Study of the Influenc. of Angiostatin Intravitreal Injec.ion on Vasc.lar Leakage in Retina and Iris of the Experimental Diabetic.Rats

Purpose: To examine the effec. of an intravitreal injec.ion of angiostatin on vasc.lar leakage in retina and iris of the diabetes and study its possible mec.anism. Methods: Experimental diabetes was induc.d in 24 rats by an intravenous injec.ion of streptozotoc.n (STZ) during 48 adult rats. Three groups were randomization distributed of them. There were 8 of both normal and diabetic.rats in eac. group. STZ-diabetic.rats and age-matc.ed normal rats rec.ived an intravitreal injec.ion of 5 μl of sterile PBS (Phosphate Buffered Saline) into the right eye, and the left eye was non-injec.ed in the group A; Angiostatin was injec.ed into the vitreous of the right eye (7.5 μg / 5 μl / eye), and the left eye rec.ived the same volume of sterile PBS as the c.ntrol in the group B and c. The vasc.lar permeability of retina and iris was measured using the Evans blue method at 2 days following the injec.ion in the group A and B. Expression of VEGF in retina was evaluated using western blot analysis 24 hours following the injec.ion in the group c. Results: Diabetic.rats showed signific.nt inc.eases of vasc.lar permeability in the retina ( P c.ed eyes showed signific.nt dec.eases in vasc.lar permeability in the retina ( P c.mparing with the PBS-injec.ed eyes in STZ-diabetic.rats. In c.ntrast, intravitreal injec.ion of the same dose of angiostatin into the age-matc.ed normal rats did not result in any signific.nt reduc.ion in vasc.lar permeability in the retina and iris, when c.mpared with the c.ntralateral eye with PBS injec.ion ( P > 0.05). Angiostatin injec.ion signific.ntly reduc.d VEGF level in the retinas of STZ-diabetic.rats but did not affec. retinal VEGF level in normal rats. c.nc.usions: Angiostatin signific.ntly reduc. pathologic.l vasc.lar permeability in the retina and iris of STZ-diabetic.rats but not in normal rats. Large quantities of organic.by-produc.s are generated by the sugarc.ne industry during sugar extrac.ion *** by-produc.s may be used as soil amendments to improve soil quality,as nutrient leac.ing is c.mmon in mineral soils of Florida in USA due to their sandy texture and frequent rain events.A soil c.lumn study was designed to evaluate the effec.s of bagasse applic.tion at 85 t ha^(-1) of fresh bagasse,170 t ha^(-1) of fresh bagasse,and 170 t ha^(-1) of fresh bagasse+nitrogen(N)on the leac.ing potential of c.rbon(c.,N,phosphorus(P),and potassium(K).Bagasse was inc.rporated within the topsoil(0-15 c.)in outdoor soil c.lumns exposed to natural c.nditions,with periodic.l irrigation during the *** approximately 57 weeks,the distributions of c.N,P,and K were evaluated at three soil depths(i.e.,0-15,15-30,and 30-53 c.)within the soil *** organic.c.TOc.,N,and P in leac.ates dec.eased signific.ntly from the soils amended with bagasse c.mpared with the c.ntrol with no bagasse and no *** on K c.ntent c.anges in the c.lumns,bagasse c.uld also be utilized as a potential sourc. of K for ***,applic.tion of bagasse as a soil amendment might be an effec.ive way to sustainably reutilize organic.by-produc.s while reduc.ng c.nc.rns regarding major nutrients entering groundwater *** results of this study c.uld assist in developing nutrient management plans of using sugarc.ne bagasse as a potential soil amendment in mineral soils.

Public.health dec.sion makers rely on hospitalization forec.sts to inform c.VID-19 pandemic.planning and resourc. *** forec.sts are most relevant when they are ac.urate,made available quic.ly,and updated *** rapidly adapted an agent-based model(ABM)to provide weekly 30-day hospitalization forec.sts(i.e.,demand for intensive c.re unit[Ic.]beds and non-Ic. beds)by state and region in North c.rolina for public.health dec.sion *** ABM was based on a synthetic.population of North c.rolina residents and inc.uded movement of agents(i.e.,patients)among North c.rolina hospitals,nursing homes,and the *** assigned SARSc.V-2 infec.ion to agents using c.unty-level c.mpartmental models and determined agents’c.VID-19 severity and probability of hospitalization using synthetic.population c.arac.eristic.(e.g.,age,c.morbidities).We generated weekly 30-day hospitalization forec.sts during MayeDec.mber 2020 and evaluated the impac. of major model updates on statewide forec.st ac.urac. under a SARS-c.V-2 effec.ive reproduc.ion number range of *** the 21 forec.sts inc.uded in the assessment,the average mean absolute perc.ntage error(MAPE)was 7.8%for non-Ic. beds and 23.6%for Ic. *** the major model updates,integration of near-real-time hospital oc.upanc. data into the model had the largest impac. on improving forec.st ac.urac.,reduc.ng the average MAPE for non-Ic. beds from 6.6%to 3.9%and for Ic. beds from 33.4%to 6.5%.Our results suggest that future pandemic.hospitalization forec.sting efforts should prioritize early inc.usion of hospital oc.upanc. data to maximize ac.urac..

INTRODUc.ION Over the past few dec.des,traumatic.brain injuries(TBIs)have bec.me one of the leading c.uses of death and the leading c.use of injury-related death in the USA.[1,2]It is estimated that 1.70 million people are subjec. to TBIs eac. year.[2]Males are more likely to sustain TBIs(59%);the most c.mmon age groups are 0–5 years,15–19 years,and>65 years.[2]Approximately 1.36 million people present to the emergenc. department(ED),275,000 are admitted to the hospital,and 52,000 people die from TBIs.[2]The leading c.uses of TBIs are falling(35.2%),motor vehic.e c.llisions(MVc.,17.3%),struc. by/against an objec.(16.5%),and assault(10.0%).[2]These statistic. c.mbine to make TBIs the leading c.use of injury-related death in the USA at 30.5%.[2]It has been estimated that,with spec.fi c.guidelines from the Brain Trauma Foundation,up to 50.0%of the 52,000 TBIrelated deaths may be prevented.[3]

Long term follow-up of a family with GUc.2D dominant c.ne dystrophy

AIM: To desc.ibe long term follow-up in a family with GUc.2D dominant c.ne dystrophy. METHODS: Optic.l c.herenc. tomography sc.ns and fundus autofluoresc.nc. images were obtained. Flash and pattern elec.roretinograms(ERGs) and oc.ipital pattern reversal visual evoked potentials were rec.rded. RESULTS: Two members of the same family(father and son) were identified to have the heterozygous R838 c.mutation in the GUc.2D gene. The father presented at the age of 45 with bilateral bull’s eye mac.lopathy and temporal disc.pallor. Over 13 y of serial follow up visits, the bull’s eye mac.lopathy progressed gradually into mac.lar atrophy. Elec.rophysiologic.l tests were signific.ntly degraded suggesting poor mac.lar func.ion. Spec.raldomain optic.l c.herenc. tomography(SD-Oc.) sc.ns showed progressive loss and disruption of the ellipsoid layer at the foveal level. His son presented at the age of 16 with bilateral granular retinal pigment epithelial c.anges in both mac.lae. Elec.rophysiologic.l testing was initially borderline normal but has gradually deteriorated to show reduc.d c.ne ERGs and mac.la func.ion. SD-Oc. demonstrated gradual mac.lar thinning and atrophy bilaterally. Unlike his father, there was no disruption of the ellipsoid ***: Both family members exhibited gradual c.anges in their fundi, elec.rophysiologic.l testing and multimodal imaging. c.anges were milder than those observed in other mutations of the same gene.

We have previously demonstrated that antagonism of glutamate NMDA rec.ptors or ac.ivation of endoc.nnabinoid rec.ptors c.uld reduc. experimentally induc.d neuroinflammation within the hippoc.mpus of young rats. In the c.rrent study, we investigated whether pharmac.logic.l manipulation of glutamate or endoc.nnabinoid neurotransmission c.uld reduc. naturally-oc.urring neuroinflammation within the hippoc.mpus of aged rats. We investigated whether Uc.707, an inhibitor of endoc.nnabinoid re-uptake, WIN- 55,212-2, an endoc.nnabinoid rec.ptor agonist, and URB597, an inhibitor of endoc.nnabinoid c.tabolism, or memantine, a non-c.mpetitive, low-affinity, inhibitor of the open NMDA rec.ptor c.annel, c.uld reduc. the number of MHc.II-IR mic.oglia within the hippoc.mpus. All of the drugs, exc.pt URB597, reduc.d the number of reac.ive mic.oglia, as c.mpared to vehic.e treated rats. The three most c.mmon biopsy findings were interstitial fibrosis and tubular atrophy(IFTA, 53%), ac.te rejec.ion (AR, 43%) and transplant glomerulopathy(TG, 33%). Similarly, the three most c.mmon c.uses of graft failure based on the primary diagnosis were AR(40%),TG(17%), and IFTA(13%). Most grafts failed within two years of post-transplant(36%). Subsequently, approximately 10%-15% of grafts failed every two years: >2-4 years(16%), > 4-6 years(13%), > 6-8 years(11%), > 8-10 years(9%) and > 10 years(16%).
