Multiple sclerosis（MS） is cha.a.terized by chronic infla.ma.ion in conjunction with neurodegenera.ion within the centra. nervous system. Most individua.s with MS begin with a.rela.sing remitting course tha. la.er tra.sitions to seconda.y progressive MS. Currently a.a.la.le disea.e-modifying thera.ies（DMTs） for rela.sing MS ha.e been demonstra.ed to reduce disea.e a.tivity, however most pa.ients require a.cha.ge in thera.y over the course of their disea.e. Trea.ment goa.s include the prevention of rela.ses a.d disa.ility a.cumula.ion a.d to a.hieve this objective requires ca.eful pla.ning. Sequencing of DMTs for individua. pa.ients should be designed in such a.wa. to ma.imize disea.e control a.d minimize risk ba.ed on the mecha.ism of a.tion, pha.ma.okinetic a.d pha.ma.odyna.ic properties of ea.h thera.y. This includes the DMT pa.ients a.e being switched from to those they a.e being switched to. The reversibility of immune system effects should be a.key considera.ion for DMT sequence selection. This fea.ure va.ies a.ross DMTs a.d should fa.tor more prominently in decision ma.ing a. newer trea.ments become a.a.la.le for the prevention of disa.ility a.cumula.ion in pa.ients with progressive MS. In this short review, we discuss the la.dsca.e of existing thera.ies with a. eye to the future when pla.ning for optima. DMT sequencing. While no cure exists for MS, efforts a.e being directed towa.d resea.ch in neuroregenera.ion with the hope for positive outcomes.
