Neuroprotective Effect of Chunghyuldan(Qing Xue Dan)on Hypoxia-Reoxygenation Induced Damage of Neuroblastoma 2a Cell Lines

Objectives： Chunghyuldan （CHD）, a combinatorial drug that has anti-hyperlipidemic and anti inflammatory activities, has been shown to reduce infarct volume in a focal ischemia-reperfusion rat model. To explore the molecular basis of CHD＇s neuroprotective effect, we examined whether CHD shows a cell-protective activity and has a regulatory effect on Bax and/or B-cell leukemia/lymphoma 2 （Bcl-2） expression in mouse neuroblastoma 2a （N2a） cells subjected to hypoxia-reoxygenation （H/R）. Methods： In order to evaluate the effects of CHD on the cytotoxicity induced from hypoxia or H/R condition, lactate dehydrogenase （LDH） assay was performed. To explore whether the suppression of neural damage when pre-treated with CHD is associated with its anti-apoptotic effect, the CHD effect on the expression of Bcl-2 and Bax was analyzed by Western blotting analysis. Results： Cytotoxicity of N2a cell line was slightly increased in 42 h hypoxia condition and dramatically increased under the H/R condition. CHD treatment markedly decreased the cytotoxicity in both conditions （P〈0.01, P〈0.05）. H/R markedly increased the expression of the pro-apoptotic protein, Bax, but slightly increased the expression of the anti-apoptotic protein, Bcl-2, compared with the normoxia or hypoxia group. CHD significantly decreased Bax expression （P〈0.01） and slightly decreased Bcl-2 expression （P〉0.05）, resulted in a reduction of Bax/Bcl-2 ratio in N2a cells subjected to H/R. Conclusion： CHD has neuroprotective effect in N2a cells subjected to H/R, which might be derived at least in part from its ability to decrease the expression of the pro-apoptotic protein, Bax.

Effect of the herbal medicine Danggui Sini plus Wuzhuyu Shengjiang Tang on erythrocyte deformability in normal subjects: a cross-over trial

OBJECTIVE: To show whether Danggui Sini plus Wuzhuyu Shengjiang Tang(DSWST) has any transient effect on erythrocyte deformability in normal ***: A total of 25 subjects [mean age(27.8± 1.8) years] was enrolled in this study. The study was designed as a cross-over trial in which the subjects took part for 2 d. On the first day, blood samples were collected at baseline and 1-2 h after administration of water, whereas, on the second day, instead of water, the subjects were administered DSWST after the baseline blood sampling. The blood samples collected at baseline and after the administration water or DSWST, were examined for erythrocyte ***: The elongation index increased significantly after 2 h(P = 0.009) compared to the baseline after DSWST intake. However, after water intake, there was no significant differenceobserved. When comparing the percent change of erythrocyte deformability between DSWST and water, we found that after 2 h of administration,DSWST improved erythrocyte deformability significantly compared to water(P < 0.001).CONCLUSION: DSWST has a transient effect on erythrocyte deformability in normal subjects.

AIM： TO investigate whether krüppel-like factor 6 （KLF6） plays an important role in the development and/or progression of colorectal cancer. METHODS： A total of 123 formalin-fixed and paraffinembedded colorectal cancer specimens were analyzed by immunohistochemistry using tissue microarray for the expression of KLF6 protein. The specimens were collected over a 3-year period in the laboratories at our large teaching hospital in Seoul, Republic of Korea. The correlation of KLF6 expression with clinicopathologic parameters was analyzed by χ^2 test and Bartholomew test. RESULTS： Normal colonic epithelium showed weak to moderate expression of KLF6, whereas reduced KLF 6 expression or loss of KLF6 expression was seen in 45 （36.6%） of the 123 colorectal carcinoma specimens. Interestingly, aberrant expression of KLF6 was detected in 25 （43.1%） of 58 cases with metastasis to regional lymph node and in 31 （47.0%） of 66 tumors more than 5 cm in size. Statistically, loss of KLF6 expression was significantly associated with tumor size （P〈0.05）. However, there was no significant correlation between KLF6 expression and Dukes＇ stage （Bartholomew test, P〉 0.05）, tumor location and lymph node metastasis （χ^2 test, P〉0.05）.CONCLUSION： Loss of KLF6 expression may be a common and early event in colorectal carcinogenesis.

Anti-inflammatory effects of traditional mixed extract of medicinal herbs(MEMH)on monosodium urate crystal-induced gouty arthritis

Korean oriental medicine prescription is widely used for the treatment of gouty diseases. In the present study, we investigated anti-inflammatory effects of modified Korean herbal formulation, mixed extract of medicinal herbs(MEMH), and its modulatory effects on inflammatory mediators associated with gouty arthritis. Both in vitro and in vivo studies were carried out to assess the anti-inflammatory efficacy of MEMH on monosodium urate(MSU) crystals-induced gouty inflammation. MSU crystals stimulated human chondrosarcoma cell line, SW1353, and human primary chondrocytes were treated with MEMH in vitro. The expression levels of pro-inflammatory mediators and metalloproteases were analyzed. The effect of MEMH on NFκB signaling pathway in SW1353 cells was examined. Effect of MEMH on the mR NA expression level of pro-inflammatory mediators and chemotactic factor from human monocytic cell line, THP-1, was also analyzed. The probable role of MEMH in the differentiation process of osteoblast like cells, SaO S-2, after MSU treatment was also observed. To investigate the effects of MEMH in vivo, MSU crystals-induced ankle arthritic model was established. Histopathological changes in affected joints and plasma levels of pro-inflammatory mediators(IL-1β and TNFα) were recorded. MEMH inhibited NFκB signaling pathway and COX-2 protein expression in chondrocytes. MSU-induced mR NA expressions of pro-inflammatory mediators and chemotactic cytokines were suppressed by MEMH. In MSU crystals-induced ankle arthritic mouse model, administration of MEMH relieved inflammatory symptoms and decreased the plasma levels of IL-1β and TNFα. The results indicated that MEMH can effectively inhibit the expression of inflammatory mediators in gouty arthritis, demonstrating its potential for treating gouty arthritis.

SUPPRESSION OF DEFECTIVE DATA ARTIFACTS FOR DEBLURRING IMAGES CORRUPTED BY RANDOM VALUED NOISE

For deblurring images corrupted by random valued noise, two-phase methods first select likely-to-be reliables （data that are not corrupted by random valued noise） and then deblur images only with selected data. Two-phase methods, however, often cause defective data artifacts, which are mixed results of missing data artifacts caused by the lack of data and noisy data artifacts caused mainly by falsely selected outliers （data that are corrupted by random valued noise）. In this paper, to suppress these defective data artifacts, we propose a blurring model based reliable-selection technique to select reliables as many as possible to make all of to-be-recovered pixel values to contribute to selected data, while excluding outliers as accurately as possible. We also propose a normalization technique to compensate for non-uniform rates in recovering pixel values. We conducted simulation studies on Gaussian and diagonal deblurring to evaluate the performance of proposed techniques. Simulation results showed that proposed techniques improved the performance of two-phase methods, by suppressing defective data artifacts effectively.

Pin1 overexpression in colorectal cancer and its correlation with aberrant β-catenin expression

AIM： To investigate clinical significance of Pin1 and β-catenin expression in colorectal cancers and to demonstrate the relationship of their expression. METHODS： The role of Pin1 and β-catenin protein in colorectal tumorigenesis and their clinicopathologic significance were analyzed by immunohistochemistry, and correlation between Pin1 and β-catenin protein expressions was also studied in 124 patients with colorectal cancer who were surgically treated. RESULTS： Normal colonic epithelium either failed to express or showed focal and weak expression of Pin1 and β-catenin. Overexpression of Pin1 and β-catenin protein was found in 23 （18.54%） and 50 （40.3%） of 124 colorectal cancers, respectively. Overexpression of both proteins was not related to the lymph node metastasis, tumor stage and survival period after excision. Survival analysis results indicated that tumor stage was a valuable predictor of survival. Interestingly, a significant correlation was found between Pin1 and β-catenin protein expression. CONCLUSION： Overexpression of Pin1 and β-catenin may be closely related with the development and/or progression of colorectal carcinoma and further supports that Pin1 overexpression might contribute to the upregulation of β-catenin.

Previous studies have demonstrated that male hypogonadism is associated with a low level of vitamin D. However, no reports have investigated the effects of vitamin D on testosterone levels in Korean men. Our aim was to investigate whether testosterone levels are associated with serum vitamin D levels and whether seasonal variation exists. This cross-sectional study analyzed serum 25-hydroxyvitamin D [25（OH）D], total testosterone （TT）, and free testosterone （FT） in 652 Korean men over 40 years of age who had undergone a comprehensive medical examination. The average age of the subjects was 56.7± 7.9 years, and the mean serum 25（OH）D, TT and FT levels were 21.23 ± 7.9 ng m1-1, 4.70 ±1.6 ng m1-1, and 8.12± 3.3 pg m1-1, respectively. In the multiple linear regression model, 25（OH）D showed positive association with TT （β =0.137, P〈 0.001） and FT （p =0.103, P= 0.008）. 25（OH） D and FT showed similar seasonal or monthly variation after adjustment for age. A vitamin D deficiency [25（OH）D 〈 20 ng m1-1] was associated with an increased risk of deficiencies of TT （〈2.30 ng ml-1）（odds ratio [OR]： 2.65; 95% confidence interval [CI]. 1.21-5.78, P = 0.014） and FT （〈6.50 pg ml-1）（OR： 1.44; 95% Ch 1.01-2.06 P = 0.048） after adjusting for age, season, body mass index, body composition, chronic disease, smoking, and alcohol use. In conclusion, we demonstrated a positive correlation between 25（OH）D and testosterone, which showed similar seasonal variation in Korean men.

Facile synthesis of fully ordered L10-FePt nanoparticles with controlled Pt-shell thicknesses for electrocatalysis

We report a simple one-step approach for the synthesis of -4 nm uniform and fully L10-ordered face-centered tetragonal （fct） FePt nanoparticles （NPs） embedded in -60 nm MCM-41 （fct-FePt NPs@MCM-41）. We controlled the Pt-shell thickness of the fct-FePt NPs by treating the fct-FePt NPs@MCM-41 with acetic acid （HOAc） or hydrochloric acid （HC1） under sonicafion, thereby etching the surface Fe atoms of the NPs. The fct-FePt NPs deposited onto the carbon support （fct-FePt NP/C） were prepared by mixing the fct-FePt NPs@MCM-41 with carbon and subsequently removing the MCM-41 using NaOH. We also developed a facile method to synthesize acid-treated fct-FePt NP/C by using a HF solution for simultaneous surface-Fe etching and MCM-41 removal. We studied the effects of both surface-Fe etching and Pt-shell thickness on the electrocatalytic properties of fct-FePt NPs for the methanol oxidation reaction （MOR）. Compared with the non-treated fct-FePt NP/C catalyst, the HOAc-treated and HCl-treated catalysts exhibit up to 34% larger electrochemically active surface areas （ECASAs）; in addition, the HCl-treated fct-FePt NP （with -1.0 nm Pt shell）/C catalyst exhibits the highest specific activity. The HF-treated fct-FePt NP/C exhibits an ECASA almost 2 times larger than those of the other acid-treated fct-FePt NP/C catalysts and shows the highest mass activity （1,435 mA·mgPt^-1, 2.3 times higher than that of the commercial Pt/C catalyst） and stability among the catalysts tested. Our findings demonstrate that the surface-Fe etching for the generation of the Pt shell on fct-FePt NPs and the Pt-shell thickness can be factors for optimizing the electrocatalysis of the MOR.

Synthesis of shape-controlled Pt nanocrystals is substantial and important for enhancing chemical and electrochemical ***,the removal of capping agents,shape-controlling chemicals,on Pt surfaces is essential prior to conducting the catalytic *** we report a facile one-pot synthesis of Pt nanocubes directly grown on carbon supports(Pt nanocubes/C) with modulating the kinetic reaction factors for shaping the nanocrystals,but without adding any capping agents for preserving the clean Pt ***-dispersed Pt nanocubes/C shows enhanced activity and long-term stability toward methanol oxidation reaction compared to the commercial Pt/C catalyst.
