Nanotechnology-based approach for safer enrichment of semen with best spermatozoa

Background: Advances in nanotechnology have permitted molecular-based targeting of cells through safe and biocompatible magnetic nanoparticles(MNP). Their use to detect and remove damaged spermatozoa from semen doses could be of great interest. Here, MNP were synthesized and tested for their ability to target apoptotic(annexin V) and acrosome-reacted(lectin) boar spermatozoa, for high-throughout retrieval in a magnetic field(nanoselection). The potential impacts of nanoselection on sperm functions and performance of offspring sired by sperm subjected to nanoselection were determined. Fresh harvested and extended boar semen was mixed with various amounts(0, 87.5, and 175 μg) of MNP-conjugates(Annexin V-MNP or Lectin-MNP) and incubated(10 to15 min) for 37 °C in exp. 1. In exp. 2, extended semen was mixed with optimal concentrations of MNP-conjugates and incubated(0, 30, 90, or 120 min). In exp. 3, the synergistic effects of both MNP-conjugates(87.5 μg– 30 min)on spermatozoa was evaluated, followed by sperm fertility assessments through pregnancy of inseminated gilts and performance of neonatal offspring. Sperm motion, viability, and morphology characteristics were evaluated in all experiments.Results: Transmission electron microscopy, atomic force microscopy, and hyperspectral imaging techniques were used to confirm attachment of MNP-conjugates to damaged spermatozoa. The motility of nanoselected spermatozoa was improved(P e viability of boar sperm, as assessed by the abundance of reactive oxygen species and the integrity of the acrosome, plasma membrane, and mitochondrial membrane was not different between nanoselected and control spermatozoa. The fertility of gilts inseminated with control or nanoselected spermatozoa, as well as growth and health of their offspring were not different between(P > 0.05).Conclusions: The findings revealed the benefit of magnetic nanoselection for high-throughput targeting of damaged sperm, for removal and rapid and effortless enrichment of semen doses with highly motile,

Fluorescence confocal laser-scanning microscopy(LSM)is one of the most popular tools for life science research.This popularity is expected to grow thanks to single-photon array detectors tailored for LSM.These detectors offer unique single-photon spatiotemporal information,opening new perspectives for gentle and quantitative superresolution imaging.However,a flawless recording of this information poses significant challenges for the microscope data acquisition(DAQ)system.We present a DAQ module based on the digital frequency domain principle,able to record essential spatial and temporal features of photons.We use this module to extend the capabilities of established imaging techniques based on single-photon avalanche diode(SPAD)array detectors,such as fluorescence lifetime image scanning microscopy.Furthermore,we use the module to introduce a robust multispecies approach encoding the fluorophore excitation spectra in the time domain.Finally,we combine time-resolved stimulated emission depletion microscopy with image scanning microscopy,boosting spatial resolution.Our results demonstrate how a conventional fluorescence laser scanning microscope can transform into a simple,information-rich,superresolved imaging system with the simple addition of a SPAD array detector with a tailored data acquisition system.We expected a blooming of advanced single-photon imaging techniques,which effectively harness all the sample information encoded in each photon.

Chitin powder and chitin-aerogel were prepared from shrimp wastes and used to uptake Y^(3+)from aqueous solutions and concentrate this rare earth element from phosphogypsum(PG).Chitin aerogel displays a specific surface area of 945 m^(2)/g,while chitin powder is 3.6 m^(2)/g,which largely influences its adsorption ability.Regarding the adsorption in synthetic solutions,the effect of pH on Y^(3+)removal is strong for chitin powder adsorbent.In contrast,no big pH influence was detected for chitin aerogel.electrostatic interactions and chelation can highlight the proposed mechanism of Y^(3+)on chitin adsorbents for the powder and aerogel.Furthermore,in addition to these interactions,pore filling/pore diffusion is the main mechanism of Y^(3+)removal in the chitin aerogel.Chitin aerogel is efficient in concentrating 8 times the Y^(3+)from PG,a very complex matrix.The complex chitin aerogel-Y^(3+)can be a secondary source of rare earth elements for other applications.

To control capillary bleeding, surgeons may use absorbable hemostatic agents, such as Surgicel® and TachoSil®. Due to their slow resorption, their persistence in situ can have a negative impact on tissue repair in the resected organ. To avoid complications and obtain a hemostatic agent that promotes tissue repair, a zinc-supplemented calcium alginate compress was developed: HeMO-IONIC®. This compress is non-absorbable and is therefore removed once hemostasis has been achieved. After demonstrating the hemostatic efficacy and stability of the blood clot obtained with HeMO-IONIC, the impact of Surgicel, TachoSil, and HeMO-IONIC on cell activation and tissue repair were compared (i) in vitro on endothelial cells, which are essential to tissue repair, and (ii) in vivo in a mouse skin excision model. In vitro, only HeMO-IONIC maintained the phenotypic and functional properties of endothelial cells and induced their migration. In comparison, Surgicel was found to be highly cytotoxic, and TachoSil inhibited endothelial cell migration. In vivo, only HeMO-IONIC increased angiogenesis, the recruitment of cells essential to tissue repair (macrophages, fibroblasts, and epithelial cells), and accelerated maturation of the extracellular matrix. These results demonstrate that a zinc-supplemented calcium alginate, HeMO-IONIC, applied for 10 min at the end of surgery and then removed has a long-term positive effect on all phases of tissue repair.

Optimizing the perception of external cues and regulating physiology accordingly help plants to cope with the constantly changing environmental conditions to which they are exposed.An array of photoreceptors and intricate signaling pathways allow plants to convey the surrounding light information and synchronize an endogenous timekeeping system known as the circadian clock.This biological clock integrates multiple cues to modulate a myriad of downstream responses,timing them to occur at the best moment of the day and the year.Notably,the mechanism underlying entrainment of the light-mediated clock is not clear.This review addresses known interactions between the light-signaling and circadian-clock networks,focusing on the role of light in clock entrainment and known molecular players in this process.

Reconsidering sex differences during place learning in tungara frogs

In tungara frogs, female mate choice requires remembering the location and/or calls of preferred males who advertise from fixed positions within a breeding pond. A previous study found that, when solving a place discrimination task in the laboratory, female tCingara frogs were able to learn a visual cue to solve the task, whereas males were not. In that task, male performance appeared to be inhibited, in part, by their attempt to use egocentric cues. We tested whether the sex difference in place learning previously reported would generalize to other training parameters with different cues available by eliminating the potential to use egocentric cues and increasing the number of trials per day. As before, frogs were given a choice between a red or yellow door, one of which led to shelters and return to their home cage. In the current testing conditions, we detected a preference for the red door;thus, we only considered frogs rewarded to the yellow door. Training was associated with an in crease in correct choices and an in creased preference for the yellow door. However, there was no evide nee for a sex d iff ere nee in learni ng. In summary, un der the curre nt training conditions, we fou nd that the appare nt female advantage in place learning was no Ion ger evide nt. Future studies that in vestigate sex d iff ere nces in cue pref ere nee and/or ability to switch among cues will further illuminate the conditions under which sex differences in learning are manifest in tungara frogs.

Dear editor effective vaccines and some antiviral agents against SARS-CoV-2,the causative agent of coronavirus disease 2019(COVID-19),have been developed.Nonetheless,additional therapeutics are needed since emerging SARS-CoV-2 variants evade immunity induced by vaccination or previous infection and are resistant to therapeutic antibodies and fusion inhibitors.^(1)In addition,many patients experience prolonged post-acute sequelae of SARS-CoV-2.

Treatment of O-allyl N-tosyl carbamates with aromatic compounds in the presence of Cu(OTf)_(2) or TMSOTf as promoters affords N-substituted 1-arylpropan-2-amines,1,2-diarylpropanes,1,1-diarylpropanes,or indanes,depending on the nature of the promoter and of the aryl substrates.A full mechanistic rational allowing appreciation of the outcome of these novel C–H based cascades is proposed.An initial acid promoted decarboxylative/deamidative Friedel–Crafts allylation takes place.After protonation of the allylated arene,evolution of the resulting cation may follow different paths depending on the nature of the arene partner and of the allyl moiety in the carbamate.

effect of acetyl-L-carnitine on hypersensitivity in acute recurrent caerulein-induced pancreatitis and microglial activation along the brain’s pain circuitry

BACKGROUND Acute pancreatitis(AP)and recurring AP are serious health care problems causing excruciating pain and potentially lethal outcomes due to sepsis.The validated caerulein-(CAe)induced mouse model of acute/recurring AP produces secondary persistent hypersensitivity and anxiety-like behavioral changes for study.AIM To determine efficacy of acetyl-L-carnitine(ALC)to reduce pain-related behaviors and brain microglial activation along the pain circuitry in CAe-pancreatitis.MeTHODS Pancreatitis was induced with 6 hly intraperitoneal(i.p.)injections of CAe(50μg/kg),3 d a week for 6 wk in male C57BL/6J mice.Starting in week 4,mice received either vehicle or ALC until experiment’s end.Mechanical hypersensitivity was assessed with von Frey filaments.Heat hypersensitivity was determined with the hotplate test.Anxiety-like behavior was tested in week 6 using elevated plus maze and open field tests.Microglial activation in brain was quantified histologically by immunostaining for ionized calcium-binding adaptor molecule 1(Iba1).ReSULTS Mice with CAe-induced pancreatitis had significantly reduced mechanical withdrawal thresholds and heat response latencies,indicating ongoing pain.Treatment with ALC attenuated inflammation-induced hypersensitivity,but hypersensitivity due to abdominal wall injury caused by repeated intraperitoneal injections persisted.Animals with pancreatitis displayed spontaneous anxiety-like behavior in the elevated plus maze compared to controls.Treatment with ALC resulted in increased numbers of rearing activity events,but time spent in“safety”was not changed.After all the abdominal injections,pancreata were translucent if excised at experiment’s end and opaque if excised on the subsequent day,indicative of spontaneous healing.Post mortem histopathological analysis performed on pancreas sections stained with Sirius Red and Fast Green identified wide-spread fibrosis and acinar cell atrophy in sections from mice with CAe-induced pancreatitis that was not rescued by treatment with ALC.Microglial Iba1

An integrated multiscale approach for characterization of rock masses subjected to tunnel excavation

The design of tunnels must be conducted based on the knowledge of the territory. The longer the structure, the larger the area to be investigated, and the greater the number of surveys and tests to be performed in order to thoroughly examine all the relevant features. Therefore, optimization of the investigation process is strongly required to obtain complete and reliable data for the design of the infrastructure. The fast development of remote sensing technologies and the affordability of their products have contributed to proving their benefits as supports for investigation, encouraging the spreading of automatic or semi-automatic methods for regional scale surveys. Similarly, considering the scale of the rock outcrop, photogrammetric and laser scanner techniques are well-established techniques for representing geometrical features of rock masses, and the benefits of non-contact surveys in terms of safety and time consumption are acknowledged. Unfortunately, in most cases, data obtained at different scales of investigations are only partially integrated or compared, probably due to the missing exchange of knowledge among experts of different fields（e.g. geologists and geotechnical engineers）. The authors,after experiencing such a lack of connection among the results of different surveys concerning tunnels,propose a multiscale approach for the optimization of the investigation process, starting from the regional scale, to obtain the data that can be useful not only for planning more detailed surveys in a preliminary phase, but also for making previsions on the discontinuity sets that are present in the rock masses subjected to excavations. A methodological process is proposed and illustrated by means of a case study. Preliminary results are discussed to highlight the potentiality of this method and its limitations.
