Tetramic acid-motif natural products from a marine fungus Tolypocladium cylindrosporum FB06 and their anti-Parkinson activities

Tetramic acid-containing natural products are attracting significantly increasing attention from biologists and chemists due to their intriguing structures and biological activities.In the present study,two new tetramic acid alkaloids tolypyridone I(1)and tolypyridone J(2),together with five known ones(3–7),were isolated from cultures of a marine fungus Tolypocladium cylindrosporum FB06 isolate obtained from a marine sediment in Beaufort sea of North Alaska.Their structures were elucidated using 1D,2D NMR,and HRESIMS.Their configurations were established on the basis of 1H coupling constants,ROESY correlations and DP4 calculations.Compound 2 was isolated as mixtures of rotational isomers with C-3 to C-7 axis between 4-hydroxy-2-pyridone and 1-ethyl-3,5-dimethylcyclohexane,hindering rotation.In our unbiased screening to discover neuroprotective compounds in an in vitro Parkinson’s disease(PD)model,SH-SY5Y dopaminergic cells were treated with isolated compounds followed by treatment with 1-methyl-4-phenylpyridinium(MPP^(+)),a parkinsonian neurotoxin.Among tested compounds,F-14329(7)significantly protected cells from MPP^(+)-induced cytotoxicity.MPP^(+)-mediated cell death is known to be related to the regulation of Bcl-2 family proteins,specifically the down-regulation of anti-apoptotic Bcl-2 and the up-regulation of pro-apoptotic Bax levels.Treatment with 2 mmol/L of MPP^(+)for 24 h significantly reduced Bcl-2 levels compared to control treated with vehicle.However,treatment with F-14329(7)attenuated such reduction.This study demonstrates that tetramic acid-motif compounds could be potential lead compounds for treating PD.

Deep Learning Model Ensemble for the Accuracy of Classification Degenerative Arthritis

Artificial intelligence technologies are being studied to provide scientific evidence in the medical field and developed for use as diagnostic tools.This study focused on deep learning models to classify degenerative arthritis into Kellgren–Lawrence grades.Specifically,degenerative arthritis was assessed by X-ray radiographic images and classified into five classes.Subsequently,the use of various deep learning models was investigated for automating the degenerative arthritis classification process.Although research on the classification of osteoarthritis using deep learning has been conducted in previous studies,only local models have been used,and an ensemble of deep learning models has never been applied to obtain more accurate results.To address this issue,this study compared the classification performance of deep learning models,includingVGGNet,DenseNet,ResNet,TinyNet,EfficientNet,MobileNet,Xception,and ViT,on a dataset commonly used for osteoarthritis classification tasks.Our experimental results verified that even without applying a separate methodology,the performance of the ensemble was comparable to that of existing studies that only used the latest deep learning model and changed the learning method.From the trained models,two ensembles were created and evaluated:weight and specialist.The weight ensemble showed an improvement in accuracy of 1%,and the proposed specialist ensemble improved accuracy,precision,recall,and F1 score by 5%,6%,6%,and 6%,respectively,compared with the results of prior studies.

Delusional parasitosis as premotor symptom of parkinson's disease:A case report

BACKGROUND Delusional parasitosis is characterized by a false belief of being infested with parasites,insects,or worms.This illness is observed in patients with Parkinson’s disease and is usually related to dopaminergic treatment.To our knowledge,no cases of delusional parasitosis have been reported as a premotor symptom or nonmotor symptom of Parkinson’s disease.CASE SUMMARY A 75-year-old woman presented with a complaint of itching that she ascribed to the presence of insects in her skin,and she had erythematous plaques on her trunk,arms,buttocks,and face.These symptoms started two months before the visit to the hospital.She took medication,including antipsychotics,with a diagnosis of delusional parasitosis,and the delusion improved after three months.A year later,antipsychotics were discontinued,and anxiety and depression were controlled with medication.However,she complained of bradykinesia,masked face,hand tremor,and mild rigidity,and we performed fluorinated N-3-fluoropropyl-2β-carbomethoxy-3β-(4-iodophenyl)nortropane positron emission tomography(PET),which showed mildly decreased DAT binding in the right anterior putamen and caudate nucleus.Parkinson’s disease was diagnosed on the basis of PET and clinical symptoms.CONCLUSION In conclusion,delusional parasitosis can be considered a non-motor sign of Parkinson’s disease along with depression,anxiety,and constipation.

Composite solid electrolytes(CSEs)have emerged as promising candidates for safe and high-energy–density solid-state lithium metal batteries(SSLMBs).However,concurrently achieving exceptional ionic conductivity and interface compatibility between the electrolyte and electrode presents a significant challenge in the development of high-performance CSEs for SSLMBs.To overcome these challenges,we present a method involving the in-situ polymerization of a monomer within a self-supported porous Li_(6.4)La_(3)Zr_(1.4)Ta_(0.6)O_(12)(LLZT)to produce the CSE.The synergy of the continuous conductive LLZT network,well-organized polymer,and their interface can enhance the ionic conductivity of the CSE at room temperature.Furthermore,the in-situ polymerization process can also con-struct the integration and compatibility of the solid electrolyte–solid electrode interface.The synthesized CSE exhibited a high ionic conductivity of 1.117 mS cm^(-1),a significant lithium transference number of 0.627,and exhibited electrochemical stability up to 5.06 V vs.Li/Li+at 30℃.Moreover,the Li|CSE|LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2) cell delivered a discharge capacity of 105.1 mAh g^(-1) after 400 cycles at 0.5 C and 30℃,corresponding to a capacity retention of 61%.This methodology could be extended to a variety of ceramic,polymer electrolytes,or battery systems,thereby offering a viable strategy to improve the electrochemical properties of CSEs for high-energy–density SSLMBs.

Heat stress adversely affects sows' performance, which can be improved by applying proper nutritional strategies. This study was conducted to investigate the interactive effects of dietary fiber levels and sources on sows' reproductive performance, metabolic response during gestation, and the carry-over influence on litter performance in the lactation period during heat stress(average room temperature of 27.1°C). Fifty-four multiparous sows(Landrace × Yorkshire; initial body weight of 236.3 ± 16 kg; 2, 3and 4 parities) at d 90 of gestation were assigned to a 2 × 3 factorial arrangement(9 sows/treatment),involving 2 dietary fiber levels(4.5% and 6% crude fiber) and 3 dietary fiber sources(wheat bran [WB],palm kernel meal [PK], and beet pulp [BP]). Sows fed the BP diet had highest(P min and120 min after the meal. The concentration of phthalic acid, succinic acid, phenylethylamine, hydrocinnamic acid, iron, linoleic acid, glycerol, ketone, and formamide were increased(P < 0.05) in the BP treatment compared with the WB. The BP treatment with high soluble fiber content improved the constipation index, farrowing duration, and litter performance, while high insoluble fibers increased sows comfort and reduced stress factors including respiratory rate and rectal temperature. Tau protein is encoded by the microtubuleassociated protein tau(MAPT)gene and stabilizes microtubules in the neurons.Tau has unique biophysical properties that make it both highly water-soluble and positively charged.Although tau is an intrinsically disordered protein lacking defined secondary structures,it can acquire highly organized-sheet structures and stack into a filamentous inclusion such as a paired helical filament(PHF).

Ni-rich layered oxides in lithium-ion batteries have problems with gas generation and electrochemical performance reduction due to residual lithium's reaction on the surface with the electrolyte.To address this issue,in this study,the Acid solvent evaporation(AsE)method has been proposed as a potential method to remove residual lithium while promoting the formation of a new LiNO_(3)-derived coating layer on the cathode surface.The reduction of residual lithium using the ASE method and the construction of a LiNO_(3)-derived coating layer suppresses gas evolution caused by the side effects of the electrolyte,improves electrochemical performance,and improves thermal stability by facilitating the smooth movement of lithium ions.Furthermore,the structural stability and resistance change due to the LiNO_(3)-derived coating layer effects is guaranteed through cycling and DCIR of the pouch cell.As a result,compared to Pristine,the capacity retention of coin cells increased by 8%after 100 cycles,and pouch cells increased by 25%after 160 cycles.In addition,after cycling the pouch cell,CO_(2) gas has significantly reduced by about 30%compared to Pristine using gas chromatography.The ASE method effectively forms a robust LiNO_(3)-derived coating layer on the cathode active material,which helps minimize electrolyte reactivity,suppress ,CO_(2) emissions,enhance surface structure stability,improve thermal stability,and improveoverallbatteryperformance.

Low molecular weight polysaccharides can be isolated from Sargassum thunbergii(LMPST)and in vitro experiments were conducted to evaluate the inhibitory effects on lipids.Two natures of LMPST were attained from S.thunbergii and appraised their LMPST on palmitic acid(PA)induced lipid accretion in Hep G2,and 3T3-L1 cells.LMPST treatment lessened lipid deposition and intracellular free fatty acid and triglyceride intensities in PA-treated above mentioned cells.The mechanistic study publicized that LMPST2 significantly suppressed adipogenesis and stimulated the PA-treated 3T3-L1 cells occupied in the lipolysis pathway.Furthermore,in PA-treated Hep G2 cells,the free fatty acid oxidation was significantly increased by LMPST2.Given these constructive properties of LMPST2 from S.thunbergii,is a potential candidate for diminishing the intracellular lipids,and for a therapeutic agent in those conditions.

This review article provides overall understanding of stainless,environment-friendly,and nonflammable Mg alloys(SEN alloys)recently developed at the Korea Institute of Materials Science.SEN alloys are produced by adding small amounts of Ca and Y(eachminor amounts of Ca and Y renders excellent mechanical properties due to improved strengthening effects.These enhanced properties are attributed to more pronounced dynamic recrystallization and grain refining behaviors caused by the second-phase particles during extrusion.(4)Despite the presence of various types of second-phase particles,the fatigue properties of SEN9 alloys are similar to those of commercial AZ91 alloys.(5)Simultaneous introduction of Ca and Y suppresses the formation of Mg17Al12 discontinuous precipitates during aging,leading to the enhanced elongation of aged SEN alloys.(6)Adding mischmetal into the SEN9 alloy leads to a six-fold enhancement in extrudability.Consequently,the studies conducted on SEN alloys demonstrate their excellent ignition and corrosion resistances and mechanical properties,which broaden the industrial applications of Mg alloys by addressing their inherent weaknesses.
