印度家族中先天性白内障的突变分析:sNPs和CRYBB2基因中新的致病性等位基因的鉴定

PURPOsE. to study some functional candidate genes in cataract families of Indi an descent. MEtHODs. Nine Indian families, clinically documented to have congeni tal/childhood cataracts, were screened for mutations in candidate genes such as CRYG (A→D), CRYBB2, and GJA8 by PCR analyses and sequencing. Genomic DNA sample s of either probands or any representative affected member of each family were P CR amplified and sequenced commercially. Documentation of single nucleotide poly morphisms (sNPs) and candidate mutations was done through BLAst sEARCH(http://ww ***/ blast/Blast.- cgi?). REsULts. several single nucleotide polymorphisms in CRYG, CRYBB2, and G JA8 genes were observed. Because they do not co-segregate with the phenotype, t hey were excluded as candidates for the cataract formation in these patients. However, a substitution (W151C in exon 6 of CRYBB2) was identi fied as.t.e most likely caus.t.ve mutation underlying the phenotype of central n uclear cataract in all affected members of family C176. Protein structural inter pretations demonstrated that no major structural alterations could be predicted and that even the hydrogen bonds.t. the neighboring Leu166 were unchanged. surpr isingly, hydropathy analysis of the mutant βB2-crystallin featuring the amino acids at pos.t.on 147 to 155, further increased the hydrophobicity, which might impair the solubility of the mutant protein. Finally, the Cys residue at pos.t.o n 151 might possibly be involved in intramolecular disulphide bridges with other cysteines during translation, possibly leading to dramatic structural changes. CONCLUsIONs. Exon 6 of CRYBB2 appears.t. be a critical region susceptible for mu tations leading to lens opacity.

Analysis and behaviour of FRP-confined short concrete columns subjected to eccentric loading

Fibre-reinforced polymer (FRP) compos.t.s were widely utilized in civil engineering structures as.t.e retrofit of reinforced concrete (RC) columns. to design FRP jackets safely and economically, the behaviour of such columns should be predicted first. this paper is concerned with the analysis and behaviour of FRP-confined RC circular and rectangular short col- umns subjected to eccentric loading. A simple design-oriented stress-strain model for FRP-confined concrete in a section analysis was first proposed. the accuracy was.t.en proved by two test data. Following that, a parametric study including amount of FRP confinement, FRP strain capacity, unconfined concrete strength and shape of column section is provided. some conclusions were obtained at the end of the paper. the work here will provide a comprehensive understanding of the behaviour of FRP-confined concrete columns. the simplicity of the model also enables a simple equivalent stress block to be developed for direct use in practical design.

Free vibration and transient response of double curved beams connected by intermediate straight beams

this paper investigates.t.e free vibration and transient response of interconnected structures including double curved beams and intermediate straight beams,which are all constructed from functionally graded porous(FGP) materials. the strain potential and kinetic energies of each beam along with the work done by the external force are calculated. Additionally, a higher-order beam element is introduced to derive stiffness and mass matrices, along with the force vector. the curved and straight beams are discretized, and their assembled stiffness, mass matrices, and force vectors, are *** conditions at the joints are used to derive the total matrices of the entire structure. subsequently, the natural frequencies and transient response of the system are determined. the accuracy of the mathematical model and the self-developed computer program is validated through the comparison of the obtained results with those of the existing literature and commercial software ANsYs, demonstrating excellent ***, a comprehensive study is conducted to investigate the effects of various parameters on the free vibration and transient response of the considered structure.

Influence of sputtering Power on the structure and Electrical Properties of Bi_2Fe_4O_9 thin Films

Bismuth ferrite（Bi_2Fe_4O_9） thin films were grown on p-type si（100） substrate by radio-frequency magnetron sputtering at 873 K. X-ray diffraction, field emission scanning electron microscopy and Raman spectroscopy studies revealed that the grown films have single-phase polycrystalline nature and are crystallized in orthorhombic structure. the grain size of the grown thin films was found to increase（56–130 nm） with sputtering power. Atomic force microscopy images clearly illustrated that the grown thin films have smooth surface. Energy-dispersive X-ray analysis revealed the presence of Bi, Fe and O elements with desired ratio and also the absence of impurities in the grown films. Analysis of ferroelectric hysteresis loops revealed that the remanent polarization and coercive field increase with the increase in sputtering power. Vicker＇s hardness analysis showed that the hardness of films strongly depends on the grain size and film thickness, which are mainly determined by the sputtering power. the above observations revealed that Bi_2Fe_4O_9 thin film depos.t.d at higher sputtering power has good crystallinity and shows better electrical properties.

Experimental study of friction dissipators for seismic protection of building structures

this paper presents.t.e results from unidirectional shaking table tests of two reduced scale steel models of a building frame, with one and two floors, respectively. these frames incorporate friction dissipators at every floor. the inputs are sine-dwells and artificial and registered earthquakes. this study is part of a larger research project aiming to assess.t.e seismic efficiency of friction dissipators by means of an integrated numerical and experimental approach. Inside this framework, the main objectives of these experiments are to：（i） collect a wide range of results.t. calibrate a numerical model derived within the project, （ii） clarify some of the most controversial issues about friction dissipators （including behavior for inputs containing pulses, capacity to reduce resonance peaks, introduction of high frequencies in the response, and self- generated eccentricities）, （iii） better understand their dynamic behavior, （iv） provide insight on the feasibility and reliability of using simple friction dissipators for seismic protection of building structures and （v） characterize the hysteretic behavior of these devices. Most of these objectives are s.t.sfactorily reached and relevant conclusions are stated.

Influence of austenization temperature on the erosion behavior of austempered ductile irons

the erosion behavior of austempered ductile irons austenized at different temperatures was studied. the results indicate that the erosion rate well correlates with the mechanical properties. At high impact angles, increasing ductility and mechanical energy dens.t. results in decreasing erosion rate, whereas increasing hardness reduces.t.e erosion rate at low impact angles. 2008 Univers.t. of science and technology Beijing. the experiments with solar, atmospheric, reactor and accelerator neutrinos have provided compelling evidences for oscillations of neutrinos caused by nonzero neutrino masses and neutrino mixing. the data imply the existence of 3-neutrino mixing in vacuum. We review the theory of neutrino oscillations, the phenomenology of neutrino mixing, the problem of the nature - Dirac or Majorana, of massive neutrinos, the issue of CP violation in the lepton sector, and the current data on the neutrino masses and mixing parameters. the open questions and the main goals of future research in the field of neutrino mixing and oscillations are outlined.
