Successful double fertilization and subsequent seed development in flowering plants requires the delivery of two sperm cells, transported by a pollen tube, into the embryo sac of an ovule. The embryo sac cells tightly control synergid cell death, and as a result the polyspermy block. Arabinogalactan proteins are highly glycosylated proteins thought to be involved in several steps of the reproductive process. We show that JAGGER, Arabinogalactan Protein 4, is an important molecule necessary to prevent the growth of multiple pollen tubes into one embryo sac in Arabidopsis thaliana. In jagger, an AGP4 knockout mutant, the pistils show impaired pollen tube blockage as a consequence of the survival of the persistent synergid. JAGGER seems to be involved in the signaling pathway that leads to a blockage of pollen tube attraction. Our results shed light on the mechanism responsible for preventing polyspermy in Arabidopsis and for safe- guarding successful fertilization of all ovules in one pistil, ensuring seed set and the next generation.

目的哮喘-慢性阻塞性肺疾病肺重叠症(Asthma-chronie obst ructive pulmonary disease overlap,A-CO)患者的生活质量较低,肺功能较差,病情经常加重。由于暴露于污染环境会加重其症状,增加发病率和病死率,环境管理对其至关重要。旨在通过系统性综述,寻找证据,以指导制订护理干预措施,促进ACO患者对空气质量的自我管理。方法2023年3月 ,系统检索CINAHL、Medline、Academie Search Complete .Scopus、WebofScience、Cochrane图书馆、乔安娜布里格斯研究所(JoannaBriggsInstitute,JBI)循证实践数据库,以及GoogleScholar。对纳人研究进行数据提取和综合,并对护理干预措施进行分类。采用JBI研究质量评估工具进行方法学质量评估。结果共纳入5篇来自不同国家的研究。促进ACO患者自我监控空气质量的干预措施包括:警觉性干预措施(医务人员定期巡诊、进行症状评估),监测干预措施(监测室内外诱发因素) ,教育干预措施(空气质量警报、避免接触过敏原),以及相关政策的影响,如无烟政策和改善空气质量的综合战略。尽管这些研究报告了不同的干预措施,方法质量也存在差异,但这些干预措施显示了预防患者病情恶化、尽量减少急诊就诊和住院治疗的潜力。这些关键点可作为护理干预措施的关键组成部分。结论有必要采取全面措施,让护士参与促进患者自我监控空气质量的多学科团队.提高认识、制订和实施护理干预,从而使ACO患者受益。同时,需要进--步开展研究,以扩大证据基础并完善这些干预措施。

Herein,the synthesis and characterization methods of natural deep eutectic solvents based on monoterpenoids have been presented.Low viscous fluids with suitable physicochemical properties are produced.The materials are non-toxic,biodegradable,and cost-effective.Thus,they can be used to develop sustainable solvents for various processes and can find their applications in various fields.A theoretical study based on quantum chemistry and classical molecular dynamics is used for the nanoscopic characterization of structure,dynamics,and hydrogen bonding.The reported results help analyze the properties of this new family of solvents.The required information for developing structure–property relationships for proper solvent design to form a sustainable chemistry framework is obtained.

The broke of a tailings dam from Vale S.A. destroyed a gigantic area killing 270 persons in Brumadinho/Brazil. Organizing activities after a mass disaster is a complex process that requires the involvement of many people and resources in the laboratory. To DNA identification of the victims, a daunting work had to be made by DNA laboratory staff in a collaborative effort with many partners. Efforts to implement good practice guidelines in Disaster Victim Identification (DVI) have revealed several important aspects that need to change in the forensic DNA laboratory. This article highlights the challenges of implementing DVI best practice guidelines in resource-poor settings, but with professionals from different sectors engaged in the same goal of briefly identifying victims and helping families and society.

Chronic wounds have become one of the most important issues for healthcare systems and are a leading cause of death worldwide.Wound dressings are necessary to facilitate wound treatment.Engineering wound dressings may substantially reduce healing time,reduce the risk of recurrent infections,and reduce the disability and costs associated.In the path of engineering of an ideal wound dressing,hydrogels have played a leading role.Hydrogels are 3D hydrophilic polymeric structures that can provide a protective barrier,mimic the native extracellular matrix(ECM),and provide a humid environment.Due to their advantages,hydrogels(with different architectural,physical,mechanical,and biological properties)have been extensively explored as wound dressing platforms.Here we describe recent studies on hydrogels for wound healing applications with a strong focus on the interplay between the fabrication method used and the architectural,mechanical,and biological performance achieved.Moreover,we review different categories of additives which can enhance wound regeneration using 3D hydrogel dressings.Hydrogel engineering for wound healing applications promises the generation of smart solutions to solve this pressing problem,enabling key functionalities such as bacterial growth inhibition,enhanced re-epithelialization,vascularization,improved recovery of the tissue functionality,and overall,accelerated and effective wound healing.

This research investigates the soil conditioning effect of sugar cane fields with biochar produced from sugarcane bagasse.Its influence on the physicochemical and microbiological conditions of the soil and the agronomic performance of sugarcane seedlings was verified.To this end,the biochar from sugarcane bagasse was produced by pyrolysis in a double drum oven and mixed with soil at rates of 0,1%,3%,and 5%(in weight).The experiment was installed in a greenhouse with two evaluation periods(30 days and 60 days)and five replicates.Each pot received a pre-sprouted cane seedling,and the analysis of colony-forming units(bacteria and fungi),physicochemical characteristics of the soil,and growth and development of sugarcane were performed per pot.The biochar showed a high fixed carbon content(72%)and a specific surface area estimated by the adsorption of methylene blue of 50 m2 g−1.Furthermore,its structure is porous and contains important nutrients(e.g.,CaO,K2O,and P2O5).In the periods evaluated,the applications of the 3%and 5%(in weight)rates of biochar reduced most of the soil fertility parameters.However,the chemical analyses indicated that the sugarcane field soil collected already possessed high fertility.The microbiota was influenced,but only the application of the 3%(in weight)rate at 60 days after planting showed a significant positive effect on the number of bacteria forming units,with an increase of approximately 385%.On the other hand,no significant positive effect on sugarcane development was found.Therefore,biochar application in high-fertility sugarcane plantation soil did not result in gains for sugarcane development in the periods evaluated.

Mindfulness interventions during pregnancy:A narrative review

Pregnancy is a period of major transformations in a woman’s life;increased stress,and mood and sleep disorders are frequent.This review evaluates mindfulness interventions during pregnancy and their ability to help manage stress,anxiety,depression,emotional regulation,level of mindfulness and sleep quality.A search of English language scientific literature relevant to mindfulness interventions for pregnant women was conducted using PubMed,Scopus and Web of Science,without restriction on publication date.Inclusion criteria were randomized clinical trials with pregnant women,using mindfulness as an intervention for at least three weeks,in one of our main areas of interest,and using only validated scales to measure outcomes.Two hundred and thirty studies were identified in our searches of research databases,and thirteen were included in our analysis.We found a large diversity of mindfulness programs,heterogeneity among the instruments used to evaluate outcomes,and inconsistency in the gestational periods used in the studies.Mindfulness interventions were beneficial for stress,anxiety and depression.Mindfulness was also effective when applied in pregnant women with a history of depression or experiencing depression.Considering emotional regulation and the level of mindfulness,there were signs of improvement,but more studies are needed.None of the studies evaluated sleep quality.Our review provides information about current mindfulness programs,an overview of the effects of mindfulness interventions,a description of the measurements used so far,and recommendations for developing highquality mindfulness protocols for pregnant women.

Posterior corneal astigmatism modifications after cataract surgery and its role on total corneal astigmatism

Background:In recent years posterior corneal astigmatism and its effect on total corneal astigmatism has been studied,with research showing that this can impact total astigmatism.This study aims to ascertain if there is significant change in the posterior corneal astigmatism after cataract surgery and its impact on the total astigmatism.Methods:Analysis of 76 eyes that underwent cataract surgery with monofocal intraocular lens implantation.Corneal topography was performed with Pentacam(OCULUS®)pre-and post-operatively.Total corneal astigmatism was calculated with the algorithm of vergence tracing.We compared preoperative and postoperative changes in the magnitude and axis differences of anterior corneal curvature astigmatism,posterior corneal curvature astigmatism and the calculated total corneal astigmatism.We calculated the correlation between the total preoperative astigmatism and the difference between total corneal astigmatism and anterior corneal astigmatism.Results:The mean preoperative and postoperative posterior astigmatism was 0.31±0.02 D,showing no significant differences before and after surgery(P=0.989).Statistically significant differences between the calculated total corneal astigmatism and anterior corneal astigmatism were registered preoperatively and postoperatively in the with-the-rule anterior(WTR)corneal astigmatism(P=0.004,P<0.0001);against-the-rule(ATR)anterior corneal astigmatism(P<0.0001,P<0.0001)and in the oblique(P=0.026,P=0.019)subgroups.The posterior corneal astigmatism and the total corneal astigmatism correlated positively with the differences between the total corneal and anterior corneal astigmatism(R=0.378,P=0.001).Conclusions:There were statistically significant differences between the magnitude of the total astigmatism and anterior corneal astigmatism,underlining the impact of posterior corneal astigmatism.A positive correlation between the preoperative posterior astigmatism and the difference between the total corneal and the anterior corneal astigmatism suggests a specially r

Firmness is one of the most important fruit quality traits in strawberries.The postharvest shelf life of this soft fruit is highly limited by the loss of firmness,where cell wall disassembly plays an important role.Previous studies demonstrated that the polygalacturonase FaPG1 has a key role in remodelling pectins during strawberry softening.In this study,FaPG1 knockout strawberry plants have been generated using the CRISPR/Cas9 system delivered via Agrobacterium tumefaciens.Ten independent lines,cv.“Chandler”,were obtained,and all of them were successfully edited as determined by PCR amplification and T7 endonuclease assay.The targeted mutagenesis insertion and deletion rates were analyzed using targeted deep sequencing.The percentage of edited sequences varied from 47%up to almost 100%,being higher than 95%for seven of the selected lines.Phenotypic analyses showed that 7 out of the eight lines analyzed produced fruits significantly firmer than the control,ranging from 33 to 70%increase in firmness.There was a positive relationship between the degree of FaPG1 editing and the rise in fruit firmness.Minor changes were observed in other fruit quality traits,such as colour,soluble solids,titratable acidity or anthocyanin content.Edited fruits showed a reduced softening rate during postharvest,displayed a reduced transpirational water loss,and were less damaged by Botrytis cinerea inoculation.The analysis of four potential off-target sites revealed no mutation events.In conclusion,editing the FaPG1 gene using the CRISPR/Cas9 system is an efficient method for improving strawberry fruit firmness and shelf life.

Performance of Geometric Multigrid Method for Two-Dimensional Burgers’Equations with Non-Orthogonal,Structured Curvilinear Grids

This paper seeks to develop an efficient multigrid algorithm for solving the Burgers problem with the use of non-orthogonal structured curvilinear grids in L-shaped geometry.For this,the differential equations were discretized by Finite Volume Method(FVM)with second-order approximation scheme and deferred correction.Moreover,the algebraic method and the differential method were used to generate the non-orthogonal structured curvilinear grids.Furthermore,the influence of some parameters of geometric multigrid method,as well as lexicographical Gauss–Seidel(Lex-GS),η-line Gauss–Seidel(η-line-GS),Modified Strongly Implicit(MSI)and modified incomplete LU decomposition(MILU)solvers on the Central Processing Unit(CPU)time was investigated.Therefore,several parameters of multigrid method and solvers were tested for the problem,with the use of nonorthogonal structured curvilinear grids and multigrid method,resulting in an algorithm with the combination that achieved the best results and CPU time.The geometric multigrid method with Full Approximation Scheme(FAS),V-cycle and standard coarsening ratio for this problem were utilized.This article shows how to calculate the coordinates transformation metrics in the coarser grids.Results show that the MSI and MILU solvers are the most efficient.Moreover,theMSI solver is faster thanMILU for both grids generators;and the solutions are more accurate for the Burgers problem with grids generated using elliptic equations.
