Design and preparation of a new multi-targeted drug delivery system using multifunctional nanoparticles for co-delivery of siRNA and paclitaxel

Drug resistance is a great challenge in cancer therapy using chemotherapeutic agents. Administration of these drugs with siRNA is an efficacious strategy in this battle. Here, the present study tried to incorporate siRNA and paclitaxel(PTX) simultaneously into a novel nanocarrier. The selectivity of carrier to target cancer tissues was optimized through conjugation of folic acid(FA) and glucose(Glu) onto its surface. The structure of nanocarrier was formed from ternary magnetic copolymers based on FeCopolyethyleneimine(FeCo-PEI) nanoparticles and polylactic acid-polyethylene glycol(PLA-PEG) gene delivery system. Biocompatibility of FeCo-PEI-PLA-PEG-FA(NPsA), FeCo-PEI-PLA-PEG-Glu(NPsB) and FeCo-PEI-PLA-PEG-FA/Glu(NPsAB) nanoparticles and also influence of PTX-loaded nanoparticles on in vitro cytotoxicity were examined using MTT assay. Besides, siRNA-FAM internalization was investigated by fluorescence microscopy. The results showed the blank nanoparticles were significantly less cytotoxic at various concentrations. Meanwhile, siRNA-FAM/PTX encapsulated nanoparticles exhibited significant anticancer activity against MCF-7 and BT-474 cell lines. NPsAB/siRNA/PTX nanoparticles showed greater effects on MCF-7 and BT-474 cells viability than NPsA/siRNA/PTX and NPsB/siRNA/***, they induced significantly higher anticancer effects on cancer cells compared with NPsA/siRNA/PTX and NPsB/siRNA/PTX due to their multi-targeted properties using FA and Glu. We concluded that NPsAB nanoparticles have a great potential for co-delivery of both drugs and genes for use in gene therapy and chemotherapy.

Combining Knowledge-and Data-Driven Fuzzy Approach to Evaluate Shelf-Life of Various Seafood Products

Due to the complexity of the deterioration process of seafood products,relying on one indicator is not adequate to determine the quality of such ***,shelf-life was estimated based on various indicators complicating the decision-making *** Support Systems are considered as a good *** current study aims to establish a simple and novel fuzzy model based on a combination of knowledge-and data-driven approaches to define a fuzzy quality deterioration index(FQDI)in various seafood products(rainbow trout,threadfin bream,and white shrimp samples)during cold *** volatile basic nitrogen(TVB-N)and psychrotrophic microorganisms counts(PMCs)were determined based on traditional *** sensory analysis was performed by a data-driven fuzzy ***,the shelf-life of the rainbow trout fillet was estimated to be 8 d,based on all the freshness ***,the shelf-life of the Japanese threadfin bream fillet was 5-7 d according to the microbial and chemical parameters,*** time for shrimp samples was 6-8 d using sensory score and TVB-N *** results of data-driven fuzzy approach showed all of the quality properties were considered as the'Important'-'Very Important'(defuzzification score>75).The TVB-N and PMCs were the most and weakest freshness quality properties(defuzzified-values:84.64 and 78.75,respectively).Based on FQDI,the shelf-life of the rainbow trout,Japanese threadfin bream,and shrimp samples were estimated to be 8,5,and 7 d,*** method was able to successfully provide a comprehensive deterioration index for evaluating the seafood *** a total index can be considered as a comprehensive output(y variable)to predict seafood freshness by rapid and nondestructive method.

Validity and Reliability of an Information-Motivation-Skill-Behavior Questionnaire: A Gender-Sensitive HIV Survey

Background: Despite the essence of HIV-related surveys, their precision in measurement of intended constructs has been an area of controversy. Considering the increasing trend of sexual transmission of HIV among women, standard gender-sensitive tools can yield pivotal information for researchers and policy-makers in Iran. Aim: To evaluate the reliability and validity of a questionnaire (IMSB) based on the Information-Motivation-Behavior model of HIV-related behavior prediction. Methods: The IMSB survey constituted of five main domains: HIV-related knowledge, attitudes toward people living with HIV, motivation to practice safe-sex, skills to practice safe-sex, and HIV-related behaviors. Completed questionnaires (n = 189) from pilot phases of three projects were accessed. We assessed face and content validity through expert consultation and respondent feed-backs for the primary pool of items. Discriminant validity was measured utilizing non-parametric tests. Cronbach’s alpha was calculated for each domain to assess internal consistency, and paired t-tests were used to measure stability over time. Results: The primary pool of items was reduced in the process of validation. The discriminant validity was appraised to be favorable since the survey can differentiate between age groups, women with different levels of education and marital status. The survey was reliable in 4 domains;but a Cronbach’s alpha of 0.6 was reported for the fifth domain (HIV-related behaviors). Younger women had higher knowledge and motivation scores;single women were more motivated to practice safe-sex and divorced/widowed women had lower motivation and skills scores. Conclusion: This questionnaire can be used as a valid and reliable tool to measure the IMB constructs. The lower reliability of the “behavior” domain is probably rooted in societal norms and cultural definitions.
