Minimal volume vitrification of epididymal spermatozoa results in successful in vitro fertilization and embryo development in mice

This study compared three cryopreservation protocols on sperm functions, IVF outcomes, and embryo development. Epididymal spermatozoa cryopreserved using slow-cooling （18% w/v raffinose, RS-C） were compared with spermatozoa vitrified using 0.25 M sucrose （SV） or 18% w/v raffinose （RV）. The motility, vitality, and DNA damage （TUNEL assay） of fresh control （FC） spermatozoa were compared with post-thawed or warmed RS-C, RV, and SV samples. Mouse oocytes （n = 267） were randomly assigned into three groups for insemination： RV （n = 102）, RS-C （n = 86）, and FC （n = 79）. The number and the proportion of two-cell embryos and blastocysts from each treatment were assessed. Sperm motility （P 〈 0.01） and vitality （P 〈 0.05） were significantly reduced after vitrification compared with slow-cooled spermatozoa. However, DNA fragmentation was significantly reduced in spermatozoa vitrified using sucrose （15 - 1.8% [SV] vs 26 - 2.8% [RV] and 27 - 1.2% [RS-C]; P 〈 0.01）. Although the number of two-cell embryos produced by RS-C, RV, and FC spermatozoa was not significantly different, the number of blastocysts produced from two-cell embryos using RV spermatozoa was significantly higher than FC spermatozoa （P = 0.0053）. This simple, small volume vitrification protocol and standard insemination method allows successful embryo production from small numbers of epididymal spermatozoa and may be applied clinically to circumvent the need for ICSI, which has the disadvantage of bypassing sperm selection.

A glasshouse pot experiment was conducted to investigate effects of the arbuscular mycorrhizal fungus Glomus mosseae on the growth of Vicia faba and toxicity induced by heavy metals （HMs）（Cu, Zn, Pb and Cd） in a field soil contaminated by a mixture of these metals. There was also uninoculation treatment （NM） simultaneously. Mycorrhizal （GM） plants have significantly increased growth and tolerance to toxicity induced by heavy metals compared with NM plants. P uptake was significantly increased in GM plants. Mycorrhizal symbiosis reduced the transportation of HMs fi＇om root to shoot by immobilizing HMs in the mycorrhizal, shown by increasing the ratios of HMs from root to shoot. Oxidative stress, which can induce DNA damage, is an important mechanism of heavy metal toxicity. GM treatment decreased oxidative stress by intricating antioxidative systems such as peroxidases and non-enzymic systems including soluble protein. The DNA damage induced by heavy metals was detected using comet assay, which showed DNA damage in the plants was decreased by the GM treatment.

For the design and optimization of functional peptides, unravelling the structures of individual building blocks as well as the properties of the ensemble is paramount. TI＇R1, derived from human transthyretin, is a fibril-forming peptide implicated in diseases such as familial amyloid polyneuropathy and senile systemic amyloidosis. The functional peptide TTR1-RGD, based on a TFR1 scaffold, was designed to specifically interact with cells. Here, we used scanning tunneling microscopy （STM） to analyze the assembly structures of TTRl-related peptides with both the reverse sequence and the modified forward sequence. The site- specific analyses show the following： i） The TIR1 peptide is involved in assembly, nearly covering the entire length within the ordered [3-sheet structures, ii） For TTR1-RGD peptide assemblies, the TTR1 motif forms the ordered [3-sheet while the RGDS motif adopts a flexible conformation allowing it to promote cell adhesion. The key site is clearly identified as the linker residue Gly13. iii） Close inspection of the forward and reverse peptide assemblies show that in spite of the difference in chemistry, they display similar assembling characteristics, illustrating the robust nature of these peptides, iv） Glycine linker residues are included in the ^-strands, which strongly suggests that the sequence could be optimized by adding more linker residues. These garnered insights into the assembled structures of these peptides help unravel the mechanism driving peptide assemblies and instruct the rational design and optimization of sequence- programmed peptide architectures.

When discussing the tradition of ancient Chinese philosophy the intellectual community tends to focus their attention on the diachronic historical sources and pay less attention to the spatial regional characteristics. In ancient China communications were not developed and the dissemination of philosophical knowledge was greatly restricted by geographical conditions. Moreover, the spread of ancient philosophical knowledge depended mainly on private tutoring, much like the handing down of handicraft skills. Therefore, different schools of philosophy with regional traditions evolved with flourishing lecturing places （academies of classical learning during the Song and Ming dynasties and afterwards） as their centers. The formation of various schools of thought during pre-Qin times was closely linked with regional cultural traditions.

Precise control over the charge carrier dynamics throughout the device can result in outstanding performance of perovskite solar cells(PSCs).Poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate)(PEDOT:PSS)is the most actively studied hole transport material in p-i-n structured ***,charge transport in the PEDOT:PSS is limited and inefficient because of its low conductivity with the presence of the weak ionic conductor *** addition,morphology of the underlying PEDOT:PSS layer in PSCs plays a crucial role in determining the optoelectronic quality of the active perovskite absorber *** work is focused on realization of a non-wetting conductive surface of hole transport layer suitable for the growth of larger perovskite crystalline *** is accomplished by employing a facile solventengineered(ethylene glycol and methanol)approach resulting in removal of the predominant PSS in PEDOT:*** consequence of acquiring larger perovskite crystalline domains was observed in the charge carrier dynamics studies,with the achievement of higher charge carrier lifetime,lower charge transport time and lower transfer impedance in the solvent-engineered PEDOT:PSS-based *** of this solventengineered treatment for the fabrication of MAPbI3 PSCs greatly increased the device stability witnessing a power conversion efficiency of 18.18%,which corresponds to^37%improvement compared to the untreated PEDOT:PSS based devices.

What turned out to be an object of strongest public condemnation in modern education in China was the mechanical copying of foreign experience without any focus on its real needs. In 1928, the adoption of Three People＇s Principles as the ruling ideology of the Kuomintang and the educational purpose around these principles was an attempt on the part of the government at the establishment of dominant ideology and its legitimate authority, but with no marked effects. This was considered one of the most important reasons for the Kuomintang to lose the mainland to the Communist Party. An analysis of the relations between the education on national centrism and the education on the Three People＇s Principles in three different stages will reveal the troubled destiny of the latter.
