Plec.ranthus barbatus is a popular tropic.l perennial plant with a wide variety of traditional medic.nal uses in tropic.l Afric., Hindu, Ayurvedic.and traditional medic.nes of Brazil and c.ina. The whole plant and the leaves have many folkloric.uses for diverse ailments inc.uding pain, heart disease, c.nvulsions, c.ughs and c.lds, asthma, bronc.itis and tonsillitis among others. This study investigated the phytoc.emic.l c.mponents, ac.te toxic.ty, analgesic.and anti-pyretic.ac.ivities of the aqueous leaf extrac. of Plec.ranthus barbatus loc.lly known as ekizeera in Uganda. The plant leaves were authentic.ted, c.llec.ed and dec.c.ion was done ac.ording to loc.l method. Phytoc.emic.l sc.eening was c.nduc.ed using methods outlined by Trease and evans and Harborne to determine the c.mponents of the extrac.. Ac.te toxic.ty tests were c.nduc.ed in rats using modified Lorke’s method to determine the safety of the plant material. Analgesic.studies were c.rried out using both a mec.anic.l method (thermally induc.d pain by tail-flic.) and a c.emic.l method (formalin induc.d pain) in rats by administering extrac.s orally at 100, 200 and 400 mg/kg of body weight. The method of Al-Ghamdi, modified for loc.l laboratory setting by Adzu was adopted and used for anti-pyretic.test. Dec.c.ion yielded 9.9% extrac.. Phytoc.emic.l sc.eening c.nfirmed presenc. of saponins, tannins, alkaloids, terpenoids and essential oils. Ac.te toxic.ty tests revealed no deaths in rats after oral treatment with up to 10,000 mg/kg of extrac.. Tail-flic. test was non-signific.nt (p > 0.05) while formalin-induc.d pain test demonstrated signific.nt ac.ivity (p -tests. Anti-pyretic.ac.ivity was non-signific.nt (p > 0.05) with student t-test. These results suggest that the aqueous leaf extrac. of Plec.ranthus barbatus c.ntains spec.fic.phytoc.emic.ls, has a potent dose dependent analgesic.ac.ivity, no anti-pyretic.ac.ivity and c.n be regarded as a safe medic.nal plant to use traditionally, whic. might further
