Fucoidan Induces G_1 Phase Arrest and Apoptosis through Caspases-dependent Pathway and ROS Induction in Human Breast Cancer MCF-7 Cells

Fucoidan is an active component of seaweed, which inhibits proliferation and induces apoptosis of several tumor cells while the detailed mechanisms underlying this process are still not clear. In this study, the effect of Fucoidan on the proliferation and apoptosis of human breast cancer MCF-7 cells and the molecular mechanism of Fucoidan action were investigated. Viable cell number of MCF-7 cells was decreased by Fucoidan treatment in a dose-dependent manner as measured by MTT assay. Fucoidan treatment resulted in G1 phase arrest of MCF-7 cells as revealed by flow cytometry, which was associated with the decrease in the gene expression of cyclin D 1 and CDK-4. Annexin V/PI staining results showed that the number of apoptotic cells was associated with regulation of cytochrome C, cas- pase-8, Bax and Bcl-2 at transcriptional and translational levels. Both morphologic observation and Hoechst 33258 assay results confirmed the pro-apoptotic effect of Fucoidan. Meanwhile, the ROS pro- duction was also increased by Fucoidan treatment, which suggested that Fucoidan induced oxidative damage in MCF-7 cells. The results of present study demonstrated that Fucoidan could induce GI phase arrest and apoptosis in MCF-7 cells through regulating the cell cycle and apoptosis-related genes or proteins expression, and ROS generation is also involved in these processes.

Summary： Fucoidan is one of the main bioactive components of polysaccharides. The current study was focused on the anti-tumor effects of fucoidan on human heptoma cell line HepG2 and the possible mechanisms. Fucoidan treatment resulted in cell cycle arrest and apoptosis of HepG2 cells in a dose-dependent manner detected by MTT assay, flow cytometry and fluorescent microscopy. The results of flow cytometric analysis revealed that fucoidan induced G2/M arrest in the cell cycle progression. Hoechst 33258 and Annexin V/PI staining results showed that the apoptotic cell number was increased, which was associated with a dose-dependent up-regulation of Bax and down-regulation of Bcl-2 and p-Stat3. In parallel, the up-regulation of p53 and the increase in reactive oxygen species were also observed, Which may play important roles in the inhibition of HepG2 growth by fucoidan. In the meantime, Cyelin B 1 and CDK1 were down-regulated by fucoidan treatment. Down-regulation of p-Stat3 by fucoidan resulted in apoptosis and an increase in ROS in response to fucoidan exposure. We therefore concluded that fucoidan induces apoptosis through the down-regulation of p-Stat3. These results suggest that fucoidan may be used as a novel anti-cancer agent for hepatocarcinoma.

Germacrone Induces Apoptosis in Human Hepatoma HepG2 Cells through Inhibition of the JAK2/STAT3 Signalling Pathway

Summary： Previous studies have shown that STAT3 plays a vital role in the genesis and progression of cancer. In this study, we investigated the relationship between the JAK2/STAT3 signalling pathway and germacrone-induced apoptosis in HepG2 cells. HepG2 cells were incubated with germacrone for 24 h, the protein expression of p-STAT3, STAT3, p-JAK2 and JAK2 was detected by Westem Blotting, and RT-PCR was used to determine the expression of STAT3, p53, Bcl-2 and Bax at transcriptional levels. Besides that, HepG2 cells were pre-treated with AG490 or IL-6 for 2 h, and then incubated with ger- macrone for 24 h. The expression ofp-JAK2, JAK2, p-STAT3, STAT3, p53, Bax and Bcl-2 was detected by Western blotting. The activity of HepG2 cells was tested by MTT assay. The apoptosis of HepG2 cells and levels of reactive oxygen species （ROS） were flow cytometrically measured. The results showed that germacrone exposure decreased p-STAT3 and p-JAK2 and regulated expression of p53 and Bcl-2 family members at the same time. Moreover, IL-6 enhanced the activation of the JAK2/STAT3 signalling pathway and therefore attenuated the germacrone-induced apoptosis. Suppression of JAK2/STAT3 signalling pathway by AG490, an inhibitor of JAK2, resulted in apoptosis and an increase in ROS in response to germacrone exposure. We therefore conclude that germacrone induces apoptosis through the JAK2/STAT3 signalling pathway.

Flow of Burgers' fluid over an inclined stretching sheet with heat and mass transfer

Effects of heat and mass transfer in the flow of Burgers fluid over an inclined sheet are discussed. Problems formulation and relevant analysis are given in the presence of thermal radiation and non-uniform heat source/sink. Thermal conductivity is taken temperature dependent. The nonlinear partial differential equations are simplified using boundary layer approximations. The resultant nonlinear ordinary differential equations are solved for the series solutions. The convergence of series solutions is obtained by plotting theη-curves for the velocity, temperature and concentration fields. Results of this work describe the role of different physical parameters involved in the problem. The Deborah numbers corresponding to relaxation time(β1 and β2) and angle of inclination(α) decrease the fluid velocity and concentration field. Concentration field decays as Deborah numbers corresponding to retardation time(β3) and mixed convection parameter(G) increase. Large values of heat generation/absorption parameters A/B, and the temperature distribution across the boundary layer increase. Numerical values of local Nusselt number,-θ′(0), and local Sherwood number,-f′(0), are computed and analyzed. It is found that θ′(0) increases with an increase in β3.

Traditional medicine systems around the globe,like Unani,Ayurveda and traditional Chinese medicine,include a number of sugar-based formulations,which contain a large amount of saccharide-containing sweetener,such as honey,sucrose or *** pervasive lifestyle disorders throughout the world,there have been discussions to consider alternative sweetening ***,from the perspective of Unani medicine,we discuss how the saccharide-based sweeteners may be an essential component of these traditional preparations,like electuaries,which may be deprived of their bioactivities without these *** contemporary researches,it is known that apart from their own therapeutic effects,saccharides also form deep eutectic solvents which help in enhancing the bioactivity of other ingredients present in crude *** addition,they provide energy for fermentation which is essential for biotransformation of ***,the sugars also increase the shelf-life of these compound drugs and act as natural *** the basis of this review,we strongly believe that saccharide-based sweeteners are an essential component of traditional medicines and not merely an excipient.

Analysis for flow of Jeffrey fluid with nanoparticles

An analysis of the boundary layer flow and heat transfer in a Jeffrey fluid containing nanoparticles is presented in this paper. Here, fluid motion is due to a stretchable cylinder. The thermal conductivity of the fluid is taken to be temperature-dependent. The partial differential equations of velocity, temperature, and concentration fields are transformed to a dimensionless system of ordinary differential equations. Nonlinear governing analysis is computed for the homotopy solutions. The behaviors of Brownian motion and thermophoresis diffusion of nanoparticles have been examined graphi- cally. Numerical values of the local Nusselt number are computed and analyzed.

A CFD study of the transport and fate of airborne droplets in a ventilated office:The role of droplet−droplet interactions

Previous studies reported that specially designed ventilation systems provide good air quality and safe environment by removing airborne droplets that contain viruses expelled by infected *** water droplets can be stable in the environment and remain suspended in air for prolonged *** between droplets may occur and droplet interactions should be ***,the previous studies focused on other physical phenomena(air flow,drag force,evaporation)for droplet transport and neglected droplet *** this work,we used computational fluid dynamics(CFD)to simulate the transport and fate of airborne droplets expelled by an asymptomatic person and considered droplet *** drag with turbulence for prediction of transport and fate of droplets indicated that the turbulence increased the transport of 1μm droplets,whereas it decreased the transport of 50μm *** contrast to only considering drag and turbulence,consideration of droplet interactions tended to increase both the transport and *** the length scale of the office is much larger than the droplet sizes,the droplet interactions,which occurred at the initial stages of release when droplet separation distances were shorter,had a significant effect in droplet fate by considerably manipulating the final locations on surfaces where droplets ***,it is proposed that when an exact prediction of transport and fate is required,especially for high droplet concentrations,the effects of droplet interactions should not be ignored.
