Leukemic stromal hematopoietic microenvironment negatively regulates the normal hematopoiesis in mouse model of leukemia

Background and Objective: Leukemic microenvironment has a major role in the progression of leukemia. Leukemic cells can induce reversible changes in microenvironmental components, especially the stromal function which results in improved growth conditions for maintaining the malignant leukemic cells. This study aimed to investigate the survival advantage of leukemic cells over normal hematopoietic cells in stromal microenvironment in long term. Methods: The mice were injected intraperitoneally with N-N’ ethylnitrosourea (ENU) to induce leukemia; the mice received injection of normal saline were used as control. At 180 days after ENU induction, the mice were killed and the bone marrows were cultured for 19 days. Colony-forming assays were used to analyze the formation of various cell colonies. The expression of Sca-1, CD146, VEGFR2, CD95, pStat3, pStat5, and Bcl-xL in marrow cells were detected by flow cytometry. Results: Long-term leukemic bone marrow culture showed abnormal elongated stromal fibroblasts with almost absence of normal hematopoietic cells. Adherent cell colonies were increased, but CFU-F and other hematopoietic cell colonies were significantly decreased in leukemia group (P< 0.001). Primitive progenitor-specific Sca-1 receptor expression was decreased with subsequent increased expression of CD146 and VEGFR-2 in leukemic bone marrow cells. Decreased Fas antigen expression with increased intracellular pStat3, pStat5 and Bcl-xL proteins were observed in leukemic bone marrow cells. Conclusions: Stromal microenvironment shows altered morphology and decreased maturation in leukemia. Effective progenitor cells are decreased in leukemia with increased leukemia-specific cell population. Leukemic microenvironment plays a role in promoting and maintaining the leukemic cell proliferation and survivability in long term.

Effect of TiO_2 Functionalization on Nano-Porous Silicon for Selective Alcohol Sensing at Room Temperature

Room temperature operated sensor for detection of alcohol vapours in low ppm range based on TiO_2 functionalized nano-porous silicon（PSi） is demonstrated. The effect of functionalization by TiO_2 on PSi is investigated using SEM, EDX, Raman spectroscopy, XRD and contact angle measurements. Sensing is accomplished by measuring change in resistance of the sensing layer using Cr-Au inter-digitatedelectrode（IDE） structure formed on top of the functionalized PSi layer. The sensors were tested for volatile organic compounds（VOCs） and water vapours in the wide range of 5–500 ppm concentration at room temperature. Functionalization of the nanostructured PSi by sputter deposited TiO_2 results in significant enhancement of sensitivity and inverse change in selectivity. PSi sensors have displayed strong response to water vapours whereas after functionalization, selective sensing to ethanol is depicted. Minimum detection by PSi sensors is portrayed at 100 ppm and that of functionalized sensors is at 10 ppm. Sensing mechanism is explained on the basis of surfaces and structures of both PSi and TiO_2. This study incites the importance of surface treatment of PSi for tuning the sensing properties and is useful in the development of selective alcohol sensors.

Azo-dye adsorption activity of iron(Ⅲ) loaded novolac-based network sorbents

Iron(Ⅲ) loaded novolac-based network adsorbents 1 and 2 were studied for efficient removal of azo-dye pollutants from aqueous solutions. The adsorption behavior was evaluated by using methyl orange and orange-G as model azo dyes. The effect of parameters such as contact time and initial dye concentration on the adsorption of azo dyes was studied. The results showed that loading of Fe(Ⅲ) onto the sorbent networks has noticeable effect on azo-dye sorption capacity. The adsorption equilibrium data were fitted to Freundlich isotherm model. Besides, the reusability of the dye loaded sorbents was investigated on adjusting pH of solutions.

Microbial Solubilization of Phosphorus from Nano Rock Phosphate

Phosphorus （P） is a major growth-limiting nutrient, and unlike the case of nitrogen （N）, there is no large atmospheric source that can be made biologically available. Moreover, P governs crucial role in plant as it stimulates root development and growth, gives plant rapid and vigorous start leading to better tillering and essential for many metabolic processes for seed formation. Soil microbes play very important role in bio-weathering and biodegradation. The microorganisms produce low molecular mass organic acids, which attack the phosphate structure and transform phosphorus from non-utilizable to the utilizable for the plants form. The test of the relative efficiency of isolated strains is carried out by selecting the microorganisms that are capable of producing a halo/clear zone on a plate owing to the production of organic acids into the surrounding medium. It is a well-known fact that as the particle size of rock phosphate decreases, the microbe mediated solubilization of rock phosphate increases in soil. In the present investigation, microbial solubilization of nano rock phosphate （〈 100 nm） particles was studied. Experimental results revealed that Pseudomonas striata solubilized 11.45% of the total P after 24 h of incubation from nano rock phosphate particles while 28.95% and 21.19% of the total P was solubilized by Aspergillus niger （black pigmented） and Aspergillus niger （green pigmented）, respectively. It was also observed that Aspergillus niger has the higher ability to dissolve Udaipur rock phosphate than Pseudomonas striata.

Software Cost Estimation Using Social Group Optimization

This paper introduces the integration of the Social Group Optimization(SGO)algorithm to enhance the accuracy of software cost estimation using the Constructive Cost Model(COCOMO).COCOMO’s fixed coefficients often limit its adaptability,as they don’t account for variations across *** fine-tuning these parameters with SGO,we aim to improve estimation *** train and validate our SGO-enhanced model using historical project data,evaluating its performance with metrics like the mean magnitude of relative error(MMRE)and Manhattan distance(MD).Experimental results show that SGO optimization significantly improves the predictive accuracy of software cost models,offering valuable insights for project managers and practitioners in the ***,the approach’s effectiveness may vary depending on the quality and quantity of available historical data,and its scalability across diverse project types and sizes remains a key consideration for future research.

Utilization of Nano Rock Phosphate by Maize （Zea mays L.） Crop in a Vertisol of Central India

An attempt was made in the present investigation to utilize the vast deposit of rock phosphate （RP） of India by preparing nano RP using modem tool of nano-science and nano-technology for field application to maize crop （Zea mays L.）. To evaluate the efficacy of different nano RP, a field experiment was conducted during kharif season （June to September） of 2012 at India Institute of Soil Science （IISS）, Bhopal, taking maize as a test crop. The experiment consisted of five treatments （control, NK （100%）, NPK （100%）, NK （I00%）＋ 60 kg P205） as nano RP through Udaipur nano RP （34% P205） and Udaipur nano RP （31% P205） were tested in randomized block design with four replications. The results showed that the shelling percentage, 1,000 grain weight, grain and stover yield, etc., were significantly higher in the maize plant treated with Udaipur nano RP （34% P205） as compared to the control, which was being at par with single super phosphate （SSP） treated plants. The total P content and its uptake were higher （0.65% and 40.29 kg/ha） in SSP treated plant, which was closely followed by Udaipur nano RP 34% （0.63% and 38.29 kg/ha） over the control （0.60% and 27.82 kg/ha）. The SSP treated plants had significantly highest apparent P recovery （47.98%） which was comparable to Udaipur nano RP （34% P205）, where as the Udaipur nano RP （31% P205） had the lowest. The additional N and K uptake by the plant was also observed with application of P sources, which were ranged from 16.5 kg/ha to 37.2 kg/ha and 27.9 kg/ha to 64.7 kg/ha under different P treatments. These results showed that crop utilization of P from nano RP was at par with that of P from SSP while yield response to P from nano RP was marginally lower than to P from SSP but serve as a cheaper source.

Objective:To record the fractional components of lipid and polyunsaturated fatty acids of Lingula anatina(***),a Precambrian intertidal benthic brachiopod,giving emphasis on-ω series group especially eicosapentaenoic acid(EPA)and docosahexaenoic acid(DHA)alongside assessing their biotransformation within the population and mangrove-estuarine associated ***:Different biological samples after being collected from three contrasting study sites ***,SII and SIII at Talsari(Longitude 87°5′E to 88°5′E and Latitude 20°30′N to 22°2′N)were stored at-20°C until *** lipids were extracted from each sample following Bligh and Dryer *** and conformation of fatty acids were done by following Ackman ***:On analyzing different collected samples,muscles of *** exhibited the highest amount of total lipids(2.95%)of which 54.03%belongs to phospholipid *** body parts of studied species contained appreciable and greater amount of EPA and DHA than α-linolenic ***:Different collected samples exhibited variabilities in respect of total lipids and its fractional fatty acid *** muscles of *** showed maximum storage of lipids and fatty *** occurrences of EPA and DHA in different body parts of *** are supposed to be due to the biotransformation process converting theα-linolenic acid from its primary food sources.

Effect of Copper Oxide Nano Particle on Seed Germination of Selected Crops

Nano particles are finding their way into the environment through deliberate and accidental actions, ecotoxicological properties and the risks of these nano particles have yet not been fully characterized. In this present investigation, experiments were carried out to know the effect of Cu oxide-nano particles （〈 50 nm） on germination and growth of seeds of soybean and chickpea. In both the crops, germination was not checked up to 2,000 ppm Cu （applied through Cu oxide-nano particles）, but the root growth was prevented above 500 ppm Cu. With increasing concentration of NPs, the elongation of the roots was severely inhibited as compared to that in control. In many cases root necrosis was occurred. Massive adsorption of Cu oxide-nano particles into the root system was responsible for the toxicity. A parallel experiment was also carried out to know the effect of copper sulphate solution on seed germination, above 200 ppm Cu, it restricted the germination of seeds, because of high salinity.
