database consisting of over 1,000,000 sentences that cover a broad range of travel-related conversations. Recent evaluation of the overall system showed that speech-to-speech translation quality is high, being at the level of a person having a TOEIC score of 650 out of the perfect score of 990. Additionally, field experiment results in real various places will be introduced

Long-term survival after resection of pancreatic cancer:A single-center retrospective analysis

AIM: To retrospectively analyze factors affecting the long-term survival of patients with pancreatic cancer who underwent pancreatic resection.METHODs: From January 2000 to December 2011,195 patients underwent pancreatic resection in our hospital.The prognostic factors after pancreatic resection were analyzed in all 195 patients.After excluding the censored cases within an observational period,the clinicopathological characteristics of 20 patients who survived ≥ 5(n = 20) and s were compared.For this comparison,we analyzed the patients who underwent surgery before June 2008 and were observed for more than 5 years.For statistical analyses,the log-rank test was used to compare the cumulative survival rates,and the χ2 and Mann-Whitney tests were used to compare the two groups.The CoxHazard model was used for a multivariate analysis,and P values less than 0.05 were considered significant.A multivariate analysis was conducted on the factors that were significant in the univariate analysis.REsULTs: The median survival for all patients was 27.1 months,and the 5-year actuarial survival rate was 34.5%.The median observational period was 595 d.With the univariate analysis,the UICC stage was significantly associated with survival time,and the CA19-9 ≤ 200 U/m L,DUPAN-2 ≤ 180 U/m L,t u m o r s i ze ≤ 2 0 m m,R 0 re s e c t i o n,a b s e n c e o f lymph node metastasis,absence of extrapancreatic neural invasion,and absence of portal invasion were favorable prognostic factors.The multivariate analysis showed that tumor size ≤ 20 mm(HR = 0.40; 95%CI: 0.17-0.83,P = 0.012) and negative surgical margins(R0 resection)(HR = 0.48; 95%CI: 0.30-0.77,P = 0.003) were independent favorable prognostic factors.Among the 96 patients,20 patients survived for 5 years or more,and 76 patients died within 5 years after operation.Comparison of the 20 5-year survivors with the 76 non-survivors showed that lower concentrations of DUPAN-2(79.5 vs 312.5 U/mL,P = 0.032),tumor size ≤ 20 mm(35% vs 8%,P = 0.008),R0 resection(95% vs 61%,P =

Effects of L-carnitine and Branched-Chain Amino Acids on Energy Metabolism,Body Composition,and Delayed-Onset Muscle soreness after Exercise in Healthy subjects

Background specific nutrients combined with exercise may prevent a loss of lean body mass and consequently prevent sarcopenia.We examined the effects of 2 weeks of supplementation with L-carnitine and a single dose of branched-chain amino acids(BCAAs)on energy metabolism and body composition before and after exercise and on post-exercise muscle soreness in healthy subjects.Methods A total of 12 young,untrained,healthy women were randomly assigned to two groups who received either 2 weeks of supplementation with L-carnitine(1000 mg/day)and a single dose of BCAAs plus 200 kcal energy(LCAR+BCAA group,n=6)or 200 kcal energy alone(control group,n=6)before 60 min of exercise.Energy substrate metabolism and body composition by bio-impedance were examined before and after exercise.Delayed-onset muscle soreness(DOMs)was evaluated by using a visual analogue scale for 3 days after exercise.Results serum concentrations of free and acyl-L-carnitine in the LCAR+BCAA group were not significantly different from those in the control group.There were no significant differences in body composition before or after exercise between the two groups.Although serum free fatty acid levels 120 min after the oral ingestion of BCAAs were significantly lower in the LCAR+BCAA group than in the control group,there were no significant differences in serum free fatty acid levels immediately after and 60 min after exercise.DOMs for 3 days after exercise was lower in the LCAR+BCAA group than in the control group,but this difference was not significant.Conclusion Continuous supplementation with L-carnitine combined with a dose of BCAAs in young,untrained,healthy subjects caused a blunted response of stimulated lipolysis after exercise.

Application of TERRA/MODIs images, TM images and weather data to assess the effect of cold damage on rice yield

Data assimilation is extensively applied in agricultural remote sensing application.However,integration of multi-temporal and high spatial resolution images with crop growth model to evaluate the effect of cold damage on paddy rice was still lacking.In this paper,authors applied data assimilation combining LANDsAT/TM,a series of terra MODIs images with sIMRIW model to detect how cold damage affected paddy rice yield per unit in the Wuchang county,Heilongjiang province for the year 2006.In the study,MODIs images selected corresponding to a series of the key rice growth phases were utilized to retrieve daily LAI values that were needed in the sIMRIW model.Meanwhile,TM was applied to accurately extract paddy rice sown areas.The study results showed that the yield per unit was 10,628.5840 kg/ha under cold damage condition,which was little less than 10,768.3210 kg/ha under optimal condition.Moreover,the ratio of the calculated yield per value under cold damage condition to the actual value of paddy rice yield per unit was 0.56.The result was better than that acquired in UsA and Japan.The results of this study expected to provide suggestions to policy-makers and reference to related research.

Detection of Non-Typhoidal salmonella Using a Mechanism for Controlling Hydrogen sulfide Production

To know the relation between NaCl concentration and hydrogen sulfide (H2s) production by MY Phenomenon of salmonella, we developed a method to control H2s production by salmonella in this study. Desoxycholate-hydrogen sulfide-lactose (DHL) agar was used as the selective medium for salmonella. Triple sugar iron (TsI) agar was used as the screening medium. Citric acid disks were placed on DHL agar. NaCl was added to both media. After 24 hours incubation, H2s production on DHL agar was stimulated by citric acid, whereas production was suppressed by adding NaCl. The color change in the bottom of the TsI medium was due to the low production of black Fes. The production of Fes was reduced by lower H2s production by salmonella with an increasing concentration of NaCl from 0.5% to 3%. The inhibition of the black color meant that the color change due to glucose fermentation could be seen clearly in the bottom of the agar. The current method cannot detect glucose fermentation but the addition of more NaCl to the TsI medium allowed the visualization of glucose fermentation by salmonella. Although H2s production of salmonella was promoted by citrate etc., being controlled by sodium chloride was proved in this study. Thus, we had developed a possibility of novel technique for the detection of salmonella etc.

Depletion of CD25^+CD4^+T cells (Tregs) enhances the HBV-specific CD8^+ T cell response primed by DNA immunization

AIM: Persistent hepatitis B virus (HBV) infection is characterized by a weak CD8+ T cell response to HBV. Immunotherapeutic strategies that overcome tolerance and boost these suboptimal responses may facilitate viral clearance in chronically infected individuals. Therefore, we examined whether CD25+CD4+ regulatory T (Treg) cells might be involved in a inhibition of CD8+T cell priming or in the modulation of the magnitude of the'peak' antiviral CD8+ T cell response primed by DNA immunization. METHODs: B10.D2 mice were immunized once with plasmid pCMV-s. Mice received 500 μg of anti-CD25 mAb injected intraperitoneally 3 d before DNA immunization to deplete CD25+ cells. Induction of HBV-specific CD8+ T ceils in peripheral blood mononuclear cells (PBMCs) was measured by s28-39 peptide loaded DimerX staining and their function was analyzed by intracellular IFN-γ staining.REsULTs: DNA immunization induced HBV-specific CD8+ T cells. At the peak T cell response (d 10), 7.1±2.0% of CD8+ T cells were HBV-specific after DNA immunization, whereas 12.7±3.2% of CD8+ T cells were HBV-specific in Treg-depleted mice, suggesting that DNA immunization induced more antigen-specific CD8+ T cells in the absence of CD25+ Treg cells (n = 6, P＜0.05). similarly, fewer HBVspecific memory T cells were detected in the presence of these cells (1.3±0.4%) in comparison to Treg-depleted mice (2.6±0.9%) on d 30 after DNA immunization (n = 6, P＜0.01). Both IFN-γ production and the avidity of the HBV-specific CD8+ T cell response to antigen were higher in HBV-specific CD8+ T cells induced in the absence of Treg cells.CONCLUsION: CD25+ Treg cells suppress priming and/or expansion of antigen-specific CD8+ T cells during DNA immunization and the peak CD8+ T cell response is enhanced by depleting this cell population. Furthermore, Treg cells appear to be involved in the contraction phase of the CD8+ T ceil response and may affect the quality of memory T cell pools. AIM: To study the different death pathways in human cervical cancer HeLa and melanoma A375-s2 cells Initiated by evodiamine.
