Comparative oral metagenome insight into acute and chronic root canal infections

Background:In the dynamic progression of root canal infections,the resultant change in environmental conditions directs the adaptation of the microbiome in terms of richness,evenness,and diversity.Objective:The goal of this study is to contrast the interradicular bacterial community structures of teeth in acute and chronic stages and investigate potential microorganisms associated with persistent apical periapical infections.Methods:Interradicular samples were collected from 10 teeth in the apical periodontitis group and 10 teeth in the chronic apical periodontitis group.High-throughput next-generation sequencing(NGS)–based microbial identification through the Illumina platform was used to characterize the complex intraradicular microbial communities in acute and chronic root canal infections.Results:Bacteria were the major domain,with a prevalence of>97%(acute cases:98.11%and chronic cases:97.6%)compared to eukaryotes and archaea.Bacterial diversity was relatively higher in acute compared to chronic conditions under 10 and 9 phyla,79 and 64 genera,and 172 and 135 species,respectively.These endodontically infecting bacteria belonged to Firmicutes(47.21%),Bacteroidetes(20.07%),and Proteobacteria(10.72%)in acute conditions,while in the chronic stage,Firmicutes(40.85%),Proteobacteria(27.80%),and Actinobacteria(14.79%)were abundant.The phylum Candidatus Saccharibacteria was exclusively found in acute samples.Spirochaetes and Synergistetes were found predominantly in acute infections compared to chronic infections.Bacillus and Lactobacillus were found in equal and highest abundance under the phylum Firmicutes in both conditions.Conclusion:This study provides information on the differential microbial community present in the apical root canal system of teeth with acute and chronic apical periodontitis and contributes to new approaches or channels for future research on preventive measures and therapeutic protocols during disease progression and treatment.However,functional characterization and detection of antimicrobia

Fractal analysis of major faults and fractal dimension of lineaments in the Indo-Gangetic Plain on a regional scale

The Indo-Gangetic Plain(IGP)is one of the most seismically vulnerable areas due to its proximity to the Himalayas.Geographic information system(GIS)-based seismic characterization of the IGP was performed based on the degree of deformation and fractal dimension.The zone between the Main Boundary Thrust(MBT)and the Main Central Thrust(MCT)in the Himalayan Mountain Range(HMR)experienced large variations in earthquake magnitude,which were identified by Number-Size(NS)fractal modeling.The central IGP zone experienced only moderate to low mainshock levels.Fractal analysis of earthquake epicenters reveals a large scattering of earthquake epicenters in the HMR and central IGP zones.Similarly,the fault fractal analysis identifies the HMR,central IGP,and south-western IGP zones as having more faults.Overall,the seismicity of the study region is strong in the central IGP,south-western IGP,and HMR zones,moderate in the western and southern IGP,and low in the northern,eastern,and south-eastern IGP zones.

Otitis media(OM),the most underrated middle ear disease,often onsets during childhood and gets complicated in susceptible patients.Inflammation of the middle ear(ME)leads to its degradation and abnormal physiological functions.Prolonged ME inflammation sometimes results in lethal secondary complications(Penido et al.,2016).OM is a corroboration of multiple facets that leads to disabling hearing loss and associated complications.Some of the facets constituted are genetic,epigenetic,immunogenetic,anatomical.

ML-Fresh: Novel Routing Protocol in Opportunistic Networks Using Machine Learning

Opportunistic Networks(OppNets)is gaining popularity day-by-day due to their various applications in the real-life world.The two major reasons for its popularity are its suitability to be established without any requirement of additional infrastructure and the ability to tolerate long delays during data communication.Opportunistic Network is also considered as a descendant of Mobile Ad hoc Networks(Manets)and Wireless Sensor Networks(WSNs),therefore,it inherits most of the traits from both mentioned networking techniques.Apart from its popularity,Opportunistic Networks are also starting to face challenges nowadays to comply with the emerging issues of the large size of data to be communicated and blind forwarding of data among participating nodes in the network.These issues lower the overall performance of the network.Keeping this thing in mind,ML-Fresh-a novel framework has been proposed in this paper which focuses to overcome the issue of blind forwarding of data by maintaining an optimum path between any pair of participating nodes available in the OppNet using machine learning techniques viz.pattern prediction,decision tree prediction,adamic-adar method for complex networks.Apart from this,ML-Fresh also uses the history of successful encounters between a pair of communicating nodes for route prediction in the background.Simulation results prove that the ML-Fresh outperforms the existing framework of Opportunistic Networks on the grounds of standard Quality-of-Service(QoS)parameters.

Fake News Classification Using a Fuzzy Convolutional Recurrent Neural Network

In recent years,social media platforms have gained immense popularity.As a result,there has been a tremendous increase in content on social media platforms.This content can be related to an individual’s sentiments,thoughts,stories,advertisements,and news,among many other content types.With the recent increase in online content,the importance of identifying fake and real news has increased.Although,there is a lot of work present to detect fake news,a study on Fuzzy CRNN was not explored into this direction.In this work,a system is designed to classify fake and real news using fuzzy logic.The initial feature extraction process is done using a convolutional recurrent neural network(CRNN).After the extraction of features,word indexing is done with high dimensionality.Then,based on the indexing measures,the ranking process identifies whether news is fake or real.The fuzzy CRNN model is trained to yield outstanding resultswith 99.99±0.01%accuracy.This work utilizes three different datasets(LIAR,LIAR-PLUS,and ISOT)to find the most accurate model.

Sentiment Analysis on Social Media Using Genetic Algorithm with CNN

There are various intense forces causing customers to use evaluated data when using social media platforms and microblogging sites.Today,customers throughout the world share their points of view on all kinds of topics through these sources.The massive volume of data created by these customers makes it impossible to analyze such data manually.Therefore,an efficient and intelligent method for evaluating social media data and their divergence needs to be developed.Today,various types of equipment and techniques are available for automatically estimating the classification of sentiments.Sentiment analysis involves determining people’s emotions using facial expressions.Sentiment analysis can be performed for any individual based on specific incidents.The present study describes the analysis of an image dataset using CNNswithPCA intended to detect people’s sentiments(specifically,whether a person is happy or sad).This process is optimized using a genetic algorithm to get better results.Further,a comparative analysis has been conducted between the different models generated by changing the mutation factor,performing batch normalization,and applying feature reduction using PCA.These steps are carried out across five experiments using theKaggledataset.The maximum accuracy obtained is 96.984%,which is associated with the Happy and Sad sentiments.

Scrub typhus is a neglected disease and one of the most serious health problems in the Asia-Pacific region.The disease is caused by an obligate intracellular bacteria Orientia tsutsugamushi,which is transmitted by chigger bites or larval mite bites.Scrub typhus is a threat to billions of people worldwide causing different health complications and acute encephalitis in infants and growing children.The disease causes multiple organ failure and mortality rates may reach up to 70%due to a lack of appropriate healthcare.Currently available genome and proteome databases,and bioinformatics methods are valuable tools to develop novel therapeutics to curb the pathogen.This review discusses the state-of-the-art of information about Orientia tsutsugamushi-mediated scrub typhus and delineates the role of omics technologies to develop drugs against the pathogen.The role of proteome-wide in silico approaches for the identification of therapeutic targets is also highlighted.

Single Molecule Analysis of the Arabidopsis FRA1 Kinesin Shows that It Is a Functional Motor Protein with Unusually High Processivity

The Arabidopsis FRA1 kinesin contributes to the organization of cellulose microfibrils through an unknown mechanism. The cortical localization of this kinesin during interphase raises the possibility that it transports cell wallrelated cargoes along cortical microtubules that either directly or indirectly influence cellulose microfibril patterning. To determine whether FRA1 is an authentic motor protein, we combined bulk biochemical assays and single molecule fluorescence imaging to analyze the motor properties of recombinant, GFP-tagged FRA1 containing the motor and coiled-coil domains （designated as FRAI（707）-GFP）. We found that FRAI（707）-GFP binds to microtubules in an ATP-dependent manner and that its ATPase activity is dramatically stimulated by the presence of microtubules. Using single molecule studies, we found that FRAI（707）-GFP moves processively along microtubule tracks at a velocity of about 0.4 μm s-1. In addition, we found that FRAI（707）-GFP is a microtubule plus-end-directed motor and that it moves along microtubules as a dimer. Interestingly, our single molecule analysis shows that the processivity of FRAI（707）-GFP is at least twice the processivity of conventional kinesin, making FRA1 the most processive kinesin to date. Together, our data show that FRA1 is a bona fide motor protein that has the potential to drive Iong-distance transport of cargo along cortical microtubules.

Quercetin attenuates the development of 7,12-dimethyl benz(a)anthracene(DMBA) and croton oil-induced skin cancer in mice

To evaluate the chemopreventive potential of quercetin in an experimental skin carcinogenesis mouse model.Skin tumor was induced by topical application of 7,12-dimethyl Benz（a） anthracene（DMBA） and Croton oil in Swiss albino mouse.Quercetin was orally administered at a concentration of 200 mg/kg and 400 mg/kg body weight daily for 16 weeks in mouse to evaluate chemopreventive potential.Skin cancer was assessed by histopathological analysis.We found that quercetin reduced the tumor size and the cumulative number of papillomas.The mean latent period was significantly increased as compared to carcinogen treated controls.Quercetin significantly decreased the serum levels of glutamate oxalate transaminase,glutamate pyruvate transaminase,alkaline phosphatase and bilirubin.It significantly increased the levels of glutathione,superoxide dismutase and catalase.The elevated level of lipid peroxides in the control group was significantly inhibited by quercetin.Futhermore,DNA damage was significantly decreased in quercetin treated mice as compared to DMBA and croton oil treated mice.The results suggest that quercetin exerts chemopreventive effect on DMBA and croton oil induced skin cancer in mice by increasing antioxidant activities.

Assessment of compatibility of rhlGF-1/rhlGFBP-3 with neonatal intravenous medications

Background Recombinant human(rh)IGF-1/IGFBP-3 protein complex,administered as a continuous intravenous infusion in preterm infants,is being studied for the prevention of complications of prematurity.Methods We conducted in vitro studies to evaluate the physical and chemical compatibility of rhIGF-1/IGFBP-3 with medications routinely administered to preterm neonates.In vitro mixing of rhIGF-1/IGFBP-3 drug product with small-molecule test medications plus corresponding controls was performed.Physical compatibility was defined as no color change,precipitation,turbidity,gas evolution,no clinically relevant change in pH/osmolality or loss in medication content.Chemical compatibility of small molecules was assessed using liquid chromatography(e.g.,reverse-phase HPLC and ion chromatography),with incompatibility defined as loss of concentration of≥10%.A risk evaluation was conducted for each medication based on in vitro compatibility data and potential for chemical modification.Results In vitro physical compatibility was established for 11/19 medications:caffeine citrate,fentanyl,fluconazole,gen-tamicin,insulin,intravenous fat emulsion,midazolam,morphine sulfate,custom-mixed parenteral nutrition solution(with/without electrolytes),parenteral nutrition solution+intravenous fat emulsion,and vancomycin(dosed from a 5 mg/mL solu-tion),but not for 8/19 medications:amikacin,ampicillin,dopamine,dobutamine,furosemide,meropenem,norepinephrine,and penicillin G,largely owing to changes in pH after mixing.Small-molecule compatibility was unaffected post-mixing,with no loss of small-molecule content.For physically compatible medications,risk analyses confirmed low probability and severity of a risk event.Conclusion Co-administration of rhIGF-1/rhIGFBP-3 drug product with various medications was assessed by in vitro studies using case-by-case risk analyses to determine the suitability of the products for co-administration.
