Construction,expression and binding specificity of bispecific CD3×VEGFR-2 and CD3×NCAM antibodies in the single chain and diabody format

Bispecific antibodies are recombinant proteins with novel immunological properties and therapeutic potential. Recombinant protein quality and activity of several bispecific antibodies comprising different variable domain combinations with respect to the parental monospecific single chain fragments (scFv) were evaluated after expression in bacteria or mammalian cells. The parental scFv proteins humanized anti-NCAM scFv, murine anti-VEGFR-2 scFv, murine and humanized anti-CD3 scFv, respectively, could successfully be expressed in E. coli, whereas the murine anti-NCAM scFv version could not be reliably detected. Bispecific CD3 × VEGFR-2 and CD3 × NCAM anti-bodies were expressed in the bispecific single chain and the single chain diabody format. However, the diabody derived from the murine anti-NCAM scFv could not efficiently be expressed in E. coli or in mammalian cells. Significant binding of the CD3 × NCAM single chain diabody comprising the humanized version of anti-CD3 and humanized version of anti-NCAM was efficient to both antigens. Nevertheless, binding of the bispecific single chain version to the NCAM antigen was inefficient in comparison to CD3 binding. In conclusion, the data could indicate that the result of scFv expression in bacteria may be predictive for the chances of success for functional expression of more complex bispecific derivatives.

The interest in using the Datura stramonium plant is due to its natural products,which are used in many pharmaceutical *** objective of the current study was to assess the therapeutic and cytotoxic effects of the *** plant on two types of human cancer cell models(MCF7 and HT29)in vitro.A soxhlet apparatus was used to obtain methanolic extract from dried plant *** recovered crude,after the solvent had evaporated,was then dispersed at varied concentrations of extract 100,50,20,and 0.0µg/mL and tested to see how the cells ***,the cancer-testis antigen(CTA)gene transcription in the two cell types exposed to the plant extract was examined using a semi-quantitative real-time polymerase chain *** chromatography–mass spectrometry(GC-MS)results produced the significant main metabolites Nonanoic acid,Tropine N-Oxide,3,6-Ditigloyloxy-7-hydroxytropane,Hexadecanoic acid,2-Pentadecanone,6,10,14-trimethyl-,Carvenone,methyl ester,Phytol,Aposcopolamine,Hyoscyamine,4,8,12,16-Tetramethylheptadecan-4-olide,Scopolamine,Alpha.-Tocospiro A,1,2-Cycloheptanedione,3,3,7,7-tetramethyl-,dihydrazone,Campesterol,Stigmasterol,Gamma-Sitosterol and dl-.alpha.-*** results showed that the two types of cell lines impacted by *** extract,through untreated type 1 cells(MCF7)gave a highly significant transcription according to all applicable *** implemented analyses cleared the strong genetic impacts of Datura extract on cancer cells’***2LY and C2orf63 transcript accumulation were also significantly elevated when exposed to plant extract at a level of 50µg/mL in cell line type 2(HT29),but TGIF2LY and P53 had the lowest relative expression at a level of 100µg/mL when treated the same cell line type.

目的汉化护理学校归属感量表,并检验中文版护理学校归属感量表的信效度。方法依据Brislin原则翻译护理学校归属感量表,由8名专家评价中文版护理学校归属感量表的内容效度。2022年8月至2023年10月,采用中文版护理学校归属感量表调查330名护理本科生以检验其信效度。结果中文版护理学校归属感量表包含4个分量表共19个条目。量表水平的内容效度指数为0.947,条目水平的内容效度指数为0.875~1.000,量表的Cronbach′sα系数为0.965。探索性因子分析提取4个公因子,累计方差贡献率为83.766%;验证性因子分析结果显示,χ^(2)/df=2.672,RMSEA=0.088,CFI=0.938,RMR=0.027,NFI=0.906,IFI=0.939。结论中文版护理学校归属感量表信效度良好,可用于测评护理专业学生的学校归属感水平。

Optimizing Model Land Use and Crop Productivity in Agroforestry Farms for Food Security of Small Farmers in Burundi

Burundi faces major agricultural constraints, including land fragmentation, soil erosion, limited access to inputs, inadequate infrastructure and demographic pressures that exacerbate food insecurity. In order to address the multiple challenges faced by farmers in rural areas, a study on improving agricultural productivity and food security in Burundi through optimized land use and diversified farming practices in agroforestry systems has been carried out. The study area is the communes of Giheta and Rutegama, all located in Burundi’s humid plateau livelihood zone, and involved 164 households grouped in coffee growing cooperatives supervised by the cooperative consortium COCOCA. The study uses a mathematical programming model to determine optimal crop selection based on factors such as production costs, yields and market demand. The findings of the study revealed significant insights into the demographic and socio-economic characteristics of the sampled population. Notably, 98.8% of respondents were engaged in agriculture, confirming the predominantly agricultural nature of Burundi. The results indicated that maize is the most important crop, occupying 33.9% of the average total cultivated area, followed by cassava at 26.5% and bananas at 19.4%. Together, these three crops accounted for a substantial portion of the total cultivated area, highlighting their significance in local agriculture. Beans and potatoes also play a role, occupying 14.4% and smaller areas, respectively. In terms of profitability, the study provides a detailed analysis of profit margins by crop. Bananas emerges as the most profitable crop, with a profit margin of 97.3%, followed closely by cassava at 96.1% and rice at 90.5%. These crops not only offered substantial yields relative to their production costs but also benefited from strong market demand.
