The current study was carried out to determ.ne the bioactivity of P. lentiscus leaf extracts as potential antibacterial and antioxidant properties. The plant extracts were exam.ned for antibacterial activity against antibiotic-resistant Staphylococcus aureus, Staphylococcus haem.lyticus, Pseudom.nas aeruginosa, and Proteus m.rabilis using the agar well m.thod (according to the guidelines of Clinical and Laboratory Standard Institute). The antioxidant potential of 3 plant leaf extracts was determ.ned by their ability to convert Fe3+ to Fe2+ and scavenge the DPPH free radical. At all concentrations studied, the m.thanolic leaf extract had higher total phenolic and flavonoid content, as well as stronger antioxidant and antibacterial inhibitory activity com.ared to aqueous extract. Our findings with P. aeruginosa were especially interesting, because this bacterium.was inhibited by m.thanol extract than that of the reference antibiotics. The results also dem.nstrated a link between DPPH radical scavenging ability, reducing power, and total phenolic and flavonoid content of plant extracts (r > 0.97, R2 > 0.95, P = 0.01). As a result, the m.thanolic leaf extract of the chosen plant m.ght be em.loyed as an effective antioxidant and antibacterial agent for the treatm.nt of a variety of m.rbidities.

m.tabolic engineering and synthetic biology endeavors benefit from.prom.ters that perform.consistently(or robustly)with respect to cellular growth phase(exponential and stationary)and ferm.ntation scale(m.crotiter plates,tubes,flasks,and bioreactors).However,nearly all endogenous prom.ters(especially in Saccharom.ces cerevisiae)do not perform.in this *** this work,a hybrid prom.ter engineering strategy is leveraged to create novel synthetic prom.ters with robustness across these *** a m.lti-dim.nsional RNA-seq dataset,prom.ters with specific phase dependencies were *** enriched with functional transcription factors were identified using m.m. *** m.tif-containing fragm.nts could im.art activity dependence in the opposing ***,we obtain two new prom.ters with high and consistent expression across both phases by increasing the exponential phase activity of the starting stationary-phase scaffold by 38 and 23-fold ***,we show that these prom.ters function consistently across various laboratory growth scales over tim. in a m.crotiter plate and in ***,this work presents and validates a new strategy for engineering prom.ters in *** with high levels of expression that are robust to cellular growth phase and the scale of the culture.

Steroidal glycoalkaloids(SGAs)are specialized m.tabolites produced by hundreds of Solanum.species,including im.ortant vegetable crops such as tom.to,potato,and *** it has been known that SGAs play im.ortant roles in defense in plants and“anti-nutritional"effects(e.g.,toxicity and bitterness)to hum.ns,m.ny of these m.lecules have docum.nted anti-cancer,anti-m.crobial,antiinflam.atory,anti-viral,and anti-pyretic *** these,α-solasonine andα-solam.rgine isolated from.black nightshade(Solanum.nigrum.are reported to have potent anti-tum.r,anti-proliferative,and anti-inflam.atory ***,α-solasonine andα-solam.rgine,along with the core steroidal aglycone solasodine,are the m.st widespread SGAs produced am.ng the Solanum.***,it is still unknown how plants synthesize these bioactive steroidal *** com.arative m.tabolom.ctranscriptom.-guided approach,biosynthetic logic,com.inatorial expression in Nicotiana bentham.ana,and functional recom.inant enzym. assays,here we report the discovery of 12 enzym.s from.*** that converts the starting cholesterol precursorto solasodine aglycone,and the downstream.-solasonine,α-solam.rgine,and m.lonyl-solam.rgine SGA *** further identified six enzym.s from.cultivated eggplant that catalyze the production ofα-solasonine,α-solam.rgine,and m.lonyl-solam.rgine SGAs from.solasodine aglycone via glycosylation and atypical m.lonylation *** work provides the gene tool box and platform.for engineering the production of high-value,steroidal bioactive m.lecules inheterologoushosts usingsyntheticbiology.
