The mammalian epididymis not only plays a fun dame ntal role in the maturati on of spermatozoa,but also provides protecti on agai nst various *** foremost among these is the threat posed by oxidative stress,which arises from an imbalance in reactive oxygen species and can elicit damage to cellular lipids,proteins,and nucleic *** mice,the risk of oxidative damage to spermatozoa is mitigated through the expression and secretion of glutathione peroxidase 5(GPX5)as a major luminal scavenger in the proximal caput epididymidal ***,the loss of GPX5^-/-mediated protection leads to impaired DNA integrity in the spermatozoa of aged Gpx57 *** explore the underlying mechanism,we have conducted transcriptomic analysis of caput epididymidal epithelial cells from aged(13 months old)Gpx5^-/-m *** analysis revealed the dysregulation of several thousand epididymal mRNA transcripts,in eluding the downregulation of a subgroup of piRNA pathway gen es,in aged Gpx5^-/-*** agreeme nt with these fin dings,we also observed the loss of piRNAs,which potentially bind to the P-element-induced wimpy testis(PlWI)-like proteins PIWIL1 and *** absence of these piRNAs was correlated with the elevated mRNA levels of their putative gene targets in the caput epididymidis of Gpx5^-/-***,the oxidative stress response genes tend to have more targeting piRNAs,and many of them were among the top increased genes upon the loss of GPX5^-/-.Taken together,our findings suggest the existence of a previously uncharacterized somatic piRNA pathway in the mammalian epididymis and its possible invoIvement in the aging and oxidative stress-mediated responses.

A critical unaddressed problem in Parkinson’s disease is the lack of therapy that slows or hampers *** medications effectively manage symptoms,they offer no long-term benefit because they fail to address the underlying neuronal *** highlights that the elusive goals of halting progression and restoring damaged neurons limit the long-term impact of current *** clinical trials using gene therapy have demonstrated the safety of various vector delivery systems,dosages,and transgenes expressed in the central nervous system,signifying tangible and substantial progress in applying gene therapy as a promising Parkinson’s disease ***,at diagnosis,many dopamine neurons remain in the substantia nigra,offering a potential window for recovery and *** propose that modulating these surviving dopamine neurons and axons in the substantia nigra and striatum using gene therapy offers a potentially more impactful therapeutic approach for future ***,innovative gene therapies that focus on preserving the remaining elements may have significant potential for enhancing long-term outcomes and the quality of life for patients with Parkinson’s *** this review,we provide a perspective on how gene therapy can protect vulnerable elements in the substantia nigra and striatum,offering a novel approach to addressing Parkinson’s disease at its core.

Dengue and its effects on liver

Dengue has emerged as an important arboviral disease with significant impact on the disease burden in population residing in tropical countries. Dengue is spread by the bite of Aedes mosquito. The virus seems to have some hepatotoxic effects. Affliction of liver in form of derangements in the liver function tests is common and may include mild elevations in serum bilirubin, elevated transaminases and derangements in serum albumin. Although asymptomatic in most cases, clinical manifestations like jaundice, and acute liver failure(ALF) may occasionally complicate the clinical picture. Indeed, dengue has been implicated as an important cause of ALF in endemic countries. The present review focuses on the hepatic manifestations and the pathogenesis of the liver injury in dengue.

Analysis of a hand-foot-mouth disease model

In this paper, we have introduced a six-compartmental epidemic model with hand, foot and mouth disease （HFMD） infection. The total population is divided into six subclasses, namely susceptible, exposed, infective in asymptomatic phase, infective in symptomatic phase, quarantined and recovered class. Some basic properties such as boundedness and non-negativity of solutions are discussed. The basic reproduction number （R0） of the system is obtained using next generation matrix method. Then the deterministic dynamical behaviors of the system are studied. Our study includes the existence and stability analysis of equilibrium points of the system. The sensitivity analysis of our system helps us to find out the parameters of greater interest. Next, we deal with the epidemic model with three controls （two treatment controls with quarantine control）. We show that there exists an optimal control, which is effective in controlling the disease outbreak in a cost effective way. Numerical simulation is presented with the help of MATLAB, which shows tile reliability of our model from the practical point of view.

Proteomic signatures of infertile men with clinical varicocele and their validation studies reveal mitochondrial dysfunction leading to infertility

To study the major differences in the distribution of spermatozoa proteins in infertile men with varicocele by comparative proteomics and validation of their level of expression. The study-specific estimates for each varicocele outcome were combined to identify the proteins involved in varicocele-associated infertility in men irrespective of stage and laterality of their clinical varicocele. Expression levels of 5 key proteins （PKARIA, AK7, CCT6B, HSPA2, and ODF2） involved in stress response and sperm function including molecular chaperones were validated by Western blotting. Ninety-nine proteins were differentially expressed in the varicocele Eroup. Over 87% of the DEP involved in major energy metabolism and key sperm functions were underexpressed in the varicocele group. Key protein functions affected in the varicocele group were spermatogenesis, sperm motility, and mitochondrial dysfunction, which were further validated by Western blotting, corroborating the proteomics analysis. Varicocele is essentially a state of energy deprivation, hypoxia, and hyperthermia due to impaired blood supply, which is corroborated by down-regulation of lipid metabolism, mitochondrial electron transport chain, and Krebs cycle enzymes. To corroborate the proteomic analysis, expression of the 5 identified proteins of interest was validated by Western blotting. This study contributes toward establishing a biomarker ＂fingerprint＂ to assess sperm quality on the basis of molecular parameters.

Chemical, mechanical, and thermal expansion properties of a carbon nanotube-reinforced aluminum nanocomposite

In the present study,the chemical and mechanical properties and the thermal expansion of a carbon nanotube（CNT）-based crystalline nano-aluminum（nano Al） composite were *** properties of nanocomposites were tailored by incorporating CNTs into the nano Al matrix using a physical mixing *** elastic moduli and the coefficient of thermal expansion（CTE） of the nanocomposites were also estimated to understand the effects of CNT reinforcement in the Al *** characterization of the nanocomposite reveals that the CNTs are dispersed and embedded in the Al *** experimental results indicate that the incorporation of CNTs into the nano Al matrix results in the increase in hardness and elastic modulus along with a concomitant decrease in the coefficient of thermal expansion The hardness and elastic modulus of the nanocomposite increase by 21%and 20%,respectively,upon CNT *** CTE of CNT/A1 nanocomposite decreases to 70%compared with that of nano Al.

Stability analysis and optimal control of an epidemic model with vaccination

In this paper, we have developed a compartment of epidemic model with vaccination. We have divided the total population into five classes, namely susceptible, exposed, infective, infective in treatment and recovered class. We have discussed about basic properties of the system and found the basic reproduction number （R0） of the system. The stability analysis of the model shows that the system is locally as well as globally asymptotically stable at disease-free equilibrium E0 when R0 〈1. When R0 〉1 endemic equilibrium E1 exists and the system becomes locally asymptotically stable at E1 under some conditions. We have also discussed the epidemic model with two controls, vaccination control and treatment control. An objective functional is considered which is based on a combination of minimizing the number of exposed and infective individuals and the cost of the vaccines and drugs dose. Then an optimal control pair is obtained which minimizes the objective functional. Our numerical findings are illustrated through computer simulations using MATLAB. Epidemiological implications of our analytical findings are addressed critically.
