The Knee Joint Tissues Differ Significantly in TGFβ1 Expression and Its Sensitivity

The knee joint is the largest and most complex joint in the human body. In this study, we investigated TGFβ1 expression in the outer meniscus, inner meniscus and articular cartilage of rabbit and human knee tissue (outer and inner menisci) in order to determine the potential role of this factor in normal meniscal function. We also examined the potential of TGF-β1 stimulation to promote tissue regeneration in the two different regions of rabbit knee meniscus tissue. Immunohistochemical investigations of TGF-β1 were performed on rabbit and human knee tissue. The rabbit outer, inner and articular cartilage cells were culture and stimulated with TGF-β1 followed by cell proliferation assay and extracellular matrix analysis. Regulatory studies were performed using TGF-β1 inhibitors SB-431542 and PD98059. Gene expression was analyzed by quantitative polymerase chain reaction. We found marked regional variation in the expression of TGF-β1 in rabbit and human knee. TGF-β1 expressions are relatively greater in the outer meniscus than inner meniscus. Furthermore, we found that exogenous TGF-β1 stimulation increased cell proliferation and aggrecan synthesis more so in the outer than in the inner meniscus. Articular cartilage tissue shows moderate levels of cell proliferation and ECM synthesis when compared with outer and inner meniscus. These findings suggest that growth factors used to enhance the repair and regeneration of meniscal tissue should be tailored to enhance region-specific variation in cell proliferation and extracellular matrix synthesis.

The contamination of the environment by organic pollutants is a major risk factor,particularly for developing *** organic pollutants(SOPs)like the phenolic compounds,polyaromatic hydrocarbons(PAHs),pesticides,and herbicides pose serious environmental and health issues owing to their toxic characteristics and poor *** from their potential mutagenicity,carcinogenicity,tetragenicity and high body accumulation,these pollutants have become an increase concern *** is a promising alternative strategy for removing organic pollutants during water purification *** have several advantages such as simplicity of operation,good sorption capacity,high recoverability and *** a result,the focus and novelty of this review is on recent trends in the use of biosorbents,with a particular emphasis on the removal of SOPs from *** also cover use of bacteria biosorbents,fungal,algae and chitosan/chitin *** from that,we have also reviewed various classes of SOPs,their levels in the environment,classification and available characteristics techniques suitable for the adsorption experiments of these nanocomposites *** addition,we have provided comprehensive explanations and conclusions on possible future application of biosorbents and the mechanism of adsorption of these materials for removal of these SOPs from wastewater during water purification processes.

A microfluidic platform for continuous monitoring of dopamine homeostasis in dopaminergic cells

Isolation and Characterization of Contaminating Bacteria from Garcinia cambogia Extract: Methods to Reduce Microbial Load and Its Anti-Obesity Effect in Wistar Rats

Objectives: This study aimed to identify the contaminating bacteria in the extract of Garcinia cambogia, which is regularly used as a dietary supplement for addressing obesity in humans. Methods: The Garcinia cambogia extract was used and experiments were conducted to isolate the contaminating bacteria and antibiotic susceptibility was tested. The organism was identified using BIOLOG system. Such an extract was used in a placebo-controlled animal study when 6 eight adult male rats weighing between 200 and 220 g were randomly distributed into three groups (n = 3) and in test group 1, a single dose of 100 mg/kg bw of Garcinia cambogia extract was given while in the test group 2, 100 mg Garcinia cambogia extract + 116 mg Picrorhiza kurroa extract were administered through oral gavage. The normal control rats were given distilled water, and the treatment lasted for 30 days. Blood plasma and liver tissues were prepared for biochemical analysis and histology studies. Results: Nearly ~103 cfu/g of Bacillus atrophaeus was present in the Garcinia cambogia extract and we demonstrate >99% reduction in the microbial load with tetracycline. Such an extract at a dose of 100 mg/kg, showed weight loss in Wistar rats when administered orally for 1 month with no significant changes in liver histopathology. Picrorhiza kurroa, also known for its hepatoprotective properties, has been administered at a dose of 116 mg/kg along with Garcinia extract at 100 mg/kg orally and found to improve levels of hepatic enzymes as similar to control animals, although not statistically significant. Conclusions: The study revealed that Garcinia cambogia could prevent weight gain in Wistar rats when given orally and the weight gain in Garcinia-treated animals was almost 4 times less (7.31%), as against weight gain of 25.36% seen in vehicle control animals. Development and Validation of New RP-HPLC Method for the Estimation of Alfuzosin Hydrochloride in Bulk and Tablet Dosage Form

A reverse phase high performance liquid chromatographic method (HPLC) has been developed for the estimation of Alfuzosin hydrochloride (ALH) in the pharmaceutical formulation using RP-C18 column. The mobile phase (Tetrahydrofuran, Acetonitrile and buffer (pH 3.50) was pumped at a flow rate of 1.5 ml/min in the ratio of 1:20:80 and the eluents were monitored at 254.0 nm. Linearity was obtained in the concentration range of 80 - 120 μg/mL for ALH. The method was statistically validated and RSD was found to be less than 2% indicating high degree of accuracy and precision of the proposed HPLC method. Due to its simplicity, rapidness, high precision and accuracy, the proposed HPLC method may be used for determination of Alfuzosin hydrochloride in bulk drugs and in pharmaceutical dosage form.
