The integration of climate change,spatial dynamics,and habitat fragmentation:A conceptual overview

A growing number of studies have looked at how climate change alters the effects of habitat fragmentation and degradation on both single and multiple species;some raise concern that biodiversity loss and its effects will be exacerbated.The published literature on spatial dynamics(such as dispersal and metapopulation dynamics),habitat fragmentation and climate change requires synthesis and a conceptual framework to simplify thinking.We propose a framework that integrates how climate change affects spatial population dynamics and the effects of habitat fragmentation in terms of:(i)habitat quality,quantity and distribution;(ii)habitat connectivity;and(iii)the dynamics of habitat itself.We use the framework to categorize existing autecological studies and investigate how each is affected by anthropogenic climate change.It is clear that a changing climate produces changes in the geographic distribution of climatic conditions,and the amount and quality of habitat.The most thorough published studies show how such changes impact metapopulation persistence,source-sink dynamics,changes in species’geographic range and community composition.Climate-related changes in movement behavior and quantity,quality and distribution of habitat have also produced empirical changes in habitat connectivity for some species.An underexplored area is how habitat dynamics that are driven by climatic processes will affect species that live in dynamic habitats.We end our discussion by suggesting ways to improve current attempts to integrate climate change,spatial population dynamics and habitat fragmentation effects,and suggest distinct areas of study that might provide opportunities for more fully integrative work.

Osteochondral(OC)repair is an extremely challenging topic due to the complex biphasic structure and poor intrinsic regenerative capability of natural osteochondral tissue.In contrast to the current surgical approaches which yield only short-term relief of symptoms,tissue engineering strategy has been shown more promising outcomes in treating OC defects since its emergence in the 1990s.In particular,the use of multizonal scaffolds(MZSs)that mimic the gradient transitions,from cartilage surface to the subchondral bone with either continuous or discontinuous compositions,structures,and properties of natural OC tissue,has been gaining momentum in recent years.Scrutinizing the latest developments in the field,this review offers a comprehensive summary of recent advances,current hurdles,and future perspectives of OC repair,particularly the use of MZSs including bilayered,trilayered,multilayered,and gradient scaffolds,by bringing together onerous demands of architecture designs,material selections,manufacturing techniques as well as the choices of growth factors and cells,each of which possesses its unique challenges and opportunities.

Landscape effects on the thermotolerance of carabid beetles and the role of behavioral thermoregulation

Physiological thermotolerance and behavioral thermoregulation are central to seasonal cold adaptation in ectothermic organisms.For species with enhanced mobility,behavioral responses may be of greater importance in the cold stress response.Employing the carabid beetles as a study organism,the current study compared physiological thermotolerance and behavioral thermoregulation in carabid species inhabiting cereal fields in different landscape contexts,from fine grain heterogeneous“complex”landscapes to homogenous“simple”landscapes.Physiological thermotolerance was determined via measurement of the CTmin and chill coma temperature.Behavioral responses to cold temperature exposure were determined employing a purpose built arena,and thoracic temperature measured to estimate the efficacy of the behavior as a form of behavioral thermoregulation.Results revealed an influence of landscape composition on the cold tolerance of carabid beetles,although species differed in their sensitivity to landscape intensification.A reduced effect of landscape on the thermotolerance of larger carabid beetles was observed,thought to be the consequence of greater mobility preventing local acclimation to microclimatic variation along the landscape intensification gradient.Investigation into behavioral thermoregulation of the 3 largest species revealed burrowing behavior to be the main behavioral response to cold stress,acting to significantly raise carabid body temperature.This finding highlights the importance of behavioral thermoregulation as a strategy to evade cold stress.The use of behavioral thermoregulation may negate the need to invest in physiological thermotolerance,further offering explanation for the lack of landscape effect on the physiological thermotolerance of larger carabids.

The Chloroplast kinase Network： New Insights from Large-Scale Phosphoproteome Profiling

Protein phosphorylation is one of the most important posttranslational modifications in eukaryotic cells and affects almost all basic cellular processes. The chloroplast as plant-specific cell organelle with important metabolic functions is integrated into the cellular signaling and phosphorylation network. Recent large-scale chloroplast phosphoproteome analyses in Arabidopsis have provided new information about phosphorylation targets and expanded the list of chloroplast metabolic and regulatory functions that are potentially controlled by protein phosphorylation. Phosphorylated peptides identified from chloroplast proteins provide new insights into phosphorylation motifs, protein kinase activities, and substrate utilization. Phosphorylation sites in protein kinases can reveal chloroplast phosphorylation cascades that may network different functions by integrating signaling chains. Our review provides a meta-analysis of currently available chloroplast phosphoproteome information and discusses biological insights from large-scale chloroplast phosphoprotein profiling as well as technological constraints of kinase network analysis.

The Secret Life of Chloroplast Precursor Proteins in the Cytosol

Eukaryotic cells contain semi-autonomous organelles that have lost most of their coding capacity for essential proteins to the nucleus.Therefore,their functioning depends on the cytosolic translation products of nuclear-encoded genes that must be imported by a specialized protein import machinery.Chloroplasts import between 2500 and 3500 proteins for their functioning,demonstrating that the bulk of the chloroplast proteome is shuttied through the cytosol as precursor proteins.Precursor proteins usually contain an N-terminal transit peptide that is cleaved off by a signa/processing peptidase(SPP)and degraded by the chloroplast protease network upon import.In the cytosol,precursors can be co-and post-translationally modified,interact with a set of chaperones,co-chaperones,14-3-3 proteins,and/or E3 ligases before they are either imported or degraded by the cytosolic proteasome complex.It is now clear that precursors are not only a transient transport form of functional proteins but that their cytosolic transition is of regulatory importance.

Quantification of changes in zero valent iron morphology using X-ray computed tomography

Morphological changes within the porous architecture of laboratory scale zero valent iron （ZVI） permeable reactive barriers （PRBs）, after exposure to different groundwater conditions, have been quantified experimentally for different ZVI/sand ratios （10%, 50% and 100%, W/W） with the aim of inferring porosity changes in field barriers. Column studies were conducted to simulate interaction with different water chemistries, a synthetic groundwater, acidic drainage and deionised （DI） water as control. Morphological changes, in terms of pore size and distribution, were measured using X-ray computed tomography （CT）. CT image analysis revealed significant morphological changes in columns treated with different water chemistries. For example, 100% ZVI （W/W） columns had a higher frequency of small pores （0.6 mm） was observed in ZVI grains reacted with typical groundwater, resulting in a porosity of 27%, compared to 32% when exposed to DI water. In comparison, ZVI grains treated with the acidic drainage had higher porosity （44%） and larger average pore size （2.8 mm）. 10% ZVI PRB barrier material had the highest mean porosity （56%） after exposure to any water chemistry whilst 100% ZVI （W/W） columns always had the lowest （34%） with the 50% ZVI （W/W） in between （40%）. These results agree with previously published PRB field data and simultaneously conducted geochemical monitoring and mass balance calculation, indicating that both the geochemical and hydraulic environment of the PRB play an important role in determining barrier lifespan. This study suggests that X-ray CT image analysis is a powerful tool for studying the detailed inter pores between ZVI grains within PRBs.

Effects of the neurotrophic factor artemin on sensory afferent development and sensitivity

Artemin is a neuronal survival and differentiation factor in the glial cell line-derived neurotrophic factor family.Its receptor GFRα3 is expressed by a subpopulation of nociceptor type sensory neurons in the dorsal root and trigeminal ganglia(DRG and TG).These neurons co-express the heat,capsaicin and proton-sensitive channel TRPV1 and the cold and chemical-sensitive channel TRPA1.To further investigate the effects of artemin on sensory neurons,we isolated transgenic mice(ARTN-OE mice) that overexpress artemin in keratinocytes of the skin and tongue.Enhanced levels of artemin led to a 20% increase in the total number of DRG neurons and increases in the level of mRNA encoding TRPV1 and TRPA1.Calcium imaging showed that isolated sensory neurons from ARTN-OE mice were hypersensitive to the TRPV1 agonist capsaicin and the TRPA1 agonist mustard oil.Behavioral testing of ARTN-OE mice also showed an increased sensitivity to heat,cold,capsaicin and mustard oil stimuli applied either to the skin or in the drinking water.Sensory neurons from wildtype mice also exhibited potentiated capsaicin responses following artemin addition to the media.In addition,injection of artemin into hindpaw skin produced transient thermal hyperalgesia.These findings indicate that artemin can modulate sensory function and that this regulation may occur through changes in channel gene expression.Because artemin mRNA expression is up-regulated in inflamed tissue and following nerve injury,it may have a significant role in cellular changes that underlie pain associated with pathological conditions.Manipulation of artemin expression may therefore offer a new pain treatment strategy.

The neurotrophin and glial cell line-derived neurotrophic factor(GDNF) family of growth factors have been extensively studied because of their proven ability to regulate development of the peripheral nervous system.The neurotrophin family,which includes nerve growth factor(NGF),NT-3,NT4/5 and BDNF,is also known for its ability to regulate the function of adult sensory neurons.Until recently,little was known concerning the role of the GNDF-family(that includes GDNF,artemin,neurturin and persephin) in adult sensory neuron function.Here we describe recent data that indicates that the GDNF family can regulate sensory neuron function,that some of its members are elevated in inflammatory pain models and that application of these growth factors produces pain in vivo.Finally we discuss how these two families of growth factors may converge on a single membrane receptor,TRPV1,to produce long-lasting hyperalgesia.

Successful import of hundreds of nucleus-encoded proteins is essential for chloroplast biogenesis. The import of cytosolic precursor proteins relies on the Toc- （translocon at the outer chloroplast membrane） and Tic- （translocon at the inner chloroplast membrane） complexes. In Arabidopsis thaliana, precursor recognition is mainly mediated by outer membrane receptors belonging to two gene families： Toc34/33 and Toc159/132/120/90. The role in import and precursor selectivity of these receptors has been intensively studied, but the function of Toc90 still remains unclear. Here, we report the ability of Toc90 to support the import of Toc159 client proteins. We show that the overexpression of Toc90 partially complements the albino knockout of Toc159 and restores photoautotrophic growth. Several lines of evidence including proteome profiling demonstrate the import and accumulation of proteins essential for chloroplast biogenesis and functionality.

Tax distortions in cross-border flows of intangible assets

The phenomenon of global fragmented production and associated trade in intermediate products,including intangible assets,has changed how economists study globalization and how new public policies are shaped.Understanding cross-border flows of disembodied knowledge,often associated with intellectual property(IP),is essential for analyzing how modern economies operate.Available data to document these international IP-related knowledge flowsdnamely cross-border payments for IPdare distorted by various factors.Tax planning by multinational enterprises has seriously distorted the measurement of cross-border IP flows,affecting national measurements of imports,exports,GDP,and productivity.The tax-induced mismeasurement could be more than 35% of global charges for use of intellectual property,and greater for individual countries,particularly high-taxrate countries.International initiatives to address the effects of tax base erosion,profit shifting,and other statistical initiatives on global value chains will improve future measurements of cross-border IP flows,improving the understanding of both the creation and uses of IP.
