HISP-1 Inhibits HSv.1 Infection in Cultured v.ro Cells

Herpes simplex v.rus-1 (HSv.1) remains a leading cause of v.ral disease worldwide and is spread by direct contact with infected lesions. There is no v.ccine against HSv.1 infections and there remains a need to identify therapeutics that could reduce the spread. In this study v.rious hispolon compounds were analyzed to determine their antiv.ral potential against HSv.1 infections in cultured v.ro cells. To determine the effects on infectiv.ty and possible mechanisms of inhibition, the following assays were conducted. In v.tro cytotoxicity assays were conducted to determine the effect of the compounds on cell v.ability and the maximum non-cytotoxic concentrations. Antiv.ral assays measured cell v.ability, percent inhibition of infection following treatment with the compounds, and the effect on the v.ral infection cycle. These effects were v.sualized using inv.rted light and fluorescent microscopy. Of the 24 hispolons tested, only hispolon pyrazole-1 (HISP-1) demonstrated antiv.ral effects. HISP-1 was demonstrated to effect early stages in HSv.1 infection in cultured v.ro cells (attachment, penetration, and post-penetration). In silico modeling analyses were conducted to analyze the interactions between HISP-1 and v.ral glycoprotein D (gD). HISP-1 is safe at concentrations tested and is effectiv. in inhibiting infection of HSv.1 in cultured cells. HISP-1 has potential for therapeutic use as an antiv.ral against HSv.1 infection, could work in synergy with other antiv.rals that work be a different modality, and could be dev.loped as a component of a topical agent to reduce the spread of HSv.1 infections.

In this paper we will be concerned with the problem -Δu-1/2Δ(a（x）u2)u+v.x）u=f（u）,x∈R2，where v.is a potential continuous and f : R → R is a superlinear continuous function with exponential subcritical or exponential critical growth. We use as a main tool the Nehari manifold method in order to show existence of nonnegativ. solutions and existence of nodal solutions. Our results complement the classical result of solutions for quasilinear Schrodinger equations v.a the Nehari method due to Jia-quan Liu, Ya-qi Wang and Zhi-Qiang Wang in the sense that in this article we are considering nonlinearity of the exponential type.

Bioethics falls within the controv.rsial ethical issues from the new adv.nces and emerging technology in pre-clinical and clinical settings which inspired medical policy and practice. The purpose of this paper is to present an ov.rv.ew of ethical issues in stem cell-based therapies and to prov.de substantial argument for the need of a Bioethics Research Consortium that will be tasked with conv.ning a disciplined board of experts that will apply their principles to biomedical research, and emerging technology to further stipulate the socioeconomic influence of such entities and their therapeutic impact to society. Not too long ago, the successful therapeutic studies in Parkinson’s disease and stroke were the highlights of bioethical issues. The precedent for study selection was based on public feedback, gov.rnment reception, and scientific analyses dev.loped by these spearhead studies. From all the negativ. publicity that researchers hav. been getting before, the whiplash from decades of fear and misunderstanding has hindered the progress of scientific study. There is a huge figurativ. tug of war between what is making profit and what is needed to improv. the health of afflicted patients worldwide. Poor management of education and funding may hav. actually hindered potential life changing cures and treatments from coming to fruition. We propose the creation of a Bioethics Research Consortium that would determine how ethical matters are handled with careful consideration to public need. The goal is to restructure drug dev.lopment policy, and improv. upon the ways in which research methods and funds are handled today.

The distribution of photo-crosslinkable moieties onto a protein backbone can affect a biomaterial’s crosslinking behav.or, and therefore also its mechanical and biological properties. A profound insight in this respect is essential for biomaterials exploited in tissue engineering and regenerativ. medicine. In the present work, photo-crosslinkable moieties hav. been introduced on the primary amine groups of: (i) a recombinant collagen peptide (RCPhC1) with a known amino acid (AA) sequence, and (ii) bov.ne skin collagen (COL BS) with an unknown AA sequence. The degree of substitution (DS) was quantified with two conv.ntional techniques: an ortho-phthalic dialdehyde (OPA) assay and ^(1)H NMR spectroscopy. Howev.r, neither of both prov.des information on the exact type and location of the modified AAs. Therefore, for the first time, proteomic analysis was ev.luated herein as a tool to identify functionalized AAs as well as the exact position of photo-crosslinkable moieties along the AA sequence, thereby enabling an in-depth, unprecedented characterization of functionalized photo-crosslinkable biopolymers. Moreov.r, our strategy enabled to v.sualize the spatial distribution of the modifications within the ov.rall structure of the protein. Proteomics has prov.n to prov.de unprecedented insight in the distribution of photo-crosslinkable moieties along the protein backbone, undoubtedly contributing to superior functional biomaterial design to serv. regenerativ. medicine.
