Norway spruce(Picea abies) laccases:Characterization of a laccase in a lignin-forming tissue culture

Secondarily thickened cell walls of water-conducting vessels and tracheids and support-giving sclerenchyma cells contain lignin that makes the cell walls water impermeable and strong. To what extent laccases and peroxidases contribute to lignin biosynthesis in muro is under active evaluation. We performed an in silico study of Norway spruce （Picea abies （L.） Karst.） laccases utilizing available genomic data. As many as 292 laccase encoding sequences （genes, gene fragments, and pseudogenes） were detected in the spruce genome. Out of the 112 genes annotated as laccases, 79 are expressed at some level. We isolated five fun-length laccase cDNAs from developing xylem and an extracellular lignin-forming cell culture of spruce. In addition, we purified and biochemically characterized one culture medium laccase from the lignin-forming cell culture. This laccase has an acidic pH optimum （pH 3.8-4.2） for coniferyl alcohol oxidation. It has a high affinity to coniferyl alcohol with an apparent Km value of 3.5μM; however, the laccase has a lower catalytic efficiency （Vmax/Km） for coniferyl alcohol oxidation compared with some purified culture medium peroxidases. The properties are discussed in the context of the information already known about laccases/coniferyl alcohol oxidases of coniferous plants.

Background: Spinal dysraphism represents a wide spectrum of congenital abnormalities of the spine. Myelomeningocele is considered the most common malformation and the most common we saw in our community, with its morbidity problems seen commonly in the postoperative period. ASQ-3TM Scores are the ages and stages questionnaire, third edition, and represent a tool to assess the development progress, especially in toddlers. Objectives: Evaluation of neurodevelopmental outcome among Sudanese toddlers with spinal dysraphism after surgical closure with or without a VP shunt using ASQ-3TM Scores. Methodology: This is a retrospective hospital-based study of 84 patients who underwent myelomeningocele repair at the National Center for Neurological Sciences (NCNS) during the period from 2017 up to 2019. Data were collected through a constructed questionnaire, including ASQ-3TM Scores. Data were processed and analyzed using the Statistical Package for Social Science (SPSS) computer program. Version 25. Results: 84 patients were included in this study;all patients were diagnosed with spinal dysraphism. Out of them, 51 (60.7%) were 2 years old, 33 (39.3%) were 3 years old, 45 (53.6%) were male, 45 (53.6%) of patients mothers attended ANC irregularly, and 54 (64.3%) their mothers didn’t receive folate supplements. 44 (52.3%) of patients underwent MMC repair only, while 40 (47.7%) underwent MMC repair and VP shunt. The commonest postoperative complication was infection, reported in 12 (14.3%) of patients, followed by VP shunt revision in 9 (10.7%) of patients. Neurological assessment showed that the majority of patients need further assessment with a professional, 57 (67.9%) of children don’t walk, run, or climb like other toddlers as their parent’s state;also, half of patients (42, 50%) had medical problems, and 27 (32.1%) of their parent’s state that they do not talk like other toddlers their age. Genetic susceptibility to metabolic associated fatty liver disease(MAFLD)is complex and poorly *** characterization of the genetic background of hepatic fat content would provide insights into disease etiology and causality of risk *** performed genome-wide association study(GWAS)on two noninvasive definitions of hepatic fat content:magnetic resonance imaging proton density fat fraction(MRI-PDFF)in 16,050 participants and fatty liver index(FLI)in 388,701 participants from the United Kingdom(UK)Biobank(UKBB).Heritability,genetic overlap,and similarity between hepatic fat content phenotypes were analyzed,and replicated in 10,398 participants from the University Medical Center Groningen(UMCG)Genetics Lifelines Initiative(UGLI).Meta-analysis of GWASs of MRI-PDFF in UKBB revealed five statistically significant loci,including two novel genomic loci harboring CREB3L1(rs72910057-T,P=5.40E−09)and GCM1(rs1491489378-T,P=3.16E−09),respectively,as well as three previously reported loci:PNPLA3,TM6SF2,and *** of FLI in UKBB identified 196 genome-wide significant loci,of which 49 were replicated in UGLI,with top signals in ZPR1(P=3.35E−13)and FTO(P=2.11E−09).Statistically significant genetic correlation(rg)between MRI-PDFF(UKBB)and FLI(UGLI)GWAS results was found(rg=0.5276,P=1.45E−03).Novel MRI-PDFF genetic signals(CREB3L1 and GCM1)were replicated in the FLI *** identified two novel genes for MRI-PDFF and 49 replicable loci for *** a difference in hepatic fat content assessment between MRI-PDFF and FLI,a substantial similar genetic architecture was *** is identified as an easy and reliable approach to study hepatic fat content at the population level.

Production of cowpea (Vigna unguiculata (L.) Walp.), a staple legume crop in Sub-Saharan Africa, faces challenges due to biotic and abiotic constraints. Induced mutagenesis was deployed to create genetic variation in two cowpea varieties (KVX396-4-5-2D and Moussa local). The radio-sensitivity tests led to determe the lethal dose 50 (LD50) corresponding to 230 Gy and 220 Gy for KVX396-4-5-2D and Moussa local varieties, respectively. Dried seeds (M0) of each variety were gamma-ray irradiated with LD50 − 50, LD50 and LD50 + 50. M1 seeds were advanced to generate M2, M3 and M4 mutants using the single-seed-descent method. M4 mutant lines were evaluated in rain-fed conditions using a randomized complete block design to assess phenotypic differences. Data on seven qualitative and eleven quantitative traits were collected. The results indicated that the mutation induced variability in three qualitative traits: in KVX 396-4-5-2D mutant lines, with flower and seed color frequencies at 2.61% and 0.56% respectively, and pod dehiscence at a frequency of 0.24%. While in Moussa local mutants, a pod color changed at a frequency of 17%. ANOVA results revealed significant differences between mutants of both varieties for all quantitative traits, including photosynthetic parameters. Positive correlations were observed between leaf diameter and 100-seed weight, and between branch number and 100-seed weight. Hierarchical clustering revealed three clusters among KVX 396-4-5-2D mutants and six clusters among Moussa local mutants. Early maturity and high foliage were induced traits in Cluster 3 of KVX 396-4-5-2D mutants while high hundred-seed weight was induced in Cluster 6 of Moussa local mutants.

Objective:To investigate whether major dengue outbreaks in the last two decades in Kaohsiung follow a precise temporal ***:Government daily lab-confirmed dengue case data from three major dengue outbreaks occurring during the last two decades in Kaohsiung in2002,2014 and 2015,is utilized to compute the corresponding weekly cumulative percentage of total case *** divide each of the three time series data into two periods to examine the corresponding weekly cumulative percentages of case numbers for each ***’s correlation coefficient was calculated to compare quantitatively the similarity between the temporal patterns of these three ***:Three cutoff points produce the most interesting comparisons and the most different ***’s correlation coefficient indicates quantitative discrepancies in the similarity between temporal patterns of the three years when using different cutoff ***:Temporal patterns in 2002 and 2014 are comparatively more similar in early *** 2015 outbreak started late in the year,but ended more like the outbreak in 2014,both with record-breaking number of *** retrospective analysis shows that the temporal dynamics of dengue outbreaks in Kaohsiung can strongly vary from one year to another,making it difficult to identify any common predictor.

This retrospective single-center registry study included all consecutive patients who underwent percutaneous coronary intervention (PCI) for a de novo left main coronary artery lesion using drug coated-balloon (DCB)-only strategy between August 2011 and December 2018. To best of our knowledge, no previous studies of DCB-only strategy of treating de novo left main coronary artery disease, exist. The primary endpoint was major adverse cardiovascular events (MACEs) including cardiac death, non-fatal myocardial infarction, and target lesion revascularization (TLR). The cohort was divided into two groups depending on weather the lesion preparation was done according to the international consensus group guidelines. Sixty-six patients (mean age 75±8.6, 72% male), 52% of whom had acute coronary syndrome, underwent left main PCI with the DCB-only strategy. No procedural mortality and no acute closures of the treated left main occurred. At 12 months, MACE and TLR occurred in 24% and 6% of the whole cohort, respectively. If the lesion preparation was done according to the guidelines, the MACE and TLR rates were 21.2% and 1.9%. Left main PCI with the DCB only-strategy is safe leading to acceptable MACE and low TLR rates at one year, if the lesion preparation is done according to the guidelines.
