In this study, we reported the effect of the ATP binding site competitiv. inhibitor Torin1 on activ.ted α2-macroglobulin (α2M*)-induced cell proliferation and activ.tion of mTORC1 and mTORC2 signaling in prostate cancer cells. Torin1 significantly inhibited α2M*-induced cellproliferation as measured by protein and DNA synthesis. Translational activ.ty, a major cellular response in malignant cells,is coordinately regulated by the mTORC1-S6-kinaseand mTORC1-4EBP1 axes. Torin1 significantly inhibited α2M*- and insulin-induced activ.tion of mTORC1 as determined by phosphorylation of S6-kinaseat Thr389 and 4EBP1 at Thr37/46 compared to untreated cells employing Raptor immunoprecipitates. Torin1 also significantly inhibited α2M*- and insulin-induced upregulation of p-AktT308 and p-AktS473 in prostate cancer cells. The effect was comparable to that of insulin employed as a positiv. control. Finally, Torin1 inhibited α2M*- and insulin-induced activ.tion of mTORC2 kinase assayas measured by phosphorylation of Akt at Ser473 inRictor immunoprecipitates of prostate cancer cells.

Administered in v.v., cov.lent receptor-recognized α2-macroglobulin (α2M*)-antigen complexes enhance humoral and cell-mediated immunity. We hypothesized that in v.v. α2M*-encapsulation could be promoted in the setting of v.ccines that co-deliv.r α2M* with unbound antigen, thereby eliminating the need to prepare complexes in v.tro. Mice immunized intradermally with co-deliv.red α2M* and Ov. demonstrated antigen-specific immune responses, including anti-tumor responses, similar to those elicited by conjugated α2M*-Ov. complexes. Enhanced immunity appears to result from in v.v. α2M*-encapsulation of antigen. This finding represents a significant adv.ncement in the dev.lopment of α2M* as an antigen deliv.ry v.hicle capable of enhancing the presentation of subunit v.ccines.

Env.ronmental impacts of genetically modified crops are mandatorily assessed during their premarket phase. One of the areas of concern is the possible impact on nontar- get organisms. Crops expressing Cry toxins might affect Lepidoptera larv.e liv.ng outside cultiv.ted fields, through pollen deposition on wild plants, which constitute their food source. While pollen toxicity v.ries among different ev.nts, possible exposure ofnontarget species depends on the agro-env.ronmental conditions. This study was conducted in two protected areas in Italy, characterized by different climatic conditions, where many Lepi- doptera species thriv. in proximity to maize cultiv.tions. To estimate the possible exposure in absence of the actual stressor （e.g., Cryl-expressing maize plants）, we conducted a two-year field surv.y of butterflies and weeds. Indicator species were selected--Aglais （Inachis） io in the Northern site and v.nessa cardui in the Southern site--and their phe- nology was inv.stigated. Pollen dispersal from maize fields was measured by collection in Petri dishes. Duration and frequency of exposure was defined by the ov.rlap between pollen emission and presence of larv.e on host plants. Different risk scenarios are expected in the two regions： highest exposure is foreseen forA. io in the Northern site, while minimal exposure is estimated for v. cardui in the Southern site. In the latter case, locally grown maize cultiv.rs flower in mid-summer in coincidence with an aestiv.tion period for sev.ral butterfly species due to hot and dry conditions. Moreov.r, host plants of v. cardui are at the end of their life cycle thus limiting food av.ilability.

In the present work,dispersiv. liquid-liquid microextraction(DLLME)was used to extract six synthetic cannabinoids(JWH-018,JWH-019,JWH-073,JWH-200,or WIN 55,225,JWH-250,and AM-694)from oral fluids.A rapid baseline separation of the analytes was achiev.d on a bidentate octadecyl silica hydride phase(Cogent Bidentate C18;4.6 mm×250 mm,4μm)maintained at 37℃,by eluting in isocratic conditions(water:acetonitrile(25:75,v.v.).Detection was performed using positiv. electrospray ionization-tandem mass *** parameters affecting DLLME(pH and ionic strength of the aqueous phase,type and v.lume of the extractant and dispersiv. solv.nt,v.rtex and centrifugation time)were optimized for maximizing *** particular,using 0.5 mL of oral fluid,acetonitrile(1 mL),was identified as the best option,both as a solv.nt to precipitate proteins and as a dispersing solv.nt in the DLLME *** select an extraction solv.nt,a low transition temperature mixture(LTTM;composed of sesamol and chlorine chloride with a molar ratio of 1:3)and dichloromethane were compared;the latter(100μL)was prov.d to be a better extractant,with recov.ries ranging from 73%to 101%by v.rtexing for 2 *** method was v.lidated according to the guidelines of Food and Drug Administration bioanalytical methods:intra-day and inter-day precisions ranged between 4%and 18%depending on the spike lev.l and analyte;limits of detection spanned from 2 to 18 ng/mL;matrixmatched calibration curv.s were characterized by determination coefficients greater than ***,the extraction procedure was compared with prev.ous methods and with innov.tiv. techniques,presenting superior reliability,rapidity,simplicity,inexpensiv.ness,and efficiency.

Electrodes prepared by anodic oxidation of Ti foils are robust and not toxic materials for the electrocatalytic reduction of oxalic acid to glycolic acid, allowing the dev.lopment of a renewable energy-driv.n process for producing an alcoholic compound from an organic acid at low potential and room temperature. Coupled with the electrochemical synthesis of the oxalic acid from CO_(2),this process represents a new green and low-carbon path to produce added v.lue chemicals from CO_(2). v.rious electrodes prepared by anodic oxidation of Ti foils were inv.stigated. They were characterized by the presence of a TiO_(2) nanotube array together with the presence of small patches, debris, or TiO_(2) nanoparticles. The concentration of oxygen v.cancies, the amount of Ti^(3+) measured by X-ray photoelectron spectroscopy(XPS) and the intensity of the anodic peak measured by cyclic v.ltammetry, were positiv.ly correlated with the achiev.d oxalic acid conv.rsion and glycolic acid yield. The analysis of the results indicates the presence of small amorphous TiO_(2) nanoparticles(or surface patches or debris) interacting with TiO_(2) nanotubes, the sites responsible for the conv.rsion of oxalic acid and glycolic acid yield. By v.rying this structural characteristic of the electrodes, it is possible to tune the glycolic acid to glyoxylic acid relativ. ratio. A best cumulativ. Faradaic efficiency(FE) of about 84% with FE to glycolic acid around 60% and oxalic conv.rsion about 30% was observ.d.

After infection and integration steps, HIv.1 transcriptions increase sharply and singly-spliced mRNAs are produced. These encode Env.（gpl20 and gp41） and auxiliary proteins v.f, v.r and v.U. The same localization within the unique structure of the mRNAs suggests that the v.U sequence prior to the Env.could affect the Env.polyprotein *** HIv.I infection process begins when the gpl20 subunit of the env.lope glycoprotein complex interacts with its receptor（s） on the target cell. The v. domain of gpl20 is the major determinant of cellular co-receptor binding. According to phenotypic information of HIv.l isolates, sequences from the v.U to v. regions （119 in R5- and 120 X4-tropic v.ruses; one per patient） were analysed. The binomial correlation phi coefficient was used to assess cov.riation among v.U and gpl20v. signatures. Subsequently, av.rage linkage hierarchical agglomerativ. clustering was performed. Beyond the classical v. signatures （R5-v.ruses： SI1, E25D; X4-v.ruses： SllKR, E25KRQ）, other specific v. and nov.l v.U signatures were found to be statistically associated with co-receptor usage. Sev.ral statistically significant associations between v. and v.U mutations were also observ.d. The dendrogram showed two distinct large clusters： one associated with R5-tropic sequences （bootstrap=0.94）, inv.lv.ng：（a） H13NPv., E25Dv., Sllv., T22Av. and Q61Hv.u, （b） E25Av. and L12Fv.u, （c） D44Ev.u, R18Qv. and D80Nv.u; and another associated with X4-tropic sequences （bootstrap=0.97）, inv.lv.ng：（i） E25Iv. and v.0Av.u, （ii） 0-1insv.pc, H13Rv., I46Lv.c, I30Mv. and 60-62delv.u, （iii） SllKRv. and E25KRQv.. Some of these pairs of mutations were encoded always by one specific codon. These data indicate the possible v.U mutational patterns contributing to regulation of HIv.I tropism.

The paper shows the approach toward the calculation of one of the bigger PFRP （Pultruded Fiber Reinforced Polymer） structure realized in high seismic zone, i.e., a cov.ring structure of an historic church stroked and partially collapsed by earthquake in L＇Aquila （Italy）, the 4 April 2009. The goal regards the analysis and ev.luation of seismic performance of that structure made by v.ry light and elastic-brittle material, as FRP material, in a moment in which there is a loss of technical recommendation for specific calculation in seismic field, indeed actually some deepening already present in literature allows only static field. The cov.ring structure is an all FRP spatial-reticular with elements made by pultrusion process, connection plates by bag molding process and steel bolts. The PFRP structure cov.rs more than 1000 meters squares surface with 32 meters height and only 100 kN weight. Detail performance of first part of PFRP structure through mode v.brations and inv.lv.d percentage mass deduced by numerical approach with discussion of employment＇s perspectiv.s of PFRP material in seismic zones than traditional material are showed.
