Shedding of TNFR1 in regenerative liver can be induced with TNF α and PMA

AIM: Liver regeneration is associated with apoptosis of hepatocytes, which is mediated via tumor necrosis factor receptor 1(TNFR1). The shedding of TNFR1 in liver regeneration and its mechanism to regulate this shedding were ***: The shedding of TNFR1 in liver regeneration and changes of TNF-α, PMA and plasma membrane purified from hepatocytes on this shedding process were measured with Western blot. Then, the relationship between TNFR1 shedding and apoptosis of hepatocytes induced by TNFα was studied by detecting apoptotic ***: The shedding of TNFR1 began at 4 hours and terminated before 2 months after partial hepatectomy. In culture system, serum from rats at 36 h after partial hepatectomy could also promote this shedding process. With the stimulation of TNF α, PMA or purified plasma membrane from hepatocytes at 36 h after partial hepatectomy or from hepatocytes treated with TNF α for 2 h, membranous TNFR1 was also shed. With the stimulation of both TNF α and plasma membrane from hepatocytes affected with TNF α for 2 hor from hepatocytes at 36 h after partial hepatectomy, apoptotic index of hepatocytes decreased from 21% to 7.52 % and 8.45 %, respectively. PMA could also reduce apoptotic index to 13.67 %. This descent occurred in hepatocytes cultured in serum from rats at 36 h after partial hepatectomy too,but not in serum from rats at 2 months after partial hepatectomy and sham-operated ***: Shedding of TNFR1 may help reduce apoptosis of hepatocytes induced by TNFα. Membraneanchored metalloprotases could play a role in shedding membranous TNFR1. At the same time, PKC may take part in regulation of this shedding process.

Platinum modification of metallic cobalt defect sites for efficient electrocatalytic oxidation of 5-hydroxymethylfurfural

Co3O4possesses both direct and indirect oxidation effects and is considered as a promising catalyst for the oxidation of 5-hydroxymethylfurfural（HMF）.However,the enrichment and activation effects of Co3O4on OH-and HMF are weak,which limits its further *** defect engineering can regulate the electronic structure,optimize the adsorption of intermediates,and improve the catalytic activity by breaking the symmetry of the material,which is rarely involved in the upgrading of *** this work,we prepare Co3O4with metal defects and load the precious metal platinum at the defect sites（PtVco）.The results of in-situ characterizatio ns,electrochemical measurements,and theoretical calculations indicate that the reduction of Co-Co coordination number and the formation of Pt-Co bond induce the decrease of electron filling in the antibonding orbitals of Co *** resulting upward shift of the d-band center of Co combined with the characteristic adsorption of Pt species synergically enhances the enrichment and activation of organic molecules and OH species,thus exhibiting excellent HMF oxidation activity(including a lower onset potential（1.14 V） and 19 times higher current density than pure Co3O4at 1.35 V).In summary,this work explores the adsorption enhancement mechanism of metal defect sites modified by precious metal in detail,provides a new option for improving the HMF oxidation activity of cobalt-based materials,broadens the application field of metal defect based materials,and gives an innovative guidance for the functional utilization of metal defect sites in biomass conversion.

基于隐藏的动态视角,探讨不同在线广告与消费者无法被直接观测的广告响应度变化之间的关系。利用中国一家大型电子商务企业的相关数据,运用连续隐马尔可夫模型分析不同广告渠道下消费者群体的行为特征以及广告响应动态转移路径。研究发现:大多数在线广告均能显著影响消费者广告响应度的动态转移;消费者发起的广告相对于企业发起的广告,对消费者广告响应的提升作用更明显;消费者发起的广告渠道下消费者的非交易行为表现基本一致,而企业发起的广告下消费者的非交易行为有较大的差异。研究从动态转移视角揭示不同在线广告产生不同效果的深层原因,为企业有效投放在线广告提供一定指导。

非自杀性自伤(Non-suicidal Self-injury,NSSI)是重大的公共卫生问题,具有高度污名化、高度复杂性和高度异质性的特点,传统NSSI研究测量和分析方法有限,获得的影响因子预测力较低。近年来机器学习逐步应用于NSSI的分析和建模中,并通过提高NSSI研究工具预测力、增加预测模型复杂度和精确度、区分NSSI类别和亚型,使整体预测性能上升到中等水平。未来需结合NSSI传统理论和研究方法使筛选标准更严格,拓展非问卷NSSI数据与深度学习、无监督学习的结合,根据“先分型、再迁移”的原则增加模型的可复制性与可比性,进一步提高预测性能。

Immune evasion behavior and immunosuppressive characteristics of tumor extensively impedethe immune initiation effect of therapy triggered immunogeniccell death (ICD). In this work, a carrier-adjuvantedimmunostimulator (designated as CoCeC) is developed toboost photodynamic immunotherapy by downregulatingprogrammed death ligand 1 (PD-L1) and impairing adenosinetriphosphate (ATP) hydrolysis. Among these, the crosslinkedchitosan oligosaccharide is applied as the drug carrier fordelivery of Ce6 and Ceritinib, which also serves as an immuneadjuvant to downregulate PD-L1. Meanwhile, the robustphotodynamic therapy (PDT) of CoCeC exhibits lethal toxicityagainst tumor cells to induce ICD and release damage-associated molecular patterns (DAMPs), which can also impairATP hydrolysis by blocking CD39. In vitro and in vivo resultsdemonstrate the robust therapeutic efficacy of CoCeC tosuppress primary tumor growth and activate a superior immune elimination against lung metastasis by amplifying theimmune initiation of ICD with the assistance of immune *** work provides a self-adjuvanted strategy to enhance the immune response of therapy induced ICD, which ispromising to activate systemic antitumor immunity in consideration of the complicated immunosuppressive factors.

The classic cartoon “Calabash Brothers” describes a story of seven brothers born from a magic gourd uniting to defeat powerful *** order to explore the secret of the magic gourd,several transition metals were used to synthesize metal silicates（CMSi） by planted gourd leaves（GLs） and then the C-MSi materials were used to fabricate supercapacitor electrodes and devices with superior electrochemical *** integrating theoretical calculations and experimental results,the supercapacitor electrodes and devices obtained from the combination of transition metals with amorphous carbon exhibit superior electrochemical *** detail,in the three-electrode system,the Na OH etched materials（C-MSi） exhibited better electrochemical performance(for instance,as for C-CdSi,607 F g-1at 0.5 A g-1and the capacitance retention of 98.2% after 10,000cycles) than the unetched ones（i-C-MSi）.Hybrid supercapacitor（HSC） devices also achieve very excellent electrochemical *** C-CdSi//AC as an example,the areal specific capacitance with 691 mF cm-2at 2 mA cm-2,the energy density with 5.04 Whm-2at the power density of 22.2 Wm-2and the cycle stability with 87.3% after 6,000 *** approach is very versatile and was also applied to produce many hierarchically structured metal-silicate materials of other biomass precursors,including roots,vines,flowers,fruits and seeds of the planted ***,it is a potential way to prepare transition metal silicates using biomaterials for the enhancement of electrochemical performance and improvement of energy storage and ***,this paper preliminarily reveals the secret of the magic gourd.

Parallel Optimization of the Crystal-KMC on Tianhe-2

The Kinetic Monte Carlo(KMC)is one of the commonly used methods for simulating radiation damage of *** team develops a parallel KMC software named Crystal-KMC,which supports the Embedded Atom Method(EAM)potential energy and utilizes the Message Passing Interface(MPI)technology to simulate the vacancy transition of the Copper(Cu)element under neutron *** make better use of the computing power of modern supercomputers,we develop the parallel efficiency optimization model for the Crystal-KMC on Tianhe-2,to achieve a larger simulation of the damage process of materials under irradiation ***,we analyze the performance bottleneck of the Crystal-KMC software and use the MIC offload statement to implement the operation of key modules of the software on the MIC *** use Open MP to develop parallel optimization for the Crystal-KMC,combined with existing MPI inter-process communication optimization,finally achieving hybrid parallel *** experimental results show that in the single-node CPU and MIC collaborative parallel mode,the speedup of the calculation hotspot reaches 30.1,and the speedup of the overall software reaches 7.43.

Secrecy has received tremendous attention in modern information society. Innovative polymer-based fluorescent materials with multiple mode emission are quite desirable to achieve multistage data security. Herein, a series of soluble and thermally stable hyperbranched poly(formyl-1,2,3-triazole)s(hb-PFTAs) with high molecular weights were facilely prepared by catalyst-free polycycloaddition of aldehyde-activated internal triyne and diazides. Functional moieties of triphenylamine and binaphthyl could be incorporated into the polymers to offer them with unique fluorescence and circular dichroism properties, *** taking advantage of activated ethynyl and aldehyde groups on their peripheries and in the internal branch chains, the hbPFTAs can undergo efficient single-and double-stage post-functionalization. More importantly, based on the remarkable fluorescence responses to hydrazine and hydrochloric acid, the hyperbranched polymers are featured with triple-mode fluorescent patterns and have been applied in quadruple-modal information encryption and storage with enhanced security and density. Collectively, this work not only enriches the structural diversity of the existing triazole-based material library, but also provides new polymeric materials for data and information security application.

目的探讨德谷胰岛素利拉鲁肽注射液(IDegLira)联合七叶皂苷钠片治疗糖尿病视网膜病变(DR)合并黄斑水肿(DME)患者的临床效果。方法选取榆林市中医医院2022年1月至2024年5月收治的DR合并DME患者120例进行随机对照试验。采用随机数字表法将其分为研究组与对照组,各60例。研究组男35例,女25例,年龄(58.49±7.84)岁,病程(14.39±2.05)年。对照组男32例,女28例,年龄(57.88±8.95)岁,病程(14.52±2.10)年。两组均接受常规治疗;对照组另给予七叶皂苷钠片治疗,研究组在对照组基础上联合IDegLira治疗。两组均治疗3个月。对比两组治疗后血糖达标率、低血糖发生率。比较两组治疗前后最小分辨角对数(logMAR)、眼压、黄斑中心凹视网膜厚度(CMT)。比较两组治疗后黄斑区渗透情况。比较两组治疗前后房水白细胞介素-6(IL-6)、血管内皮生长因子(VEGF)、单核细胞趋化蛋白(MCP-1)、一氧化氮合酶(NOS)、可溶性细胞间黏附分子-1(sICAM-1)水平。比较两组治疗有效率和不良反应发生情况。采用χ2检验、t检验进行统计分析。结果研究组血糖达标率高于对照组[85.00%(51/60)比48.33%(29/60),P0.05);治疗后,研究组logMAR、眼压、CMT均低于对照组(均P0.05);治疗后,研究组IL-6、VEGF、MCP-1、sICAM-1水平均低于对照组,NOS水平高于对照组(均P0.05]。结论IDegLira联合七叶皂苷钠片治疗DR合并DME患者能改善其血糖控制,减少低血糖事件发生,还能有效减少黄斑区渗漏,改善患者视力及房水指标,临床总疗效更佳,且安全。
