AFM-compatible microfluidic platform for affinity- based capture and nanomechanical characterization of circulating tumor cells

Circulating tumor cells(CTCs)carried by the patient’s bloodstream are known to lead to the metastatic spread of *** is becoming increasingly clear that an understanding of the nanomechanical characteristics of CTCs,such as elasticity and adhesiveness,represents advancements in tracking and monitoring cancer progression and *** the present work,we describe a combined microfluidic–atomic force microscopy(AFM)platform that uses antibody–antigen capture to routinely isolate and nanomechanically characterize CTCs present in blood samples from prostate cancer *** introduce the reversible assembly of a microfluidic device and apply refined and robust chemistry to covalently bond antibodies onto its glass substrate with high density and the desired *** a result,we show that the device can efficiently capture CTCs from patients with localized and metastatic prostate cancer through anti-EpCAM,anti-PSA,and anti-PSMA antibodies,and it is suitable for AFM measurements of captured intact *** nanomechanically characterized,CTCs originating from metastatic cancer demonstrate decreased elasticity and increased deformability compared to those originating from localized *** the average adhesion of CTCs to the AFM tip surface remained the same in both the groups,there were fewer multiple adhesion events in metastatic CTCs than there were in their *** developed platform is simple,robust,and reliable and can be useful in the diagnosis and prognosis of prostate cancer as well as other forms of cancer.

Cerium Oxide Nanoparticles Protect against Oxaliplatin Induced Testicular Damage: Biochemical, Histological, Immunohistochemical, and Genotoxic Study

Oxaliplatin is a chemotherapeutic drug used for colorectal cancer treatment. The testicular toxic effect is one of its recorded toxicities which resulted in a few studies. Oxidative stress could be a direct cause of this testicular toxicity. Cerium oxide nanoparticles (CONPs) are optimistic antioxidants for applications in medicine. The aim of the work is to study the protective effect of CONPs on testicular toxicity induced by oxaliplatin in rats. Forty adult male albino rats were divided into 4 groups: Control group, CONPs group (60 mg/kg, 5 times/week), Oxaliplatin group (4 mg/kg, twice/week), and Oxaliplatin & CONPs group, for 4 weeks. Seventy-two hours after the last administration, blood samples were taken for hormonal levels and testes were used for both histopathology and immunohistochemical microscopic examination. Sperm smears were also performed and their results were statistically analyzed to detect any sperm abnormalities. Oxaliplatin increased MDA levels. SOD and GPx activity was decreased. GSH levels were decreased. Also, it decreased the sperm cell count and serum testosterone, and anti-Müllerian hormon. In the testicular sections, significant histopathology changes were seen and immunohistochemical examination confirmed these results. Upon supplementation of CONPs with oxaliplatin decreased MDA levels. SOD and GPx activity was increased, and GSH did not change. In testicular sections, normal morphology was seen. Also, there was an increase in the sperm cell count and serum testosterone anti-Müllerian with significant improvement of testicular architecture, and immunohistochemical examination confirmed these results. The utilization of CONPs produced significant protection against all of the above-mentioned changes.

Predominantly antibody deficiency(PAD)is the most prevalent form of primary immunodeficiency,and is characterized by broad clinical,immunological and genetic *** the current gold standard of whole exome sequencing for diagnosis,pathogenic gene variants are only identified in less than 20% of *** elucidation of the causal genes underlying PAD has provided many insights into the cellular and molecular mechanisms underpinning disease pathogenesis,many other genes may remain as yet undefined to enable definitive diagnosis,prognostic monitoring and targeted therapy of *** that many patients display a relatively late onset of disease presentation in their 2^(nd) or 3^(rd) decade of life,it is questionable whether a single genetic lesion underlies disease in all ***,combined effects of other gene variants and/or non-genetic factors,including specific infections can drive disease *** this review,we define(1)the clinical and immunological variability of PAD,(2)consider how genetic defects identified in PAD have given insight into B-cell immunobiology,(3)address recent technological advances in genomics and the challenges associated with identifying causal variants,and(4)discuss how functional validation of variants of unknown significance could potentially be translated into increased diagnostic rates,improved prognostic monitoring and personalized medicine for PAD patients.A multidisciplinary approach will be the key to curtailing the early mortality and high morbidity rates in this immune disorder.

Comparison of perceived pain and patients’ satisfaction with traditional local anesthesia and single tooth anesthesia: A randomized clinical trial

BACKGROUND Contemporary innovations in the area of local anesthesia have attempted to provide an absolutely pain free experience for *** the introduction of Computer-Controlled Local Anesthetic Delivery Systems to dentistry,many studies have compared its efficacy and safety to conventional ***,very few studies have compared single tooth anesthesia(STA)and traditional local *** To compare pain rating,changes in blood pressure,and heart rate during the local anesthetic *** secondary objectives were to measure the patients’level of satisfaction and the differences in anesthetic efficiency between the STA system and traditional local *** A randomized controlled trial was conducted and a total of 80 patients with dental restorative needs were enrolled for the *** patients were evaluated for their general physical status and oral clinical findings before *** regarding perceived pain,changes in heart rate and blood pressure,and patients’satisfaction was collected using an electronic data form and was analyzed using paired and unpaired *** No significant difference was noted in perceived pain(P=0.59)and systolic blood pressure(P=0.09)during anesthetic injection using both traditional and STA *** patients had a significantly higher heart rate during anesthesia,although a statistically significant difference was noted among the traditional anesthesia and the STA groups even before *** the restorative procedure,less pain was perceived by STA patients on the Wong-Baker FACES pain scale,which was statistically significant(P<0.001).Analyses of post-procedure patient responses showed that STA patients had a significantly better treatment experience and preferred to have the same method of injection in the future(P=0.04).CONCLUSION STA system can provide less painful and more comfortable restorative treatment procedures in comparison to the traditional infil

The minimum number of colors in d-distance colorings of G is its d-distance chromatic number, denoted by χd(G). In this paper, we give the exact value of χd(G) (d = 1, 2), for some types of generalized Petersen graphs P(n, k) where k = 1, 2, 3 and arbitrary n.

Background: Familial Mediterranean fever (FMF) is an autoinflammatory genetic disorder that associated with different genetic mutations. Frequency of clinical manifestation differs according to age group, geographic region and ethnic population. Objectives: To study the clinical manifestation of FMF in relation to genotype (M680I, M694V, M694I and V726A). Result: The main presentation of studied group was abdominal pain 65.9% (203), followed by fever 60.4% (186) patients. (Mutation M694V) was the commonest 47.6% (297), followed by (Mutation V726A) in 32.8% (169%), then (Mutation M6802) in 23.4% (121) lastly (Mutation M6941) was in 22.1% (114) patients. Fever was highly associated with mutation (V729A) and it was statistically significant (*p value 0.047). Conclusion: Abdominal pain and fever were the most common manifestation of FMF patients. (Mutation M694V), (Mutation V726A) were the most detected mutation. Third age group;fever was associated with genetic mutation (V726A), abdominal pain with (M6941).

A LSA Based Image Classification Framework Utilizing Relative Spatial Arrangement

This paper focuses on the problem of automatic image classification （AIC） by proposing a framework based on latent semantic analysis （LSA） and image region pairs. The novel framework employs relative spatial arrangements for region pairs as the primary feature to capture semantics. The significance of this paper is twofold. Firstly, to the best our knowledge, this is the first study of the influence of region pairs as well as their relative spatial information in latent semantic analysis as applied to automatic image classification. Secondly, our proposed method for using the relative spatial information of region pairs show great promise in improving image semantic classi- fication compared with the classical latent semantic analysis method and 2D string representation algorithm.

Regulatory and activated effector T cells in chronic hepatitis C virus: Relation to autoimmunity

AIM To investigate how Tregs are regulated in chronic hepatitis C virus(HCV) patients via assessment of Tregs markers(granzyme 2, CD69 and FoxP3), Teffs markers [TNFRSF4(OX40), INFG] and CD4, CD25 genes. METHODS A prospective study was conducted on 120 subjects divided into 4 groups: Group Ⅰ(n = 30) treatment na?ve chronic HCV patients; Group Ⅱ(n = 30) chronic HCV treated with Peg/Riba; Group Ⅲ(n = 30) chronic HCV associated with non-organ specific autoantibody and Group Ⅳ(n = 30) healthy persons as a control group. Tregs and Teffs markers were assessed in peripheral blood mononuclear cells by quantitative real time reverse transcriptase-polymerase chain reaction. RESULTS Chronic HCV patients exhibited significant higher levels of both Teffs and Tregs in comparison to healthy control group. Tregs markers were significantly decreased in Peg/Riba treated HCV patients in comparison to treatment na?ve HCV group. In HCV patients with antinuclear antibody(ANA) +ve, Tregs markers were significantly decreased in comparison to all other studied groups. Teffs markers were significantly elevated in all HCV groups in comparison to control and in HCV group with ANA +ve in comparison to treatment na?ve HCV *** Elevated Tregs cells in chronic HCV patients dampen both CD4^+ and CD8^+ autologous T cell immune response. Interferon-α and ribavirin therapy suppress proliferation of Tregs. More significant suppression of Tregs was observed in HCV patients with autoantibodies favoring pathological autoimmune response.
