Proposal for pathogenesis-based treatment options to reduce calcium oxalate stone recurrence

Objective:Prevalence of kidney stone disease continues to increase globally with recurrence rates between 30%and 50%despite technological and scientific *** in recurrence would improve patient outcomes and reduce cost and stone *** objective was to review results of experimental studies performed to determine the efficacy of readily available compounds that can be used to prevent ***: All relevant literature up to October 2020,listed in PubMed is ***: Clinical guidelines endorse the use of evidence-based medications,such as alkaline agents and thiazides,to reduce urinary mineral supersaturation and ***,there may be additional steps during stone pathogenesis where medications could moderate stone *** calcium oxalate stones grow attached to randall’s plaques or *** of clinical and experimental studies suggest involvement of reactive oxygen species and oxidative stress in the formation of both the plaques and *** renin-angiotensin-aldosterone system(rAAS),nicotinamide adenine dinucleotide phosphate(NADPH)oxidase,mitochondria,and NOD-like receptor pyrin domain containing-3(NLrP3)inflammasome have all been implicated at specific steps during stone pathogenesis in animal ***: In addition to supersaturation-reducing therapies,the use of anti-oxidants,free radical scavengers,and inhibitors of NADPH oxidase,NLrP3 inflammasome,and rAAS may prove beneficial for stone *** such as statins and angiotensin converting enzyme inhibitors are already in use as therapeutics for hypertension and cardio-vascular disease and have previously shown to reduce calcium oxalate nephrolithiasis in *** clinical evidence for their use in stone prevention in humans is limited,experimental data support they be considered along with standard evidence-based medications and clinical expertise when patients are being counselled for stone prevention.

Development and Application of a Validated UHPLC Method for the Determination of Atropine and Its Major Impurities in Antidote Treatment Nerve Agent Auto-Injectors (ATNAA) Stored in the Strategic National Stockpiles

The development and implementation of advanced analytical technologies is essential for extending the expiry for complex drug products stored in the Strategic National Stockpiles. Consequently, a novel Ultra High-Performance Liquid Chromatographic (UHPLC) method has been developed for the analysis of atropine and its respective impurities to support the analytical research platform for auto-injectors. This study is part of a larger research effort to improve the efficiency and broaden the applicability of advanced analytical methods for medical counter-measure medications. The current HPLC compendial methodology for atropine sulfate injection requires an analysis time of 40 minutes for atropine. In comparison, the novel gradient UHPLC method required only 8 minutes to evaluate both atropine and its major pharmaceutical impurities. Improved separation was achieved on a Waters Acquity UHPLC BEH C18 1.7 μm, 2.1 × 100 mm column employing gradient elution of mobile phase solvent A (0.1% H3PO4) and solvent B (0.1% H3PO4, 90% ACN, and 10% H2O). The method was validated according to USP Category I requirements for assay. The daily standard calibration curves were linear over a concentration range from 50 μg/mL to 250 μg/mL with a correlation coefficient of >0.999. The detection limit (LOD) and quantitation limit (LOQ) were 3.9 μg/ml and 13.1 μg/ml, respectively. resolution results indicate that atropine and the following impurities, degradants and a preservative can also be separated and analyzed using this proposed method: noratropine, 4,4’-di-hy-droxydiphenyl ether, 2,4’-dihydroxydiphenyl ether, 4-bromophenol, 4-hydro-xyatropine, tropic acid, apoatropine HCl, atropic acid, hydroquinone, nitroethane, phenol and catechol. The UHPLC method demonstrated enhanced selectivity and significantly reduced the analysis time when compared with the traditional USP compendial HPLC method. In this work the structural,electronic and thermoelectric properties of YbMg_(2)X_(2)(X=P,As,Sb,Bi)zintl compounds were investigated comprehensively using first principles *** electronic properties were studied using PBE GGA,TB-mBJ and hybrid(YS-PBE0)*** the structural parameters of optimized structures are in harmonious agreement to the available *** band structure study illustrates that the titled materials manifest metallic and semi metallic nature using PBE and TB-mBJ potentials while they show wide band gap semiconducting behavior by following hybrid(YS-PBE0)*** density of states(TDOS)and partial density of states(PDOS)were also calculated to glimpse the contribution of orbitals of atoms in the formation of *** properties were studied by using BoltzTraP2 code employed to *** get enormous values of Seebeck coefficient(S),power factor(PF)and thermoelectric figure of merit(ZT)for all the samples YbMg_(2)X_(2)(X=P,As,Sb,Bi),Moreover,the overwhelming transport properties for the titled compounds indicate the optimum level of carriers’concentration which pinpoints these materials to be better thermoelectrics in the 1-2-2 zintl family.

Dependence of effective screening length in granular columns on bead and silo sizes and their ratio

Apparent mass measurements at the bottom of silos have been carried out. An important parameter in the Janssen model known as the effective screening length has been investigated for different bead and silo diameters as well as of their ratios. It is found that the effective screening length augments with the grain diameter d in addition to the granular column size. It is also revealed that λ exhibits stronger correlation with the bead diameter than that of the silo. This phenomenon is attributed to the reduced-shielding of the vertical stresses to the horizontal ones.

Dynamical system of a SEIQV epidemic model with nonlinear generalized incidence rate arising in biology

In this paper, a dynamical system of a SEIQV mathematical model with nonlinear generalized incidence arising in biology is investigated. The stability of the disease-free and endemic equilibrium is discussed. The basic reproduction number of the model is obtained. We found that the disease-free and endemic equilibrium is stable locally as well as globally asymptotically stable. For r0〈1, the disease-free equilibrium is stable both locally and globally and for r0〉1, the endemic equilibrium is stable globally asymptotically. Finally, some numerical results are presented.

In this paper, an SEIVr epidemic model with generalized incidence and preventive vaccination is considered. First, we formulate the model and obtain its basic properties. Then, we find the equilibrium points of the model, the disease-free and the endemic equilibrium. The stability of disease-free and endemic equilibrium is associated with the basic reproduction number r0. If the basic reproduction number r0〈 1, the disease- free equilibrium is locally as well as globally asymptotically stable. Moreover, if the basic reproduction number r0 〉 1, the disease is uniformly persistent and the unique endemic equilibrium of the system is locally as well as globally asymptotically stable under certain conditions. Finally, the numerical results justify the analytical results.

Mathematical modeling approach to the transmission dynamics of pine wilt disease with saturated incidence rate

The present paper investigates the dynamics of pine wilt disease with saturated incidence rate. The proposed model is stable both locally and globally. The local stability of the disease-free equilibrium is determined by the basic reproduction r0. The disease-free equilibrium is stable locally and globally whenever r0〈 1. If r0 〉 1, then the endemic state is stable both locally and globally. Further, a brief discussion with conclusion on the numerical results of the proposed model is presented.
