C3G contributes to platelet activation and aggregation by regulating major signaling pathways

C3G is a GEF(guanine nucleotide exchange factor)for Rap GTPases,among which the isoform Rap1b is an essential protein in platelet *** transgenic mouse models with platelet-specific overexpression of C3G or mutant C3GΔCat,we have unveiled a new function of C3G in regulating the hemostatic function of platelets through its participation in the thrombin-PKCRap1b pathway.C3G also plays important roles in angiogenesis,tumor growth,and metastasis through its regulation of the platelet *** addition,C3G contributes to megakaryopoiesis and ***,we used a platelet-specific C3G-KO mouse model to further support the role of C3G in hemostasis.C3G-KO platelets showed a significant delay in platelet activation and aggregation as a consequence of the defective activation of Rap1,which resulted in decreased thrombus formation in ***,we explored the contribution of C3G-Rap1b to platelet signaling pathways triggered by thrombin,PMA or ADP,in the referenced transgenic mouse model,through the use of a battery of specific *** found that platelet C3G is phosphorylated at Tyr504 by a mechanism involving *** phosphorylation was shown to be positively regulated by ERKs through their inhibition of the tyrosine phosphatase ***,C3G participates in the ADP-P2Y12-PI3K-Rap1b pathway and is a mediator of thrombin-TXA2 ***,it inhibits the synthesis of TXA2 through cPLA2 *** together,our data reveal the critical role of C3G in the main pathways leading to platelet activation and aggregation through the regulation of Rap1b.

Process performance and quality attributes of temperature and step-down relative humidity controlled hot air drying of Panax notoginseng roots

The effects of temperature and step-down relative humidity controlled hot-air drying(THC-HAD)on the drying kinetics,energy efficiency and quality,i.e.,rehydration ratio(RR),color parameters(L*,a*,b*),total color difference(ΔE*),Panax notoginseng saponins(PNS)content,and ginsenosides content(R1,Rg1,Re,Rd,Rb1)of Panax notoginseng roots were *** drying time was significantly affected by the drying temperature followed by the relative humidity(RH)of the drying *** combination of drying conditions,i.e.,drying temperature of 50°C,relative humidity of 40%for 3 h and then continuous dehumidification from 40%to 8%allowed to shorten the drying time by 25%compared to drying at the same temperature and continuous *** longer was the drying time under constant high RH of drying air,the lower was the RR of dried *** step-down RH strategy contributed to the formation of a porous structure,enhancement of drying efficiency and quality ***,the ginsenosides content increased with the increase in temperature,while no obvious trend was recorded for ginsenoside *** contents of the ginsenoside R1,Rg1,Rb1 and PNS decreased with the increase in the drying time under constant high *** into account the drying time,energy consumption and quality attributes,drying at the temperature of 50°C,constant RH of 40%for 3 h and then step-down RH from 40%to 8%was proposed as the most favorable combination of drying conditions for dehydration of whole Panax notoginseng roots.

Little is known about the long-term survivors of acute arsenic intoxication. We present here a clinical case report of a man with chronic hepatitis b virus （HbV） infection who developed hepatocellular carcinoma four years after acute arsenic poisoning. HbsAg was detected in serum in 1990 when he voluntarily donated blood. In 1991, the patient suffered from severe psychological depression that led him to attempt suicide by massive ingestion of an arsenic-containing rodenticide. He survived with polyneuropathy and paralysis of the lower limbs, and has been wheelchair-bound since then. During participation in a follow-up study conducted among HbV carders, abdominal ultrasound detected a two-centimeter liver mass consistent with hepatocellular carcinoma. The tumor was confirmed by computed tomography （CT） and magnetic resonance image （MRI）. because of his significant comorbidity, the patient received palliative treatment with transarterial lipiodol chemoembolization （TACE） on three occasions （1996, 1997 and 1999）. At his most recent visit in lay 2005, the patient was asymptomatic, liver enzymes were normal and the tumor was in remission on ultrasound.

Peroxiredoxins （Prx） catalyse the reduction of hydrogen peroxide （H2O2） and, in association with catalases and other peroxidases, may participate in signal transduction by regulating intercel ular H2O2 concentration that in turn can control gene transcription and cel signaling. Using virus-induced-gene-silencing （VIGS）, 2-Cys Peroxiredoxin （2CysPrx） family and type-II Peroxiredoxin b （PrxI b） gene were silenced in Nicotiana benthamiana, to study the impact that the loss of function of each Prx would have in the antioxidant system under control （22℃） and severe heat stress conditions （48 ℃）. The results showed that both Prxs, although in different organel es, influence the regeneration of ascorbate to a significant extent, but with different purposes. 2CysPrx affects abscisic acid （AbA） biosynthesis through ascorbate, while PrxIIb does it probably;through the xanthophyl cycle. Moreover, 2CysPrx is key in H2O2 scavenging and in consequence in the regulation of AbA signal-ing downstream of reactive oxygen species and PrxIIb provides an important assistance for H2O2 peroxisome scavenges.

Mortality and rebleeding following variceal haemorrhage in liver cirrhosis and periportal fibrosis

AIMTo investigate mortality and rebleeding rate and identify associated risk factors at 6 wk and 5 d following acute variceal haemorrhage in patients with liver cirrhosis and schistosomal periportal fibrosis. METHODSThis is a prospective study conducted during the period from March to December 2014. Patients with portal hypertension presenting with acute variceal haemorrhage secondary to either liver cirrhosis (group A) or schistosomal periportal fibroses (group b) presenting within 24 h of the onset of the bleeding were enrolled in the study and followed for a period of 6 wk. Analysis of data was done by Microsoft Excel and comparison between groups was done by Statistical Package of Social Sciences version 20 to calculate means and find the levels of statistical differences and define the mortality rates, the P value of RESULTSA total of 94 patients were enrolled in the study. Thirty-two patients (34%) had liver cirrhosis (group A) and 62 (66%) patients had periportal fibrosis (group b). Mortality: The 6-wk and 5-d mortality were 53% and 16% respectively in group A compared to 10% and 0% in group b (P value P value P value P value P value P value P value CONCLUSIONThe 6-wk and 5-d mortality and rebleeding rate were significantly higher in patients with liver cirrhosis compared to patients with schistosomal periportal fibrosis.

Prevalence of respiratory syncytial virus infection among children hospitalized with acute lower respiratory tract infections in Southern India

bACKGROUND Respiratory syncytial virus(RSV) is a leading cause of lower respiratory infections among *** To investigate the proportion of RSV and non-RSV respiratory viral infections among hospitalized children ≤ 5 *** Hospitalized children aged between August 2011-August 2013, were *** were defined as laboratory-confirmed RSV and non-RSV respiratory viruses by direct fluorescence assay from the nasopharyngeal *** Of 383 1-59 mo old children hospitalized with an acute lower respiratory infection, 33.9%(130/383) had evidence of viral infection, and RSV was detected in 24.5%(94/383). Co-infections with RSV and other respiratory viruses(influenza A or b, adenovirus, para influenza 1, 2 or 3) were seen in children 5.5%(21/383). Over 90% of the RSV-positive children were under 2 years of age. RSV was detected throughout the year with peaks seen after the monsoon *** hospitalized with RSV infection were more likely to have been exposed to a shorter duration of breastfeeding of less than 3 mo. RSV positive children had a shorter hospital stay, although there were significant complications requiring intensive care. Use of antibiotics was high among those with RSV and non-RSV viral *** Our study provides evidence of a high proportion of RSV and other virusassociated ALRI among hospitalized children in India. RSV infection was associated with fewer days of hospital stay compared to other causes of lower respiratory infections. A high level of antibiotic use was seen among all respiratory virus-associated hospitalizations. These results suggest the need for implementing routine diagnostics for respiratory pathogens in order to minimize the use of unnecessary antibiotics and plan prevention strategies among pediatric populations.

Study of corneal biomechanical properties in patients with childhood glaucoma

AIM:To study of corneal biomechanical properties and intraocular pressure(IOP)measured with Corvis Scheimpflug Technology(ST)in patients with childhood glaucoma(CG).METHODS:Cross-sectional study in which 89 eyes were included 56 of them with *** one eye per patient was *** following variables were obtained from the clinical history and the ophthalmological examination:age,sex,IOP,number of surgeries,and the cup/disc ratio(CDR).The following parameters were recorded using Corvis ST:corrected by biomechanics IOP(b IOP),not corrected IOP(nct IOP),central corneal thickness(CCT),maximum concavity[radius,peak distance(PD)and deformation amplitude],applanation 1 and 2(length and velocity).The mean age was 23±14.55 and 33±19.5 years old for the control group and CG group,*** 36 were males and 53 were *** the CG group,7 patients were controlled only with medical *** had at least one previous goniotomy,19 had at least one trabeculectomy,and 11 had an Ahmed ***:A significant and positive intraclass correlation coefficient was found between Goldman IOP and the IOP measured by Corvis in both *** differences were found between the IOP measured with Corvis and Goldman using a student *** biomechanical parameters,there were differences in the applanation length 2(A-L2),in the applanation velocity 2(A-V2)and in the *** sex,only the applanation length 1(A-L1)was found to be different in control group.A positive and significant Pearson correlation was found between CDR and the ***:Corneal biomechanical properties have shown differences between CG and healthy subjects and also between men and women.
