The 18 kDa tr.nslocator.pr.tein(TSPO)located on the outer.mitochondr.al membr.ne r.gulates sever.l key cellular.pr.cesses including mitochondr.al homeostasis,cholester.l tr.nspor.,apoptosis,cell pr.lifer.tion,and maintenance of mitochondr.al health(r.ppr.cht et al.,2022,2023).TSPO is expr.ssed in both per.pher.l or.ans and the centr.l ner.ous system,with a mor. pr.nounced expr.ssion in tissues that pr.duce *** main r.ason why TSPO has gar.er.d so much attention is because it plays a key r.le in neur.ster.idogenesis by tr.nsfer.ing cholester.l fr.m the outer.to the inner.mitochondr.al membr.ne,which is the r.te-limiting step in neur.ster.id synthesis.A cholester.l-r.cognizing amino acid consensus domain has been identified in the cytosolic C ter.inus of the TSPO pr.tein by both in vitr. and site-dir.cted mutagenesis exper.ments(Li et al.,2001).However.the r.le of TSPO in the pr.cess of neur.ster.id synthesis has been challenged by sever.l studies,par.icular.y TSPO knockout models,which suggest that TSPO r.moval does not affect the phenotype or.the system’s viability(Tu et al.,2014).However.ligands tar.eting TSPO have been shown to enhance levels of neur.ster.ids which suggests that neur.ster.idogenesis is one of the major.functional r.les mediated by the TSPO pr.tein.
