Antioxidant Properties and Glucan Compositions of Various Crude Extract from <i>Lentinus squarrosulus</i>Mycelial Culture

Various crude extracts of Lentinus squarrosulus mushroom mycelium and culture broth were prepared for antioxidant studies, glucan and carbohydrate compound analysis. Crude extract from ethanol, cold and hot water extraction was tested on several complementary test systems namely DPPH free radical scavenging, β-carotene linoleic acid oxidation, total phenolic compound, total flavonoid compound, total carbohydrate content and total glucan compositions. The antioxidant activity of the crude extracts of Lentinus squarrosulus mushroom mycelium and culture broth was determined by 2,2’ diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method and β-carotene linoleic acid assay. Crude extracted from culture broth EPS EE recorded the highest radical scavenging effects at 20 mg/mL (85%) which is closer to the control antioxidants (ascorbic acid 97%;BHT 92.4%;BHA 93.6%). Both the ethanol extract (IPS EE and EPS EE) exhibited the highest β-carotene linoleic acid oxidation by recording IC50 values of 0.6 and 0.65 mg/mL respectively. IPS EE (0.59 mg/g) and EPS WE (0.58 mg/g) showed high flavonoid content whereas total phenol content was high in EPS WE (0.85 mg/g) and IPS EE (0.84 mg/g). Total carbohydrate concentrations were high in IPS HWE and IPS CWE with 19.1 and 16.3 mg/mL respectively and these data correlate with the β-glucan content in both extracts. The highest β-glucan content found in IPS CWE and IPS HWE were 12.2 and 11.4% w/w respectively.

(-) Huperzine A is a bioactive alkaloid from Huperzia serrata (Lycopodiaceae) used for the treatment of Alzheimer's disease. High yielding (-) huperzine A species is of interest for mass propagation since it grows slowly in temperate countries. The content of (-) huperzine A from temperate countries was reported but none reported from tropical species. The aim of this study was to evaluate the content of (-) huperzine A from tropical club mosses and further identify a high yielding species. Club mosses from Lycopodiaceae family were collected from Peninsular Malaysia. The collected club mosses were dried, pulverized and extracted with methanol. A gradient reverse phase HPLC-photodiode array detector method with increasing amount of methanol in 0.01 % trifluoroacetic acid was developed. The calibration curve was linear from 5 to 100 g·mL-1 with correlation coefficient, r2, of 0.998 1. The precision for both intra-and inter-day peak area were between 0.48% to 1.24 % and 0.95% to 3.85 %, respectively. The recovery of the method was between 99.8% to 103.8 %. Though geographically segregated, (-) huperzine A content in Huperzia phlegmaria and *** found in the tropics was highest and similar to species in Australasia. Both species may provide a good source of (-) huperzine A.

New Bioactive Molecules with Potential Antioxidant Activity from Various Extracts of Wild Edible Gelam Mushroom (<i>Boletus</i>spp.)

Boletus spp. is a wild edible Gelam mushroom, which is only available at coastal area of Bachok, Kelantan. Locals have used this mushroom as a food source and as a traditional medicine. In this study, antioxidants were extracted using Mili-Q water and methanol. Extraction of 1.0 g of Boletus spp. mushroom pileus and stipe by water produced 0.22 ± 0.09 g and 0.3 ± 0.07 g of total aqueous soluble extracts, respectively. Methanol extraction of 1.0 g Boletus spp. mushroom pileus and stipe produced 0.31 ± 0.08 mg and 0.37 ± 0.05 mg of total methanol soluble extracts, respectively. DPPH assay has been used to determine the value of antioxidant efficiency of each extracts. DPPH assay revealed that the EC50 value of pileus aqueous soluble extract and both pileus and stipe methanol soluble extracts were 1.2 mg/ml. The EC50 value of stipe aqueous soluble extracts was 1.4 mg/ml. The EC50 value of ascorbic acid, an antioxidant control was 0.5 mg/ml. Liquid chromatography- DPPH (LC-DPPH) assay combined with Quadrupole Time-of-flight Mass Spectrometry (QTOF-MS) analysis of 3:2 ratio methanol and water extract has detected 43 antioxidant compounds that were involved in the reduction of DPPH. To identify these compounds, SIMCA 3.0 software that consists of PCA, OPLS-DA and s-plot analyses had been used. PCA analysis had shown the antioxidant activity from sample on DPPH as 73.6%. This antioxidant activity confirmed by OPLS-DA analysis had shown the variability between sample and control as 99.6% with 99.3% prediction. S-plot selection had shown ten antioxidant compounds significantly involved in the DPPH assay. Four of these compounds were identified by the METLIN and NIST databases. The antioxidant metabolites were 2,4,6-trimethylacetophenone imine, glutamyl tryptophan, azatadine and lithocholic acid glycine conjugate.
