Mitochondrial control of hematopoietic stem cell balance and hematopoiesis

Hematopoietic stem cells （HSCs） are stem cells from mesodermal derivation that reside in bone marrow and provide blood cells for the whole life of an adult individual, through a process called hematopoiesis. The long lasting support of HSCs for hematopoiesis is permitted by the fine regulation of quiescence and division output. Exit from the quiescent state is to produce a committed and/or stem daughter cells, in an event defined asymmetric or symmetric division. A deregulation in the proportion between asymmetric and symmetric divisions is critical in the appearance of hematological disorders ranging from bone marrow failure to hematological malignancies. Over the past years, several studies have indicated how the metabolism of HSCs is determinant in the regulation of HSC quiescence and commitment process. A metabolism shifted to the glycolytic pathway promotes HSCs quiescence and sustainment of hematopoiesis. Boosting mitochondrial respiration promotes the stem cell commitment followed by stem pool exhaustion, and minimal mitochondrial activity is required to maintain the HSCs quiescence. In the present review are discussed the most recent advances in comprehension of the roles of mitochondria in the hematopoiesis and in the division balance.

State feedback stabilization of cascaded nonlinear systems with discontinuous connection

In practical engineering, many phenomena are described as a discontinuous function of a state variable, and the discontinuity is usually the main reason for the degradation of the control performance. For example, in the set-point control problem of mechanical systems, the static friction （described by a sgn function of velocity of the contacting faces） causes undesired positioning error. In this paper, we will investigate the stabilization problem for a class of nonlinear systems that consist of two subsystems with cascaded connection. We will show the basic idea with a special case first, and then the result will be extended to more general cases. Some interesting numerical examples will be given to demonstrate the effectiveness of the proposed design approach.

The role of the CNOT1 subunit of the CCR4-NOT complex in mRNA deadenylation and cell viability

The human CCR4-NOT deadenylase complex consists of at least nine enzymatic and non-enzymatic *** evidence suggests that the non-enzymatic subunits are involved in the regulation of mRNA deadenylation,although their precise roles remain to be *** this study,we addressed the function of the CNOT1 subunit by depleting its expression in HeLa *** cytometric analysis revealed that the sub G1 fraction was increased in CNOT1-depleted ***,the same level of the sub G1 fraction was seen when cells were treated with a mixture of siRNAs targeted against all enzymatic subunits,suggesting that CNOT1 depletion induces apoptosis by destroying the CCR4-NOT-associated deadenylase *** analysis revealed that CNOT1 depletion leads to a reduction in the amount of other CCR4-NOT ***,the specific activity of the CNOT6L immunoprecipitates-associated deadenylase from CNOT1-depleted cells was less than that from control *** formation of P-bodies,where mRNA decay is reported to take place,was largely suppressed in CNOT1-depleted ***,CNOT1 has an important role in exhibiting enzymatic activity of the CCR4-NOT complex,and thus is critical in control of mRNA deadenylation and mRNA *** further showed that CNOT1 depletion enhanced CHOP mRNA levels and activated caspase-4,which is associated with endoplasmic reticulum ER stress-induced *** together,CNOT1 depletion structurally and functionally deteriorates the CCR4-NOTcomplex and induces stabilization of mRNAs,which results in the increment of translation causing ER stress-mediated *** conclude that CNOT1 contributes to cell viability by securing the activity of the CCR4-NOT deadenylase.

Our purpose was to determine how enamel matrix derivative (EMD) affects the expression ofosteogenesis- and chondrogenesis-related transcription factors in undifferentiated mesenchymal cells.C3H10T1/2 cell line,a typical pluripotential mesenchymal cell line,was cultured with or without EMD forup to 7 *** of mRNAs encoding osteogenesis-and chondrogenesis-related transcription factors(Runx2, Osterix,AJ18,Dlx5,Msx2,Sox5,Sox9 and Zfp60) was measured using real-time polymerasechain ***2 and Sox9 protein expression and the presence of bone morphogenetic protein (BMP)-6-like molecules in EMD were determined by Western *** substantially increased mRNA levels ofosteogenesis-and chondrogenesis-related transcription *** also induced Runx2 and Sox9 *** blotting analysis of EMD using anti-BMP-6 antibody revealed immunoreactive bandscorresponding to about 14 kDa and 60 *** results suggest that EMD stimulates osteogenesis- andchondrogenesis-related transcription factors,and these activities may be mediated,at least in part,by BMP-6in EMD.

Lattice Boltzmann Flow Simulation in a Combined Nanochannel

The widely used micro-flow wall-boundary conditions for lattice Boltzmann method(LBM)are evaluated in a force driven combined nanochannel *** flow field consists of a two-dimensional nanochannel(mother channel)of an infinite length having flat plates of a finite length *** flat plate is set above the bottom wall of the nanochannel with a narrow *** flow,thus,develops through this narrow gap(narrower channel)and the other side of the plate(wide gap).The Knudsen number based on the mother channel height is Kn=0.14 whereas the characteristic Knudsen number in the narrower channel is *** obtain the reference data,the molecular dynamics(MD)simulation is performed with a fully diffusive wall *** LBMs are based on the lattice BGK model and with the bounce-back/specular reflection(BSBC)and the diffuse scattering(DSBC)wall boundary *** relaxation time is modified to include sensitivity to *** DSBC shows generally satisfactory results in the test flow cases including fully developed force driven Poiseuille flows,where the BSBC performs worse at Kn>0.5 with a fixed bridge coefficient of b=*** results in its overprediction of the flow rate in the narrower channel region since the characteristic Knudsen number there is *** MD simulation suggests that the flow develops gradually through the narrower channel region though all the LBM predictions show almost instant flow *** fact suggests that the relaxation time model needs to have more sensitivity to the locally defined *** discussions of the BSBC with a different set of models suggest that the regularization process is required for predicting complex nanoscale flows.

Seemingly maladaptive refraining from infanticidal probing at the third parasitism attack by the semi-solitary parasitoid Echthrodelphax fairchildii （Hymenoptera： Dryinidae）

We assessed the adaptiveness of refraining from infanticidal probing at the third attack under triple parasitism in the parasitoid Echthrodelphaxfairchildii Perkins, by comparing fitness returns from the third attack with and without probing. Fitness returns were assessed in terms of the survival rate, mean developmental time, and mean adult head width of the progenies. Not probing was maladaptive under triple parasitism with oviposition intervals of 1/24 and 24/24 hours （where the numbers before and after the slash refer to the first-to-second and second-to-third oviposition intervals, respectively）, although no significant difference was detected in fitness returns for oviposition intervals of 24/1 hours. We suggest that the cost of probing （especially the decrease in the chance of future ovipositions） is a reason for the seemingly maladaptive absence of probing.
