【Goal and Background】 Risk assessment of environmental contaminants relies on scientific evidence from animal toxicity testing and epidemiological studies which can be time consuming,costly and provides limited mechanistic *** throughput in vitro methods are needed to accommodate the need for testing large number of chemicals known or suspected to pose risk to human *** Environmental Protecting Agency（EPA）initiated a Toxicity Forcaster（ToxCast） program in 2007,aiming to identify a battery of such predictive in vitro high throughput screening methods to reduce and eventually replace animal testing for better human health risk *** to its unique advantage of non-invasive and real time cell analysis（RTCA）,impedance based screening methods were incorporated and utilized as a key component of the ToxCast program to evaluate general toxicity,nanoparticle toxicity,and potential endocrine disruptor characteristics of testing substances.【Materials and Methods】 Human cell lines A549,T-47 D and 22 Rvl were grown in E-Plates under optimized growth and assay conditions and were monitored on xCELLigence RTCA MP or HT *** all assays,the substances were tested with eight concentrations,and with 2 to 4 replicates.【Results】 Prior to being incorporated as part of the ToxCast program,all assays were evaluated and displayed good reliability,reproducibility,sensitivity and ***,the rich real time kinetic response profiles provided additional information on the onset and rate of effects,the potential mechanisms and adverse outcome pathways,and side effects including *** 2007,320 pesticides and 65 nanoparticles were tested in human lung cancer cell line A549 for general cytotoxicity;more than 2000 chemicals were tested in human breast cancer cell line T-47 D for evaluation of estrogenic activity;and 800 chemicals were tested in human prostate cancer cell line 22 Rvl for evaluation of androgen agonist and antagonist act

Occlusion with a Vascular Plug of a Congenital Circumflex Coronary Artery—Right Ventricle Fistula with Coronary Steal Phenomenon

Background: Coronary fistulas are abnormal communications with a worldwide prevalence of 0.002% in the general population. Aim: The aim of this study is to present a case of a congenital coronary artery fistula presented with coronary steal and describe this phenomenon. Methods: A systematic search was conducted to explore the relationship between the coronary steal phenomenon and coronary artery diseases using the PubMed database. Case Presentation: A seven-month-old female patient, was admitted to the hospital presenting diaphoresis on feeding and failure to thrive. Physical examination denotes continuous murmur in the fourth intercostal space left parasternal border (IV/VI) crescendo-decrescendo, wide and permanent unfolding of S2 (S1-A2-P2). An echocardiogram shows enlargement of right cavities, dilatation of the left coronary artery, circumflex artery connected to the right ventricle, and pulmonary artery systolic pressure of 45 mmHg. Tomography angiography demonstrated dilatation of the circumflex artery and a tortuous trajectory towards the posterior wall of the right ventricle, suggesting a coronary fistula. Results: The findings were corroborated by catheterization and a vascular plug (Amplatzer AVP II) was successfully placed. During follow-up complete obliteration was observed, with adequate weight gain and no associated symptoms. Conclusion: This case illustrates a distinctive clinical manifestation of congenital coronary artery fistula associated with the coronary steal phenomenon, establishing a new paradigm for the early diagnosis and management of this condition in the pediatric population.

Background & Aims: In normal intestine, cyclic nucleotides(adenosine 3,5-cyclic monophosphate cAMP, guanosine 3,5-cyclic monophosphate) and Ca2+ inhibit neutral sodium absorption. In contrast, in the jejunum of a knock out mousemodel of cystic fibrosis (CF), agents that elevate intracellular cAMP levels did not inhibit neutral sodium absorption, suggesting that the antiabsorptive effect of cAMP is dependent on the cystic fibrosis transmembrane conductance regulator (CFTR).The aim of the present study was to determine if a prostaglandin E1 analogue, which causes elevation of intracellular cAMP and Ca2+ levels, inhibits neutral sodium absorption in patients with CF in vivo. Methods: Electrolyte and water absorption/secretion was measured during steady state perfusion of the jejunum with a balanced electrolyte solution. Patients with CF and healthy subjects were studied under basal conditions and during intraluminal infusion of a prostaglandin E1 analogue (misoprostol). Results: The rate of neutral sodium absorption in the basal state was similar in healthy subjects and patients with CF. Prostaglandin infusion markedly reduced neutral sodium absorption in both healthy subjects and patients with CF. Prostaglandin caused high rates of electrolyte and water secretion in healthy subjects but only trivial rates of secretion in patients with ***: CFTR mutations causing CF in humans do not prevent prostaglandin E1 inhibition of neutral sodium absorption, even though these mutations produce a severe defect in prostaglandin-stimulated electrolyte *** findings suggest that an intact antiabsorptive response to either cAMP or Ca2+ may contribute to the relatively low level of intestinal disease in patients with CF.

目的探讨谷氨酰胺对肝脏移植后应激蛋白表达的影响.方法 A组10例为正常肝脏;B组10例肝脏移植后不用丙氨酰谷氨酰胺二肽(Ala-Gln);C组8例肝脏移植后用Ala-Gln.观察标本的病理学改变和患者的营养状况,用免疫组织化学及原位杂交方法检测HSP70、HO-1蛋白和mRNA表达水平.结果 B组和C组均未见免疫排斥反应,营养状况尚可;HSP70、HO-1蛋白及mRNA表达水平移植肝脏均升高,且应用Ala-Gln后更高.结论谷氨酰胺能增加供肝细胞应激蛋白基因转录和表达,加强肝细胞自我保护作用.

An easy,accurate and efficient procedure to create forest fire risk maps using the SEXTANTE plugin Modeler

To prevent, detect, and protect against forest fires, forest personnel need to define rules for determining forest fire risk. In Portugal, all municipalities must annually produce forest fire risk （FFR） maps. To produce more reliable FFR maps more easily, we developed an open source model using the Modeler plugin of SEXTANTE in the program QGIS version 2.0 Dufour. The model provides all the maps involved in the FFR model （susceptibility map, hazard map, vulnerability map, economic value map, and potential loss map） and was produced according to Portuguese Forest Authority＇s （AFN, Autoridade Florestal Nacional） rules for determining the FFR. This model was tested for the Portuguese municipality santa Maria da Feira, where 40 % of the total municipality area falls in the category ＂very high＂ or ＂high＂ fire risk. The ＂very high＂ fire risk area is mainly classified as broad-leaved forest and has the steepest slopes （〉15 %）. The distance of burned areas to roads was also analyzed; the proportion of burned areas increased with increasing distance to the main roads. In addition, 92.6 % of the ＂high＂ and ＂very high＂ risk zones were located in areas with lower elevation. These results confirmed that forest fire is strongly influenced not only by environmental factors but also by anthropogenic factors. The procedure implemented here was compared with our open source application already available in QGIS and also to the same procedure implemented in GIS pro- prietary software. Although the results were obviously the same, the model developed here presents several advan- tages over the other two approaches. Besides being faster, it is easy to change the model parameters according to user needs （i.e., to the rules of different countries）, and can be modified and adapted to other variables and other areas to create risk maps for different natural phenomena （e.g., floods, earthquakes, landslides）. When α= 0 we simply denote it by Me. In this paper we prove that if Ф and ψare two Young functions, there exists a third Young function θ such that the composition Mα,ψ o MФ is pointwise equivalent to Mα,θ. As a consequence we prove that for some Young functions θ, if Mα,θf 〈∞a.e. and δ ∈（0,1） then （Mα,θf）δ is an A1-weight.
