The positive effect of levodopa in the treatment of Parkinson’s disease,although it is limited in time and has severe side effects,has encouraged the scientific community to look for new drugs that can stop the neurodegenerative process or even regenerate the neuromelanin-containing dopaminergic nigrostriatal *** preclinical studies with coenzyme Q10,mitoquinone,isradipine,nilotinib,TCH346,neurturin,zonisamide,deferiprone,prasinezumab,and cinpanemab prompted clinical ***,these failed and after more than 50 years levodopa continues to be the key drug in the treatment of the disease,despite its severe side effects after 4–6 years of chronic *** lack of translated successful results obtained in preclinical investigations based on the use of neurotoxins that do not exist in the human body as new drugs for Parkinson’s disease treatment is a big *** our opinion,the cause of these failures lies in the experimental animal models involving neurotoxins that do not exist in the human body,such as 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine and 6-hydroxydopamine,that induce a very fast,massive and expansive neurodegenerative process,which contrasts with the extremely slow one of neuromelanin-containing dopaminergic *** exceedingly slow progress of the neurodegenerative process of the nigrostriatal neurons in idiopathic Parkinson’s patients is due to(i)a degenerative model in which the neurotoxic effect of an endogenous neurotoxin affects a single neuron,(ii)a neurotoxic event that is not expansive and(iii)the fact that the neurotoxin that triggers the neurodegenerative process is produced inside the neuromelanin-containing dopaminergic *** endogenous neurotoxin that fits this degenerative model involving one single neuron at a time is aminochrome,since it(i)is generated within neuromelanin-containing dopaminergic neurons,(ii)does not cause an expansive neurotoxic effect and(iii)triggers all the mechanisms involved in t

Huntington’s disease is a neurodegenerative disease caused by the expansion mutation of a cytosine-adenine-guanine triplet in the exon 1 of the HTT gene which is responsible for the production of the huntingtin (Htt) protein. In physiological conditions, Htt is involved in many cellular processes such as cell signaling, transcriptional regulation, energy metabolism regulation, dNA maintenance, axonal trafficking, and antiapoptotic activity. When the genetic alteration is present, the production of a mutant version of Htt (mHtt) occurs, which is characterized by a plethora of pathogenic activities that, finally, lead to cell death. Among all the cells in which mHtt exerts its dangerous activity, the GABAergic Medium Spiny Neurons seem to be the most affected by the mHtt-induced excitotoxicity both in the cortex and in the striatum. However, as the neurodegeneration proceeds ahead the neuronal loss grows also in other brain areas such as the cerebellum, hypothalamus, thalamus, subthalamic nucleus, globus pallidus, and substantia nigra, determining the variety of symptoms that characterize Huntington’s disease. From a clinical point of view, Huntington’s disease is characterized by a wide spectrum of symptoms spanning from motor impairment to cognitive disorders and dementia. Huntington’s disease shows a prevalence of around 3.92 cases every 100,000 worldwide and an incidence of 0.48 new cases every 100,000/year. To date, there is no available cure for Huntington’s disease. Several treatments have been developed so far, aiming to reduce the severity of one or more symptoms to slow down the inexorable decline caused by the disease. In this context, the search for reliable strategies to target the different aspects of Huntington’s disease become of the utmost interest. Background Many cancer patients seek adjunctive therapies to biomedical cancer treatments at some point of their disease *** acupuncture is increasingly recommended by leading oncological associations,limited evidence exists concerning the evidence-informed practice and adherence to current guidelines of traditional complementary and integrative medicine(TCIM)practitioners treating cancer *** An international online-survey assessed the demographical data,clinical practice,and sources of information used by TCIM practitioners in Austria,Germany,United States of America,Australia,and New *** In total,404 respondents completed the survey,of which 254(62.9%)treated cancer *** practitioners were acupuncturists and herbalists(57.1%),had(16.8±9.9)years of clinical experience and see a median of 2(1,4)cancer patients per *** cancer(61.8%)is the most common cancer type seen in TCIM *** TCIM treatments are frequently concurrent with the patient’s cancer specific treatment(39.9%),which is also reflected by the main goal of a TCIM treatment to alleviate side effects(52.4%).However,only 28.0%of the respondents are in contact with the treating *** to the respondents,pain is most effectively treated using acupuncture,while herbal medicine is best for cancer-related *** practitioners mostly use certified courses(33.1%)or online databases(28.3%)but often believe that experts are more reliable to inform their practice(37.0%)than research publications(32.7%).Conclusion Acupuncturists and herbalists commonly treat cancer *** practitioners use TCIM as an adjunct to biomedicine as supportive care and use it largely in accordance with current oncological guidelines.

The extension of dual-comb spectroscopy(dCS)to all wavelengths of light along with its ability to provide ultralarge dynamic range and ultra-high spectral resolution,renders it extremely useful for a diverse array of applications in physics,chemistry,atmospheric science,space science,as well as medical *** this work,we report on an innovative technique of quartz-enhanced multiheterodyne resonant photoacoustic spectroscopy(QEMR-PAS),in which the beat frequency response from a dual comb is frequency down-converted into the audio frequency *** this way,gas molecules act as an optical-acoustic converter through the photoacoustic effect,generating heterodyne sound *** conventional dCS,where the light wave is detected by a wavelengthdependent photoreceiver,QEMR-PAS employs a quartz tuning fork(QTF)as a high-Q sound transducer and works in conjunction with a phase-sensitive detector to extract the resonant sound component from the multiple heterodyne acoustic tones,resulting in a straightforward and low-cost hardware *** novel QEMRPAS technique enables wavelength-independent dCS detection for gas sensing,providing an unprecedented dynamic range of 63 dB,a remarkable spectral resolution of 43 MHz(or~0.3 pm),and a prominent noise equivalent absorption of 5.99×10^(-6)cm^(-1)·Hz-1/2.

Photoacoustic spectroscopy(PAS)as a highly sensitive and selective trace gas detection technique has extremely broad application in many ***,the laser sources currently used in PAS limit the sensing *** to diode laser and quantum cascade laser,the solid-state laser has the merits of high optical power,excellent beam quality,and wide tuning *** we present a long-wave,high-power,wide-tunable,singlelongitudinal-mode solid-state laser used as light source in a PAS sensor for trace gas *** self-built solid-state laser had an emission wavelength of~2μm with Tm:YAP crystal as the gain material,with an excellent wavelength and optical power stability as well as a high beam *** wide wavelength tuning range of 9.44 nm covers the absorption spectra of water and ammonia,with a maximum optical power of~130 mW,allowing dual gas detection with a single laser *** solid-state laser was used as light source in three different photoacoustic detection techniques:standard PAS with microphone,and external-and intra-cavity quartz-enhanced photoacoustic spectroscopy(QEPAS),proving that solid-state laser is an attractive excitation source in photoacoustic spectroscopy.

Side-scan sonar(SSS)is now a prevalent instrument for large-scale seafloor topography measurements,deployable on an autonomous underwater vehicle(AUV)to execute fully automated underwater acoustic scanning imaging along a predetermined ***,SSS images often suffer from speckle noise caused by mutual interference between echoes,and limited AUV computational resources further hinder noise *** approaches for SSS image processing and speckle noise reduction rely heavily on complex network structures and fail to combine the benefits of deep learning and domain *** address the problem,Rep dNet,a novel and effective despeckling convolutional neural network is *** dNet introduces two re-parameterized blocks:the Pixel Smoothing Block(PSB)and Edge Enhancement Block(EEB),preserving edge information while attenuating speckle *** training,PSB and EEB manifest as double-layered multi-branch structures,integrating first-order and secondorder derivatives and smoothing *** inference,the branches are re-parameterized into a 3×3 convolution,enabling efficient inference without sacrificing *** dNet comprises three computational operations:3×3 convolution,element-wise summation and Rectified Linear Unit *** on benchmark datasets,a real SSS dataset and data collected at Lake Mulan aestablish Rep dNet as a well-balanced network,meeting the AUV computational constraints in terms of performance and latency.
