Evaluation of thermostable endoglucanase in Paenibacillus lautus strain BHU3 for yield enhancement

In the present study,Paenibacillus lautus strain BHU3 isolated from landfill soil was evaluated for the presence of potential endoglucanases which are the first candidate of cellulase enzyme system to act on ***-silico analysis revealed high potential thermostable endoglucanases which can efficiently interact with *** most potent and thermostable endoglucanase(locus tag ***30_18280)belonged to glycosyl family-5 and had interaction energy of−12.981 kcal/mol for the best docked cluster containing three out of ten docking conformations,and Tm value of 73.3℃.MD simulation of 100 ns proved highly stable binding interactions of CPZ30_18280 endoglucanase with cellulose with root mean square deviation(RMSD)values ranging from 0.15 to 0.30 *** interactions with characteristic active site residues(tyrosine,tryptophan and aspartate)of glycosyl family-5 endoglucanases were ***,to enhance the production of endoglucanases,the fermentation conditions were optimized employing approaches like one factor at a time(OFAT)and response surface methodology(RSM).Maximum activity of endoglucanase was determined at 60℃.The optimized condi-tions for enhanced production of endoglucanase(10.15 U/mL)were pH 6.63,yeast extract conc.3.44 g/L,wheat bran 3.59%,and inoculum size 2.65%.Hence,*** strain BHU3 has enormous potential to synthesize highly efficient thermostable endoglucanases under optimized regime using ***,it could find immense industrial applications including large scale cellulose conversion to bioethanol.

On-current modeling of short-channel double-gate(DG) MOSFETs with a vertical Gaussian-like doping profile

An analytic drain current model is presented for doped short-channel double-gate MOSFETs with a Gaussian-like doping profile in the vertical direction of the channel. The present model is valid in linear and satura- tion regions of device operation. The drain current variation with various device parameters has been demonstrated. The model is made more physical by incorporating the channel length modulation effect. Parameters like transcon- ductance and drain conductance that are important in assessing the analog performance of the device have also been formulated. The model results are validated by numerical simulation results obtained by using the commercially available ATLAS^TM, a two dimensional device simulator from SILVACO.

Optimisation of Adventitious Shoot Regeneration and Agrobacterium-mediated Transformation in Canna × generalis(Canna Lily)

Canna being ornamental plants has a significant role in agriculture, medical, economy and food industry. Canna has a limited vase life due to the rapid loss of moisture from its perianth. For improving its market value, cuticularisation of the perianth can be achieved by the expression of a heterologous cutin producing gene, using the tissue culture and transformation protocol developed in this study. Efficient, rapid and direct adventitious shoot regeneration was successfully established in Canna × generalis using recalcitrant rhizome explants. The explants were cultured on MS medium supplemented with 6-benzylaminopurine(6-BA), thidiazuron(TDZ), and kinetin. Among the four genotypes taken for tissue culture, the ‘Trinacria variegata' was the best responding cultivar. And 2 mg · L^(-1)6-BA or 1.5 mg · L^(-1) TDZ along with 0.1 mg · L^(-1) IAA was optimum for their regeneration. The highest regeneration was achieved in ‘Trinacria variegata'(36%) on 6-BA, 33% on TDZ while kinetin failed to evoke any regenerative responses. Regeneration was enhanced by supplementation of 100 mg · L^(-1) ascorbic acid(AsA), while, 100 mg · L^(-1) of l-cysteine or 100 mg · L^(-1) dithiothreitol(DTT), inhibited regeneration. Shoots were observed to develop 3–5 fibrous roots on MS medium supplemented with 0.5 mg · L^(-1) indole-3-butyric acid(IBA). The plantlets were transplanted into pots and acclimatised in glasshouse with 100%survival. For transformation of Canna, rhizome explants were co-cultivated for 60 min in Agrobacterium suspension. The explants were washed with 500 mg · L^(-1) cefotaxime solution, subjected to 100 mg · L^(-1) kanamycin selection followed by excision of the shoots and culturing them on IBA-supplemented media for root development. Transgene integration in the putative transformants was confirmed by PCR assay and copy number by Southern blot hybridisation analysis.

Synthesis,isolation,identification and characterization of new process-related impurity in isoproterenol hydrochloride by HPLC,LC/ESI-MS and NMR

One unknown impurity （Imp-II） during the analysis of laboratory batches of isoproterenol hydrochloride was detected in the level ranging from 0.04% to 0.12% by high performance liquid chromatography with UV detection. The unknown impurity structure was proposed as 4-[2-（propan-2oylamino）ethyl]benzene-l,2-diol （Imp-II） using the liquid chromatography-mass spectrophotometry （LC-MS） analysis. Imp-II was isolated by semi-preparative liquid chromatography from the impurity-enriched reaction crude sample. Its proposed structure was confirmed by nuclear magnetic spectroscopy such as 1H, 13C, DEPT （1D NMR）, HSQC （2D NMR） and infrared spectroscopy （IR）, and retention time and purity with HPLC followed by the chemical synthesis. Due to less removable nature of Imp-II during the purification, the synthetic process was optimized proficiently to control the formation of Imp-II below to the limit 〈 0.12% in the course of reaction. The new chemical route was developed for the preparation of this impurity in required quantity with purity to use as reference standard. The most probable mechanism for the formation of Imp-II was discussed in details.

The present work gives some insight into the subthreshold behaviour of short-channel double-material- gate strained-silicon on silicon-germanium MOSFETs in terms of subthreshold swing and off-current. The formu- lation of subthreshold current and, thereupon, the subthreshold swing have been done by exploiting the expression of potential distribution in the channel region of the device. The dependence of the subthreshold characteristics on the device parameters, such as Ge mole fraction, gate length ratio, work function of control gate metal and gate length, has been tested in detail. The analytical models have been validated by the numerical simulation results that were obtained from the device simulation software ATLASTM by Silvaco Inc.

Biochar-mediated removal of pharmaceutical compounds from aqueous matrices via adsorption

Pharmaceutical is one of the noteworthy classes of emerging contaminants. These biologically active compounds pose a range of deleterious impacts on human health and the environment. This is attributed to their refractory behavior, poor biodegradability, and pseudopersistent nature. Their large-scale production by pharmaceutical industries and subsequent widespread utilization in hospitals, community health centers, and veterinary facilities, among others, have significantly increased the occurrence of pharmaceutical residues in various environmental compartments. Several technologies are currently being evaluated to eliminate pharmaceutical compounds (PCs) from aqueous environments. Among them, adsorption appears as the most viable treatment option because of its operational simplicity and low cost. Intensive research and development efforts are, therefore, currently underway to develop inexpensive adsorbents for the effective abatement of PCs. Although numerous adsorbents have been investigated for the removal of PCs in recent years, biochar-based adsorbents have garnered tremendous scientific attention to eliminate PCs from aqueous matrices because of their decent specific surface area, tunable surface chemistry, scalable production, and environmentally benign nature. This review, therefore, attempts to provide an overview of the latest progress in the application of biochar for the removal of PCs from wastewater. Additionally, the fundamental knowledge gaps in the domain knowledge are identified and novel strategic research guidelines are laid out to make further advances in this promising approach towards sustainable development.

Phosphorus partitioning and recovery of low-phosphorus iron-rich compounds through physical separation of Linz-Donawitz slag

The Linz-Donawitz（LD） steelmaking process produces LD slag at a rate of about 125 kg/t. After metallic scrap recovery, the non-metallic LD slag is rejected because its physical/chemical properties are unsuitable for recycling. X-ray diffraction（XRD） studies have indicated that non-metallic LD slag contains a substantial quantity of mineral phases such as di- and tricalcium silicates. The availability of these mineral phases indicates that LD slag can be recycled by iron（Fe）-ore sintering. However, the presence of 1.2wt% phosphorus（P） in the slag renders the material unsuitable for sintering operations. Electron probe microscopic analysis（EPMA） studies indicated concentration of phosphorus in dicalcium silicate phase as calcium phosphate. The Fe-bearing phases（i.e., wustite and dicalcium ferrite） showed comparatively lower concentrations of P compared with other phases in the slag. Attempts were made to lower the P content of LD slag by adopting various beneficiation techniques. Dry high-intensity magnetic separation and jigging were performed on as-received samples with particle sizes of 6 and 3 mm. Spiral separation was conducted using samples ground to sizes of less than 1 and 0.5 mm. Among these studies, grinding to 0.5 mm followed by spiral concentration demonstrated the best results, yielding a concentrate with about 0.75wt% P and 45wt% Fe.

This paper presents the analytical modeling of subthreshold current and subthreshold swing of short- channel fully-depleted （FD） strained-Si-on-insulator （SSOI） MOSFETs having vertical Gaussian-like doping pro- file in the channel. The subthreshold current and subthreshold swing have been derived using the parabolic approx- imation method. In addition to the effect of strain on silicon layer, various other device parameters such as channel length （L）, gate-oxide thickness （tox）, strained-Si channel thickness （ts_Si）, peak doping concentration （Np）, project range （Rp） and straggle （op） of the Gaussian profile have been considered while predicting the device characteris- tics. The present work may help to overcome the degradation in subthreshold characteristics with strain engineering. These subthreshold current and swing models provide valuable information for strained-Si MOSFET design. Ac- curacy of the proposed models is verified using the commercially available ATLASTM, a two-dimensional （2D） device simulator from SILVACO.
