Clonal Fidelity of Micropropagated Psidium guajava L. Plants Using Microsatellite Markers

Micropropagation of Psidium guajava L. (guava) is a viable alternative to currently adopted techniques for large-scale plant propagation of commercial cultivars. Assessment of clonal fidelity in micropropagated plants is the first step towards ensuring genetic uniformity in mass production of planting material. In the present study, 31 plants of guava cultivar “Lucknow 49” regenerated by micropropagation were tested for genetic fidelity by comparing them to the mother plant from which explant material was obtained. Efficient rooting of in vitro proliferated shoots was obtained by culture on 1/2 strength MS medium supplemented with either 9.8 μM indole butyric acid (IBA) or 11.4 μM indole acetic acid (IAA). Leaf samples of 31 regenerated plants were compared to the mother plant using 17 simple sequence repeat (SSR) markers. While 16 SSRs detected the same allele, locus mPgCIR07 detected slight differences, where six micropropagated plants were 1 bp smaller (152 bp) than the parental genotype (153 bp). Differences in leaf tissues for anthocyanin pigmentation were also noted among micropropagated plants. Results of the study indicated efficient rooting of “Lucknow-49” cultivar for rapid propagation of planting material, and revealed that micropropagated plants were identical for 16 of the 17 loci examined. Although most mutations induced by tissue culture may not have an effect on phenotype, the possibility that novel phenotypes can be generated in a commercial setting exists.

Production of Transgenic Camelina sativaPlants via Agrobacterium-Mediated Transformation of Shoot Apical Meristems

A method to produce transgenic Camelina sativa plants in cvs. PI650159 and PI650161 was developed. Micropropagated shoot meristem cultures were established from in vitro germinated seedlings and used as target tissues for Agrobacterium-mediated transformation. A plasmid harboring enhanced green fluorescent protein, β glucuronidase and neomycin phosphotransferase II genes were used to optimize parameters for transgenic plant production. Kanamycin at 40 mg·l-1 was effective in suppression of non-transformed cells while permitting growth of transgenic tissues. Shoot apical meristems co-cultivated with Agrobacterium exhibited stable enhanced green fluorescence protein (EGFP) and β glucuronidase (GUS) expression after culture on plant regeneration medium. We observed transformation efficiencies of 53.33% in cv. PI650159 and 98.33% in cv. PI650161. The presence of transgenes in both cultivars was confirmed by PCR, while quantitative real-time PCR detected single copy integration in Pl650161 and two copy integration in Pl650159. Transgenic plants exhibited EGFP and GUS expression in all tissues including shoots, leaves, buds, floral organs, seeds, and pods. Our results demonstrate a simple and efficient technique using apical shoot meristems for production of transgenic C. sativa plants that can be used for transfer of desirable traits.

An optimized procedure for plant recovery from somatic embryos significantly facilitates the genetic improvement of Vitis

Paulownia is a genus of fast-growing and multipurpose tree species that is native to China. Due to their rapid growth and value in the timber market, many Paulownia species are cultivated in several temperate zones worldwide. Economic importance of Paulownia is increasing as new uses and related products are developed. It is also suitable as a lignocellulosic feedstock crop for the bioethanol industry in the Southeastern USA. A number of Paulownia species are valuable sources of secondary metabolites including flavonoids with high antioxidant activities. A high demand for planting material in domestic and international markets for afforestation and bioenergy production has necessitated the development of efficient micropropagation protocols for rapid and mass propagation of Paulownia. Over the past several decades, research on Paulownia species has been conducted to develop micropropagation, somatic embryogenesis and genetic transformation protocols for use in agroforestry and reforestation programs. Given the economic importance and current and potential future uses of Paulownia, this paper reviews the development of biotechnological approaches for plant propagation and genetic improvement, and antioxidant potential of secondary metabolites occurring in species.

Comparison of Ground Water Bacterial Cell Sizes from the Agricultural, Domestic and Industrial Areas of Mysore District, Karnataka State, India

A two-year study on temporal variations in the ground water heterotrophic bacterial cell sizes of free living bacteria (FLB) and particle bound bacteria (PBB) from the agricultural, domestic and industrial areas was carried out from February 2005 to January 2007. The overall mean cell length of FLB and PBB was similar in all the ground water studied. However, the season wise grouped data revealed significant seasonal changes in cell length of FLB and PBB, as smaller bacteria were noticed during rainy season in the ground water in agricultural area in both the years, and only in the second year of study in domestic and industrial areas. Generally, it was noticed that there were summer maximum and rainy minimum values of the cell length of PBB in the ground water in agricultural, domestic and industrial areas in the second year of study. The Pearson's correlations showed the presence of 8 (in agricultural area), 5 (in domestic) and 3 (in industrial) significant correlations with environmental (Physico-chemical) parameters, respectively. The regression analysis revealed that as much as 12% of variation in the mean length of FLB was due to NO3 (+) in agricultural area and 9% due to total solids (+) in domestic area. However, the 8 % variation in bacterial cell size of FLB was due to Mg (+) in industrial area. Whereas, 13 % variation in mean length of PBB was due to SO4 (+) in agricultural area and 10% due to total anions of strong acid (TASA) (+) in domestic area. Furthermore, 10 % of variation was due to PO4 (+) in industrial area. Thus, the statistical analysis revealed that several environmental variables were potentially responsible for some of the temporal variations in aquatic heterotrophic bacterial cell size, suggesting probably the stressed environment in these ecosystems.

Profiles of HIV-infected anti-retroviral therapy naïve children from Mumbai,India

New Formulas and Results for 3-Dimensional Vector Fields

New formulas are derived for once-differentiable 3-dimensional fields, using the operator . This new operator has a property similar to that of the Laplacian operator;however, unlike the Laplacian operator, the new operator requires only once-differentiability. A simpler formula is derived for the classical Helmholtz decomposition. Orthogonality of the solenoidal and irrotational parts of a vector field, the uniqueness of the familiar inverse-square laws, and the existence of solution of a system of first-order PDEs in 3 dimensions are proved. New proofs are given for the Helmholtz Decomposition Theorem and the Divergence theorem. The proofs use the relations between the rectangular-Cartesian and spherical-polar coordinate systems. Finally, an application is made to the study of Maxwell’s equations.
