Loss of disabled-2 expression is an early event in esophageal squamous tumorigenesis

AIM: Disabled-2 (DAB2) is a candidate tumor-suppressor gene identified in ovarian cancer that negatively influences mitogenic signal transduction of growth factors and blocks ras activity. In a recent study, we observed down-regulation of DAB2 transcripts in ESCCs using cDNA microarrays. In the present study, we aimed to determine the clinical significance of loss of DAB2 protein in esophageal tumorigenesis, hypothesizing that DAB2 promoter hypermethylation-mediated gene silencing may account for loss of the protein. METHODS: DAB2 expression was analyzed by immunohistochemistry in 50 primary esophageal squamous cell carcinomas (ESCCs), 30 distinct hyperplasia, 15 dysplasia and 10 non-malignant esophageal tissues. To determine whether promoter hypermethylation contributes to loss of DAB2 expression in ESCCs, methylation status of DAB2 promoter was analyzed in DAB2 immuno-negative tumors using methylation-specifi c PCR. RESULTS: Loss of DAB2 protein was observed in 5/30 (17%) hyperplasia, 10/15 (67%) dysplasia and 34/50 (68%) ESCCs. Significant loss of DAB2 protein was observed from esophageal normal mucosa to hyperplasia, dysplasia and invasive cancer (Ptrend < 0.001). Promoter hypermethylation of DAB2 was observed in 2 of 10 (20%) DAB2 immuno-negative ESCCs. CONCLUSION: Loss of DAB2 protein expression occurs in early pre-neoplastic stages of development of esophageal cancer and is sustained down the tumorigenic pathway. Infrequent DAB2 promoter methylation in ESCCs suggests that epigenetic genesilencing is only one of the mechanisms causing loss of DAB2 expression in ESCCs.

An Interacting Gauge Field Theoretic Model for Hodge Theory: Basic Canonical Brackets

We derive the basic canonical brackets amongst the creation and annihilation operators for a two(1 + 1)-dimensional(2D) gauge field theoretic model of an interacting Hodge theory where a U(1) gauge field(Aμ) is coupled with the fermionic Dirac fields(ψ andˉψ). In this derivation, we exploit the spin-statistics theorem, normal ordering and the strength of the underlying six infinitesimal continuous symmetries(and the concept of their generators) that are present in the theory. We do not use the definition of the canonical conjugate momenta(corresponding to the basic fields of the theory) anywhere in our whole discussion. Thus, we conjecture that our present approach provides an alternative to the canonical method of quantization for a class of gauge field theories that are physical examples of Hodge theory where the continuous symmetries(and corresponding generators) provide the physical realizations of the de Rham cohomological operators of differential geometry at the algebraic level.

DTM Simulation of Peristaltic Viscoelastic Biofluid Flow in Asymmetric Porous Media： A Digestive Transport Model

A biofluid dynamics mathematical model is developed to study peristaltic flow of non-Newtonian physiological liquid in a two-dimensional asymmetric channel containing porous media as a simulation of obstructed digestive （intestinal） transport. The fractional Oldroyd-B viscoelastic rheological model is utilized. The biophysical flow regime is constructed as a wave-like motion and porous medium is simulated with a modified Darcy-Brinkman model. This model is aimed at describing the diges- tive transport in intestinal tract containing deposits which induce impedance. A low Reynolds number approximation is em- ployed to eliminate inertial effects and the wavelength to diameter ratio is assumed to be large. The differential transform method （DTM）, a semi-computational technique is employed to obtain approximate analytical solutions to the boundary value problem. The influences of fractional （rheological material） parameters, relaxation time, retardation time, amplitude of the wave, and permeability parameter on peristaltic flow characteristics such as volumetric flow rate, pressure difference and wall friction force are computed. The present model is relevant to flow in diseased intestines.

Evaluation of potential surface instability using finite element method in Kharsali Village,Yamuna Valley,Northwest Himalaya

Kharsali village, located in the Northwest Himalaya near the confluence of the Yamuna River and Unta Gad, is situated on a thick(>150 m) paleolandslide deposit. The village is continuously being eroded at its base by the two rivers. Cracks are noted in most houses while the ancient Shani Temple lying to the south of the village has tilted ~5° towards the northeast. Three slope sections(S-1, S-2, S-3) were modelled and analysed to determine the displacement and shear strain patterns of the slopes. Based on surface failure conditions, potential slope instability of the Kharsali village was evaluated from 2D Finite Element Method(FEM) using Shear Strain Reduction(SSR) analysis in the Phase2 software. Results indicate a critical Stress Reduction Factor(SRF) of 1.5 for the southern edge of the village(S-1) housing the Shani Temple. The development of failure surfaces at its lower portion signifies the propagating, progressive nature of the slope. The S-2 slope section is most vulnerable to slope failure, with a critical SRF of 1.08. This has been inferred by the formation of failure surfaces with displacements of 0.05-0.08 m. The S-3 section in the northern part of the Kharsali shows highest critical SRF of 2.76. The un-metalled road section in the north of the village near S-3 hasdeveloped a failure surface with displacement of 0.003-0.004 m, and a zone of subsidence. The S-3 section is relatively stable, whereas the S-2 section is the most vulnerable portion of the village.

This paper presents a new low delay centralized cooperative spectrum sensing method based on dynamic voting rule using multiple threshold level,for indoor hospital environment,where fading and direct path loss both are together responsible for variation in signal *** the proposed algorithm,weights of Cognitive Radios(CRs)in terms of assigned votecount are estimated based on the sensed energy values with respect to multiple fixed threshold levels,so that the direct path loss can be dealt without further increasing the error,which occurs due to *** indoor hospital like environment,Rician fading model is more ***,formulation,evaluation and analysis of various sensing performance parameters have been carried out considering Rician fading *** analysis and tradeoff among three optimum parameters-energy threshold,total number of CRs participating in sensing and total error rate,are presented with respect to signal to noise ratio(SNR).Finally,the Monte Carlo simulation is designed and implemented considering the two conditions-only fading environment and fading along with direct path loss *** can be observed that in both cases,the proposed spectrum sensing rule outperforms the other contemporary rules.

An Improved Methodology to Overcome Key Issues in Human Fecal Metagenomic DNA Extraction

Microbes are ubiquitously distributed in nature, and recent culture-independent studies have highlighted the significance of gut microbiota in human health and disease. Fecal DNA is the primary source for the majority of human gut microbiome studies. However, further improvement is needed to obtain fecal metagenomic DNA with sufficient amount and good quality but low host genomic DNA contamination. In the current study, we demonstrate a quick, robust, unbiased, and cost-effective method for the isolation of high molecular weight （〉 23 kb） metagenomic DNA （260/280 ratio 〉 1.8） with a good yield （55.8 ± 3.8 ng/mg of feces）. We also confirm that there is very low human genomic DNA contamination （eubacterial： human genomic DNA marker genes = 227.9：1） in the human feces. The newly-developed method robustly performs for fresh as well as stored fecal samples as demonstrated by 16S rRNA gene sequencing using 454 FLX＋. Moreover, 16S rRNA gene analysis indicated that compared to other DNA extraction methods tested, the fecal metagenomic DNA isolated with current methodology retains species richness and does not show microbial diversity biases, which is further confirmed by qPCR with a known quantity of spike-in genomes. Overall, our data highlight a protocol with a balance between quality, amount, user-friendliness, and cost effectiveness for its suitability toward usage for cultureindependent analysis of the human gut microbiome, which provides a robust solution to overcome key issues associated with fecal metagenomic DNA isolation in human gut microbiome studies.

Integrated Multi-stage LQR Power Oscillation Damping FACTS Controller

This paper presents an approach for oscillation damping with an integrated multi-stage linear quadratic regulator(MSLQR)FACTS controller combining power oscillation damping(POD)*** particle swarm optimization(PSO)technique has been used for precise tuning initial control parameters of power system stabilizers(PSS)and FACTS devices(such as STATCOM and UPFC)which results in improved controller *** is observed that the proposed control structure damps the oscillations adequately and is modular in design *** sample power system comprising six areas is considered to demonstrate the effectiveness of the proposed *** states inter-relation which is shown with eigenvalues reflects better regulation with the proposed *** step response also validates the controller performance.
